    Analysis of Differently Expressed Proteins in Agaricus bisporus under Heat Stress

    双孢蘑菇[AgaricusbisporusLange(Imbach)]是目前世界上人工栽培最广泛、产量最高、消费量最大的食用菌。它属于稳温结实性的菌类,温度是影响其生长的主要因素。加强和深化双孢蘑菇的分子生物学研究,特别是充分运用现代生物技术研究双孢蘑菇的热胁迫蛋白质表达差异以及耐热分子机理,对培育耐温、高产、优质的菌株具有十分重要的理论和实践意义。 首先,本研究对双孢蘑菇耐温型02菌株和温度敏感型8213菌株进行ITS鉴定。经过ITS鉴定分析,02和8213菌株是双孢蘑菇种内的不同菌株。将02和8213菌株各分为4组:24℃常温培养、先32℃热诱导6h,再37℃热处理6h、37℃热处理12...Agaricus bisporus is the manual cultivation most widespread, the output highest, the consumption quantity biggest edible fungus in the present world.     通讯作者: [email protected][中文文摘]利用蛋白质组学和荧光定量PCR方法,研究双孢蘑菇(Agaricus bisporusLange(Imbach))02菌株在常温与高温胁迫下的蛋白质表达差异,发现了6个差异蛋白质点,经NCBI数据库比对分析,表明6个差异蛋白质分别为HSP70家族的HSS1热激蛋白、异柠檬酸裂解酶、细胞色素P450单氧化酶及3个未知蛋白质.这些蛋白质在双孢蘑菇受热胁迫下具有保护自身稳定的功能.[英文文摘]Agaricus bisporus is a kind of cultivated edible mushroom which has high economic value and important ecology significance.Environment temperature is the main restrained factor in its development,and this greatly limits its cultivation season.In this study,2D-eletrophoresis and real-time PCR are employed to discover the proteomic difference in Agaricus bisporus 02 strain after high temperature induction,and 6 differential expressed protein spots were finally obtained.Compared with the NCBI data base,spot 1 is the heat shock protein,spot 2 is isocitrately ase and spot 3 is cytochrome P450 monooxygenase.The other three spots are novel.Presumably, those proteins confer the thermo tolerance on strain 02 under high temperature.国家基础科学人才培养基金(J0630649);国家自然科学基金(30570254)资

    通过简并引物扩增热球菌(Thermococcus siculi)HJ21高温α-淀粉酶保守区域之间的序列,再利用Site-finding技术获得两端未知序列。构建了在N端添加了His6标签的表达载体pEt-28a-His6-THJA后转化E.coli,在IPTG诱导下表达。进一步纯化后SDS-PAGE电泳检测达到电泳纯,重组酶分子量为50 KDa。重组酶最适作用温度和pH值分别为90℃,pH5.0。重组酶在pH为5.5时最稳定;K+,Sr2+,Mg2+,Na+对α-淀粉酶活力的促进作用不显著,Cu2+,Pb2+,Hg2+,Zn2+,N-溴代丁酰亚胺和三氯乙酸对该酶有显著抑制作用

    根据2002年4-5月在东海122°~123°30′E、29°~32°N海域的海洋综合调查资料,对中型浮游生物网采集的浮游动物样品进行了分析,结果表明:本区春季夜光藻(Noctilucascintillans)平均丰度高达6531ind·m-3(41~48576ind·m-3),夜光藻呈南北高、中间海域低的分布特征;高密集区(>10000ind·m-3)位于东海北部122°30′E以东、31°30′~32°N和南部29°~29°30′N海域;长江口及杭州湾近海(122°15′E以西)丰度低。夜光藻丰度与网采浮游植物总丰度有极其显著的正相关性(R=0.75,P<0.001);与表层水样中硅藻和大型浮游桡足类丰度呈非线形相关关系,前者可用YieldDensity模型拟合,后者可用JohnsonSchumache模型描述。 【英文摘要】 On the basis of the data acquisitioned in the oceanographic comprehensive investigation conducted in the East China Sea (122°00′-123°30′E、29°00′-32°00′N) during the months of April and May, 2002, the zooplankton samples in the sea collected with a net of mesh size 0.160 mm were studied and analyzed. The results show that the average abundance of Noctiluca scintillans was as high as 6 531 ind·m-3 (41-48 576 ind·m-3).
