    复合地层盾构刀盘掘进速率及受力特性分析

    为探究复合地层中盾构刀盘的掘进速率及受力特性,基于连续-非连续单元法(CDEM),建立盾构-岩土体这一完整系统的数值计算模型。通过在CDEM中引入简单有限体积法、虚拟质量法、单元溶蚀算法等系列算法,实现盾构刀盘掘进全过程的三维模拟。建立5种刀盘面花岗岩占比的刀盘破岩数值模型,并对不同刀盘面花岗岩占比下的刀盘进尺、掘进速率、平均转矩、平均倾覆力矩等进行详细分析。数值模拟结果表明:1)在某一特定复合地层中掘进时,随着掘进时间增加,刀盘进尺基本呈线性增大的趋势,但进尺曲线中平缓段及速升段交替循环出现; 2)刀盘面花岗岩占比由0增大至100%,掌子面处拉伸破坏单元增加,剪切破坏单元减少,且刀盘的掘进速率呈指数衰减趋势,掘进速率降低88. 7%; 3)刀盘面花岗岩占比由0增大至100%,平均转矩值逐渐增大,增大比例为117. 5%,平均倾覆力矩值则呈现先增大后减小的趋势,且在花岗岩占比为50%时最大

    Modified GA-FFT for Synthesizing Shaped Pattern of Unequally Spaced Array in Presence of Mutual Coupling

    该文提出了一种虚拟的最小均方有源单元方向图展开方法,将不等间隔阵列的有源方向图展开为一个虚拟的均匀间隔阵列的若干单元辐射的叠加。通过该方法,对包含阵元耦合效应的不等间隔阵列方向图,可以使用快速傅里叶变换进行加速计算。并且,该文将这个方法与遗传算法(GA)相结合,得到一种改进的GA-FFT方法,可以应用于解决含阵元互耦的不均匀间隔阵列的赋形波束综合问题。最后,分别对不等间隔的偶极子阵列平顶方向图及微带阵列的余割平方方向图进行了综合,结果表明所提方法的有效性和优势。A new Virtual Least-Square Active Element Pattern Expansion(VLS-AEPE) method is presented in this paper, which considers each active element pattern of an unequally spaced array as the one radiated by some of equally spaced elements of a virtual array. Using the help of this method, the pattern of an unequally spaced array including mutual coupling can be efficiently calculated by FFT. In addition, this method is combined with the Genetic Algorithm(GA) to deal with the shaped pattern synthesis problem for unequally spaced linear arrays. Two synthesis experiments including the synthesis of flat-top pattern for an unequally spaced dipole array and the synthesis of cosec-squared pattern for an unequally spaced microstrip array are conducted to verify the effectiveness and advantages of the proposed algorithm.国家自然科学基金(61301009);; 中央高校基本科研业务费(20720160081)~

    Inhibitory effect of anti-human NRP-1 monoclonal antibody on hepatocellular carcinoma cell line HepG2 and its mechanism in vitro

    目的研究抗人神经纤毛蛋白-1(Neuropilin1,NRP-1)单克隆抗体对肝癌Hep G2细胞的生长抑制作用及其机制。方法小鼠腹水法制备抗NRP-1单克隆抗体(NRP-1 m Ab)并用r Protein A亲和柱纯化抗体,间接ELISA检测抗体的滴度水平。Western blot检测NRP-1 m Ab对Hep G2细胞的特异性,细胞免疫荧光和流式细胞术检测NRP-1蛋白在肝癌细胞株Hep G2上的表达,MTT法检测NRP-1 m Ab对Hep G2的生长抑制作用,Western blot检测ERK1/2、P-ERK1/2、Akt、P-Akt蛋白的表达水平。结果 SDS-PAGE和间接ELISA检测纯化的NRP-1 m Ab纯度为95%以上,效价为1×10~(-6);Western blot检测结果显示NRP-1 m Ab可与Hep G2细胞膜上的NRP-1蛋白特异性结合。细胞免疫荧光染色结果显示NRP-1定位于Hep G2细胞膜,流式细胞术结果显示NRP-1蛋白在Hep G2细胞上表达水平较高;MTT法检测结果显示NRP-1 m Ab对Hep G2细胞有生长抑制作用。Western blot检测到在不同浓度NRP-1 m Ab作用下,Hep G2细胞裂解液P-ERK1/2、P-Akt蛋白的条带信号逐渐减弱。结论纯化的NRP-1m Ab能抑制Hep G2细胞的生长,其抑制作用是通过EGF和HGF信号通路实现的。The aim of the experimental is to investigate the inhibitory effect of anti-human nerve cilia protein1(Neuropilin-1,NRP-1) monoclonal antibody(NRP-1 mAb) on hepatocellular carcinoma cell line HepG2 and its mechanism in vitro.Anti-human NRP-1 monoclonal antibody(NRP-1 mAb) was prepared from mouse ascites and purified by rProteinA affinity column assay.The titer of antibody was determined using indirect ELISA assay;the characteristic of NRP-1 mAb binding to NRP-1 was determined using Western blotting;the expression of NRP-1protein in hepatocellular carcinoma cell line HepG2 was determined using immunofluorescence assay and flow cytometry assay.Growth inhibition of HepG2 cells treated with different concentrations of NRP-1 mAb was determined using MTT assay,while Western blotting was used to detect the expression levels of ERK1/2,P-ERK1/2,Akt and P-Akt proteins.The results of SDS-PAGE and indirect ELISA showed that the purity of purified NRP-1mAb was more than 95%and the titer was 1×10~(-6).Western blotting analysis suggested that NRP-1 mAb could bind specifically to NRP-1 on HepG2 cell;immunofluorescence staining showed that NRP-1 was located in the membrane of HepG2 cells.Flow cytometry analysis showed that the expression level of NRP-1 on HepG2 cell was relatively high.Western blotting analysis suggested that P-ERK1/2 and P-Akt expression levels were down-regulated after having incubated HepG2 cells with different concentrations of NRP-1 mAb.In conclusion,NRP-1 mAb could inhibit the growth of HepG2 cells(P<0.05),and its inhibitory effect is achieved by reducing the P-ERK1/2 and P-Akt expression.南京军区医学科技创新项目(12MA061,15MS104

    The Establishment of Using Liquid Chip Technology to Quantify Prostate Specific Antigen in Human Serum

    目的建立利用液相芯片技术定量检测人血清中前列腺特异性抗原的反应模式,并对该方法进行评价。方法应用液相芯片技术检测50例前列腺增生患者血清中的前列腺特异性抗原(PSA),评价该方法的线性范围、最低检测限、精密度等指标,并将结果与化学发光免疫分析法(ClIA)进行相关性分析。结果液相芯片技术检测PSA标准曲线的线性范围为0.022~129.6ng/Ml,PSA的最低检测限为0.018ng/Ml,检测PSA的批内CV为2.18%~2.28%,批间CV为1.61%~4.18%。用液相芯片技术和化学发光法分别对受检血清标本进行PSA定量分析,结果表明两法差异无显著性(P>0.05),r=0.9984,相关性良好。结论液相芯片技术具有线性范围广、灵敏度高、重复性好、节省样品和时间等优点,具有临床应用潜力。Objective To quantify prostate specific antigen in human serum using liquid chip method and evaluate the performance of the method.Methods Liquid chip method was employed to detect the PSA in 50 cases of benign prostatic hyperplasia patients'serum,evaluating the index of linear range,detection limit and accuracy,and make a correlation analysis with chemiluminescence immunoassay(CLIA).Results The linear range for the measurement of PSA was 0.022--129.6ng/mL and the lowest detection limit for PSA was 0.018 ng/ml.The intra-and inter-assay coefficients of variation were 2.18%~2.28% and 1.61%~4.18% respectively.Liquid chip method and CLIA were used to quantify PSA in human serum.The results showed that there were no significant differences in these two methods(P>0.05),but they had a good correlation(r=0.9984).Conclusion Liquid chip immunoassay has wide linear range,high sensitivity,repeatability,saving time and samples,with potential clinical applications

    Comparison of fertilizer-effect models on winter wheat response to nitrogen and phosphorus fertilizers in saline soils in the Yellow River Delta

    通过大田试验, 研究了黄河三角洲盐碱土地区冬小麦合适的肥料效应模型。在冬小麦生长季设置4种不同的氮磷肥用量, 根据&ldquo;3414&rdquo;试验设计8 种不同的肥效试验处理, 以探讨线性加平台、一元二次、平方根和二元二次4 种不同模型的拟合效果。结果显示, 4 种肥料效应模型的拟合结果经检验都达到极显著水平 (P&lt;0.01)。在一元肥料效应模型中, 氮磷一元二次模型拟合效果最好, 最高收益分别为7 448.3 元&middot;hm-2 和7 357.7 元&middot;hm-2, 最佳经济氮磷用量分别为254.4 kg&middot;hm-2 和98.6 kg&middot;hm-2。对比一元与二元模型, 后者拟合效果较好, 最佳经济氮磷用量分别为244.1 kg&middot;hm-2 和94.2 kg&middot;hm-2, 即氮磷肥配比为2.6∶1, 经济效益为7 432.4 元&middot;hm-2, 氮肥农学利用率为6.2 kg(籽粒)&middot;kg-1(N), 磷肥农学利用率为13.8 kg(籽粒)&middot;kg-1(P2O5)。结合拟合度、最佳经济施肥量、经济收益、肥料农学利用率和一元模型的局限性分析得出, 二元二次肥料效应模型最优, 可作为黄河三角洲地区盐碱土冬小麦氮磷肥效模型的最佳选择。</p
