    Identification of One Unknown Component and Detection Analysis of Free Amino Acids in Porphyra haitanensis

    为了检测分析坛子菜中的游离氨基酸,先采用离子交换树脂层析法分离坛紫菜粗提液中的游离氨基酸,收集3 MOl/l的氨水洗脱液,分别用薄层层析、离子色谱及PITC(异硫氰酸苯酯)-HPlC柱前衍生反相高效液相色谱法对其进行检测分析。结果显示,除含有多种组成蛋白质的氨基酸外还,含有5种未知成分。采用半制备高效液相色谱系统制备分离了其中一种未知成分的衍生物TzC,经真空冷冻干燥后为白色絮状固体,结合核磁共振波谱、高分辨质谱、红外光谱数据,最终鉴定TzC去掉已知取代基团PITC后的成分为乙醇胺,分子式为C2H7nO,相对分子质量为61.08。Free amino acids were separated by ion-exchange column chromatography from the crude extracts of Porphyra haitanensis.Amino acids were eluted with 3 mol/L NH3 ·3H2 O and the eluate was collected.Many protein amino acids and five unknown components were discovered in Porphyra haitanensis by thin layer chromatography,ion chromatography and PITC-HPLC respectively.One unknown derivative component(TZC) was obtained by preparation HPLC.TZC was white flocculent solid after vacuum freeze-drying.According to analysis of NMR,HR-MS,IR spectroscopy data,the molecular structure of TZC had been elucidated.TZC was ethanolamine after removed substituent component of PITC,its molecular formula was C2 H7 NO,molecular weight was 61.08.国家海洋局海洋公益项目(201005020

    Separation and Identification of β-alanine in Sargassum horneri(Turn.) C.Ag.

    非蛋白质氨基酸在抗癌、抗菌、抗结核、抗坏血病等方面有着重要的作用。本文主要对铜藻中的游离氨基酸进行检测分析和部分分离纯化及结构鉴定方面的研究,为更好的开发利用这些天然产物提供技术支持。采用离子交换树脂层析法分离铜藻粗提液中的游离氨基酸,收集3 MOl/l的氨水洗脱液,用PITC-HPlC柱前衍生反相高效液相色谱法对其进行检测分析,结果显示粗提液中除含有多种组成蛋白质的氨基酸外还有3种未知组分,且含量较高的常见蛋白质氨基酸为丙氨酸、脯氨酸、缬氨酸。采用半制备高效液相色谱系统制备分离了其中一种未知组分的衍生物MWz2,经真空冷冻干燥后为略黄固体粉末,结合核磁共振波谱、高分辨质谱、红外光谱数据,最终鉴定MWz2去掉已知取代基团PITC后的成分为β-丙氨酸,分子式为C3H7nO2,分子量为89.09。Non-protein amino acids were found to have many bioactivities such as anti-cancer,antibacterial,anti-tuberculosis,anti-scurvy and so on.Detection,separation and identification of free amino acids in Sargassum horneri(Turn.) C.Ag.were studied in this paper,which provided technical support for development and utilization of these natural products.Free amino acids were separated by ion-exchange column chromatography from the crude extracts of Sargassum horneri(Turn.) C.Ag.Amino acids were eluted with 3 mol/L NH4OH and the eluate was collected.Many protein amino acids and three unknown components were discovered in Sargassum horneri(Turn.) C.Ag.by PITC-HPLC.The main protein amino acids were alanine,proline,valine.One unknown derivative component(MWZ2) was obtained by preparation HPLC.MWZ2 was slightly yellow solid powder after vacuum freeze-drying.According to analysis of NMR,HR-MS,IR spectroscopy data,the molecular structure of MWZ2 had been elucidated.MWZ2 was β-alanine after removed substituent component of PITC,its molecular formula was C3H7NO2,molecular weight was 89.09.国家海洋局海洋公益项目(200805041

    Phytoplankton biomarkers in surface seawater from the northern South China Sea in summer 2009 and their potential as indicators of biomass/community structure

    生物标志物已被广泛应用于重建浮游植物生产力和群落结构变化。该方法假设之一是生标的含量可以反映表层海水的浮游植物生物量,但这个假设还缺乏现场观测的充分验证。对2009年冬季南海北部表层海水颗粒物中主要生标做了分析,利用其含量及比值研究浮游植物的生物量及群落结构的分布。生标含量表明硅藻、甲藻、颗石藻的高值区位于珠江口南部及广东沿岸,在陆坡区也有高值。生标比值显示硅藻在3个类群中的相对比例最高。其中,甲藻/硅藻比值高值区位于陆坡区,这与大洋水(黑潮)的入侵,带来大量暖水性甲藻有很大关系;颗石藻/硅藻比值总体趋势与生物量的分布相反,在近岸少数站位有高值,向外海逐渐增加,主要是因为颗石藻更适于寡营养盐的环境。生标结果所指示的生物量及群落结构的空间分布与前人的调查结果类似,为利用生标重建此区域的浮游植物生产力和群落结构变化提供了依据。Biomarkers have been widely used to reconstruct phytoplankton productivity based on the assumption that biomarker contents could reflect phytoplankton productivity in the surface seawater.However,this hypothesis has not been validated with modern survey.In this study,the contents and ratios of three phytoplankton biomarkers in the surface seawater of the northern South China Sea in winter 2009 were analyzed,to indicate spatial distributions of phytoplankton biomass and community structure.The results show high values for diatoms,dinoflagellates and coccolithophorids near the Pearl River Estuary and Guangdong coastal areas.The community structure indicates diatoms are the dominant phytoplankton species in winter.High values of dinoflagellate/diatom ratio occur near the slope area owing to intrusion of the Kuroshio,which favors the growth of dinoflagellates.The coccolithophorid/diatom ratio displays an increasing trend from the Pearl River Estuary to offshore region due to the advantage of coccolithophorids in oligotrophic environment,which is opposite to the productivity pattern of all biomarkers.These results are consistent with previous studies using phytoplankton cell and pigments,which provides support for the use of biomarker to reconstruct phytoplankton productivity and community structure in the SCS.国家重点基础研究发展计划项目(2007CB815934、2009CB421201);国家自然科学基金项目(40776029

    利用全数字摄影测量得到的高精度点数据,获取不同密度的点数据,建立基于ANUDEM和TIN方法的两种DEM,比较两种DEM的高程中误差,分析数据密度、栅格尺寸与高程中误差的关系。研究结果表明,建立1∶100 00比例尺,具有高程中误差2m左右、栅格尺寸5m以下的ANUDEM,需要的点密度水平为大于7 000个/km2,而TINDEM则为8 500个/km2。同时,当栅格尺寸小于20m、数据密度介于7 000~9 400个/km2时,无论是ANUDEM还是TINDEM,高程中误差都处于一个较稳定的状态。The resolution of digital elevation models is a basic parameter to generate DEM.However,those studies about how to decide the resolution of DEM didn&rsquo;t take the pointsnumber into account.We analyze RMSE of Elevation of ANUDEM and TIN-DEM,whichare generated by the data extracted from aero photos by digital photogrammetric method.The results show that the point density for building ANUDEM with 2mRMSE of elevationand 5mgrid size should be more than 7 000points/km2,while for TINDEM,it should bemore than 8 500points/km2 at 1∶100 00scale level.Moreover,RMSE of elevation keepsstable when the grid size is finer than 20mand the point density is in the range between7 400and 9 400points/km2
