    Association of TolC expression with resistance to tetracycline in Escherichia coli K-12

    随着抗生素的开发与使用,细菌在对多种抗生素的适应过程中逐渐发展出对抗生素耐药的反应机制。TolC是药物排出转运体系的外膜成分,与AcrAB一起形成主要的药物排出泵,有关其表达量与耐药间的关系目前尚不清楚。试验将tolC克隆到pET-32a载体上进行诱导表达,镍柱纯化,免疫新西兰大白兔,获得1:4000的抗血清。Western blotting分析表明,TolC的表达量在耐四环素的大肠埃希菌K-12中比对照组提高50%。细菌TolC高表达试验发现,其MIC从100μg/ml提高到200μg/ml。结果说明TolC的表达量与四环素耐药性直接相关,提示细菌可以通过调节外膜蛋白的表达实现对抗生素的耐受。Association of TolC expression with resistance to tetracycline in Escherichia coli K-12 was studid.TolC was an outer membrane protein,which was bound with AcrAB to form a critical multidrug efflux pump.However,association of TolC amount with drug resistance was ill-defined.In the study,tolC was cloned into pET-32a vector induced expression and its recombinant proteins were purified by Ni-NTA.The purified recombinant proteins were used for preparing of anti-TolC.The prepared antiserum with titer 1:4000 was utilized as the primary antibody in Western blotting.It showed that TolC amount was increased 50% in tetracycline-resistant E.coli K-12 compared to its original control strain.When TolC was overexpressed,the strain showed strong resistance to tetracycline with its MIC from 100μg/ml to 200μg/ml.These findings indicated that TolC amount was directly correlated to bacterial resistance to tetracycline,suggesting that bacteria mounted an adaptive feedback to antibiotics by regulating outer membrane protein expression

    锂离子电池高镍正极材料掺铝的作用及掺铝方法的研究

    为了探究铝对高镍层状氧化物在结构、形貌及性能方面的影响,本文采用两种不同的方式掺铝以制备NCA正极材料(Li Ni0.8Co0.15Al0.05O2):一是固相法即将共沉淀合成的NC前驱体(Ni0.84Co0.16(OH)2)在混锂烧结过程混入铝源(纳米Al2O3或Al(NO3)3);二是共沉淀法即直接在合成前驱体过程中混入铝源(Al2(SO4)3或NaAlO2)即合成NCA前驱体(Ni0.8Co0.15Al0.05(OH)2.05)后再混锂烧结.结果表明,掺铝能够降低阳离子混排程度、维持层状结构的稳定性,改善材料在充放电循环过程的放电电压及中值电压大幅下降的情况,提高其循环性能.其中以NaAlO2为铝源合成NCA前驱体所制备的NCA材料性能最优:在3.0~4.3 V充放电区间,0.1C倍率下首圈放电比容量达198 m Ah·g-1,首次库仑效率可达94.6%,1C倍率下循环200圈后容量保持率达70%

    在常重力静止空气环境中对厚度连续变化的聚甲基丙烯酸甲酯(PMMA)平板表面火焰向下传播进行实验研究,分析了材料厚度对火焰传播特性的影响.结果表明,材料厚度增加时,火焰长度和材料热解长度单调增加,而当材料厚度减小时,材料热解长度和火焰长度先增加后减小.在这两种情况下,均存在热薄材料和热厚材料的转变厚度,其值与文献中使用均匀厚度材料得到的实验结果及理论预测一致;材料厚度小于该转变厚度时,火焰传播速度与材料厚度呈反比关系,大于该厚度时,火焰传播速度不随材料厚度变化,这分别符合热薄材料和热厚材料火焰传播速度的理论模型

    根据2004年2月在南海北部海域陆架区的采样调查,对南海北部Uterm hl浮游植物的群落结构进行了研究.初步研究结果表明,本次调查共发现浮游植物5门106属198种(不包括20种未定种).调查期内,南海浮游植物以硅藻为主,甲藻次之,以颗石藻为主要组份的金藻和固氮蓝藻也占有相当的比例,绿藻仅在珠江口个别站位被检出.菱形海线藻(Thalassionema nitzschioides Grunow)、佛氏海线藻(Thalassionema frauen feldii(Grunow) Hallegraeff)、贺胥黎艾氏颗石藻(Emiliania huxleyi(Lohmann) Hay et Mohler)、柔弱伪菱形藻(Pseudo-Nitzschia delicatissma Heiden)、长海毛藻(Thalassiothrix longissima Cleve et Grunow)、海洋桥球石藻(Gephyrocapsa oceanica Kamptner)、具齿原甲藻(Prorocentrum dentatum Stein)、具槽帕拉藻[Paraliasulcata(Ehrenberg) Cleve]和旋沟藻(Gyrodiniumspirale Bergh)为群落的优势种.南海北部浮游植物以广温、广布型为主,其次是暖水性种,热带、亚热带和冷水性种都较少.调查区浮游植物的细胞丰度介于3.8~2 810.3个/cm3,平均值为206.4个/cm3;浮游植物垂直分布总体特点是密集区位于水体表层,由水体表层到底层,细胞丰度逐渐减少;表层浮游植物细胞丰度的平面分布高值区位于调查区珠江口南部和南部海域.浮游植物群落的多样性分析显示调查海域中部的多样性较高,近岸和外海区则较低.应用典范对应分析(CCA)发现盐度和光照的梯度分布是影响南海北部浮游植物物种分布格局最重要的因素,磷酸盐和温度的梯度分布也起一定的作用.根据CCA的分析结果,将海区浮游植物物种划分为5个组,分别为:Ⅰ.高温度、高盐度、高光照和低营养盐组;Ⅱ.高温度和高光照组;Ⅲ.高磷酸盐和硅酸盐组;Ⅳ.低光强高盐度荫生物种组;Ⅴ.高硝酸盐组.They were presented that the phytoplankton species composition and cell abundance data based on the investigation in northern South China Sea(17°~23° N,111°~117° E) which was carried out from 8 th February to 4th March 2004 with 149 samples taken from 30 grid stations.A total of 195 taxa(not include uncertain species) belonging to 110 genera of 5 phyla were identified by Uterm hl method,in which Bacillariophyta were predominant and coexisted with Dinophyta.Chrysophyta which was mainly composed of coccolithophores and diazotrophs Cyanophyta were also common found in survey area.Few Chlorophyta were checked out at two stations near Zhujiang(Pearl River) Estuary.The dominant species in turn were Thalassionema nitzschioides Grunow,Thalassionema frauenfeldii Hallegraeff,Emiliania huxleyi(Lohmann) Hay et Mohler,Pseudo-nitzschia delicatissma Heiden,Thalassiothrix longissima Cleve & Grunow,Gephyrocapsa oceanica Kamptner,Prorocentrum dentatum Stein,Paralia sulcata(Ehrenberg) Cleve and Gyrodinium spirale Bergh,etc.Most of phytoplankton taxa found in survey area was cosmopolitan species,warm-water species are also common,there were just few trophic-water,subtrophic-water or cold-water species.The cell abundance of phytoplankton ranged from 3.8~2 810.3 cells/cm3,the average was 206.4 cells/cm3.Phytoplankton cell abundance were concentrated in surface water and reduced rapidly from surface layer to the bottom.Two high phytoplankton cell abundance regions can be discriminated at southern waters of Zhujiang Estuary and southern part of the survey area, this two high phytoplankton cell abundance regions were mainly caused by higher nutrients concentrations and ecotone respectively.The Shannon-Wiener diversity indices and Pielou's evenness of phytoplankton community were high in centre of survey area.Canonical correspondence analysis(CCA) was applied to explore the relationship between phytoplankton species and environmental parameters,the results showed that salinity and photosynthetically active radiation(PAR) gradient were the most important factors,and phosphate concentration and temperature gradient were secondary important factors,which influencing the phytoplankton species distribution patterns.According to CCA the first two ordination axes between the phytoplankton species and environmental factors,5 phytoplankton species groups can be discriminated,they are: Group Ⅰ.prefer to high temperature high salinity high light low nutrient environment;Group Ⅱ.prefer to high temperature high light environment;Group Ⅲ.prefer to high phosphate high silicate environment;Group Ⅳ.prefer to low light high salinity environment;Group Ⅴ.prefer to high nitrate environment.国家自然科学基金重点项目(9021102190211020);; 国家自然科学基金项目(40676089

    Vapor growth and photoconductivity of single-crystal nickel-phthalocyanine nanorods

    Nickel-phthalocyanines (NiPc) with planar aromatic structures are ideal building blocks for organic nanostructures. They can self-assemble into stacks through pi-pi interaction, exhibit high thermal and chemical stabilities, and possess outstanding electrical and optical properties. Herein, single-crystal NiPc nanorods were synthesized by a facile vapor transfer deposition method. Their nanostructures and compositions were investigated by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fourier-transform infrared (FTIR) in detail. The deposited NiPc nanorods were found to be the beta-phase single crystals. Moreover, the NiPc nanorod-based devices were fabricated and exhibited high photocurrent upon white-light illumination. This indicates that the NiPc nanorods can be considered as a candidate material for fabricating photoelectric devices

    Vapor growth and photoconductive property of single-crystalline MoO3 nanosheets

    Molybdenum trioxide (MoO3) nanosheets were synthesized on Si substrate by a facile physical vapor deposition method and fully characterized by means of scanning electron microscopy, transmission electron microscopy, X-ray diffraction spectrometry, energy dispersive X-ray spectroscopy, and Raman spectroscopy. The characteristic spectra of both Raman and XRD demonstrated that the as-deposited nanosheets are highly pure and have perfect crystallinity of the orthorhombic-phase MoO3. Moreover, prototype devices based on individual MoO3 nanosheets were fabricated and exhibited promising photoconductive response upon the white light illumination. The result indicated that the as-deposited MoO3 nanosheets were of potential applications in optoelectronic devices
