    Inhibitory effect of anti-human NRP-1 monoclonal antibody on hepatocellular carcinoma cell line HepG2 and its mechanism in vitro

    目的研究抗人神经纤毛蛋白-1(Neuropilin1,NRP-1)单克隆抗体对肝癌Hep G2细胞的生长抑制作用及其机制。方法小鼠腹水法制备抗NRP-1单克隆抗体(NRP-1 m Ab)并用r Protein A亲和柱纯化抗体,间接ELISA检测抗体的滴度水平。Western blot检测NRP-1 m Ab对Hep G2细胞的特异性,细胞免疫荧光和流式细胞术检测NRP-1蛋白在肝癌细胞株Hep G2上的表达,MTT法检测NRP-1 m Ab对Hep G2的生长抑制作用,Western blot检测ERK1/2、P-ERK1/2、Akt、P-Akt蛋白的表达水平。结果 SDS-PAGE和间接ELISA检测纯化的NRP-1 m Ab纯度为95%以上,效价为1×10~(-6);Western blot检测结果显示NRP-1 m Ab可与Hep G2细胞膜上的NRP-1蛋白特异性结合。细胞免疫荧光染色结果显示NRP-1定位于Hep G2细胞膜,流式细胞术结果显示NRP-1蛋白在Hep G2细胞上表达水平较高;MTT法检测结果显示NRP-1 m Ab对Hep G2细胞有生长抑制作用。Western blot检测到在不同浓度NRP-1 m Ab作用下,Hep G2细胞裂解液P-ERK1/2、P-Akt蛋白的条带信号逐渐减弱。结论纯化的NRP-1m Ab能抑制Hep G2细胞的生长,其抑制作用是通过EGF和HGF信号通路实现的。The aim of the experimental is to investigate the inhibitory effect of anti-human nerve cilia protein1(Neuropilin-1,NRP-1) monoclonal antibody(NRP-1 mAb) on hepatocellular carcinoma cell line HepG2 and its mechanism in vitro.Anti-human NRP-1 monoclonal antibody(NRP-1 mAb) was prepared from mouse ascites and purified by rProteinA affinity column assay.The titer of antibody was determined using indirect ELISA assay;the characteristic of NRP-1 mAb binding to NRP-1 was determined using Western blotting;the expression of NRP-1protein in hepatocellular carcinoma cell line HepG2 was determined using immunofluorescence assay and flow cytometry assay.Growth inhibition of HepG2 cells treated with different concentrations of NRP-1 mAb was determined using MTT assay,while Western blotting was used to detect the expression levels of ERK1/2,P-ERK1/2,Akt and P-Akt proteins.The results of SDS-PAGE and indirect ELISA showed that the purity of purified NRP-1mAb was more than 95%and the titer was 1×10~(-6).Western blotting analysis suggested that NRP-1 mAb could bind specifically to NRP-1 on HepG2 cell;immunofluorescence staining showed that NRP-1 was located in the membrane of HepG2 cells.Flow cytometry analysis showed that the expression level of NRP-1 on HepG2 cell was relatively high.Western blotting analysis suggested that P-ERK1/2 and P-Akt expression levels were down-regulated after having incubated HepG2 cells with different concentrations of NRP-1 mAb.In conclusion,NRP-1 mAb could inhibit the growth of HepG2 cells(P<0.05),and its inhibitory effect is achieved by reducing the P-ERK1/2 and P-Akt expression.南京军区医学科技创新项目(12MA061,15MS104

    Synthesis of Eu(BTF)_3phen and Study on Its Property

    采用常温搅拌法制备了配合物Eu(bTf)3PHEn(bTf=苯甲酰三氟丙酮,PHEn=邻菲罗啉),改善稀土铕配合物的荧光性能。对配合物的热稳定性和荧光性质进行了测定,结果表明其具有良好的热稳定性,在362 nM激发光激发下,能发射强烈的、来自于Eu3+离子的特征红光,最大发射峰在612 nM,半峰宽大约为8 nM。对邻菲罗啉中性配体进行了两种不同的分子修饰,荧光光谱测试表明,邻菲罗啉配体上引入不同数目的基基团,在一定程度上改善了Eu(bTf)3PHEn配合物的荧光性能。In order to improve the fluorescence properties of europium complex, Eu(BTF)3phen( BTF =Benzoyltrifluoroacetone, phen =o-phenanthroline) was synthesized by the method of stirring method at room temperature.The results showed that the complex has good thermal stability.The complex emitted a strong characteristic red light which was from Eu3 +under the exciting light of 362 nm.The maximum emission peak was at 612 nm.The full width at half maximum( FWHM) was 8 nm.Two different molecular modifications of the phenanthroline neutral ligands were carried out.The fluorescence spectra analysis showed that the fluorescent properties of Eu(BTF)3phen was improved by the introduction of different numbers of neighborhood groups onto the phenanthroline ligands

    微型铣刀外径视觉测量系统的照明设计

    为了满足微型铣刀外径视觉测量系统的照明需求,设计并制作了一种基于柯勒照明原理的平行光源。平行光源由白光lEd、孔径光阑和3组组合透镜组成,使用zEMAX对光路进行了仿真。设计并加工了多级可调金属圆筒机械结构,将光源各部分器件固定组合,光源总长为142 MM。搭建了微型铣刀外径视觉测量系统,并分别使用漫射背光光源和平行背光光源对铣刀进行照明成像。实验结果表明,平行背光照明提高了铣刀测量系统的测量精度,降低了铣刀的测量离差

    Three-dimensional Porous Cu_6Sn_5 Alloy Anodes for Lithium-ion Batteries

    以三维多孔泡沫铜为基底,通过直接电沉积的方法制备锂离子电池Cu6Sn5合金负极材料.发现合金表面大量的微孔和“小岛”不仅增大电极的表面积,而且显著缓解充放电过程中的体积变化.测得三维多孔Cu6Sn5合金的初始放电(嵌锂)容量为620mAh·g-1,充电(脱锂)容量为560mAh·g-1,库仑效率达到90.3%,具有较好的循环性能.扫描电子显微镜(SEM)结果显示,在泡沫铜基底上制备的Cu6Sn5合金电极具有比通常的铜片基底更好的结构稳定性,经过50周充放电循环后无明显的脱落现象.Three-dimensional porous Cu6Sn5 alloy electrodes were prepared by electroplating using copper foam as current collector.The micro-holes and small islands on surface of the Cu6Sn5 alloy increased largely the surface area of the electrode,and improved significantly the ability of the electrode in buffering the volume change in process of charge/discharge when the Cu6Sn5 alloy was employed as anode in a lithium-ion battery.Galvonostatic charging/discharging results demonstrated that the initial discharge(lithiation)and charge(delithiation)specific capacities of the Cu6Sn5 alloy electrode were 620 mAh·g-1 and 560 mAh·g-1,respectively.It demonstrated that the Cu6Sn5 alloy electrode exhibited a large initial coulomb efficiency(90.3%)and good capacity retention.SEM(scanning electron microscope)results illustrated that the Cu6Sn5 alloy deposited on copper foam substrate was more stable than that on a conventional copper substrate,and displayed no obvious exfoliation after 50 charge/discharge cycles.国家重点基础研究发展规划项目(973)(2002CB211804)资

    Preparation and Capacity Fading Mechanism of Tin Thin Film as Anode of Lithium-ion Battery

    以电镀的方法在铜基底上沉积薄膜锡作为锂离子电池负极材料.运用X射线衍射、扫描电镜、电化学循环伏安、电化学充放电和交流阻抗等多种方法对其结构和性能进行表征和研究.结果表明所制备的薄膜锡电极主要为四方晶系结构,其初始放电(嵌锂)容量为709mAh?g-1,充电(脱锂)容量为561mAh?g-1.电化学循环伏安研究发现在嵌/脱锂过程中薄膜锡经历了多种相变过程.电化学阻抗谱结果说明,首次嵌锂过程中当电极电位达到1.2V在电极表面形成SEI膜,而当电极电位低于0.4V表面SEI膜出现破裂,归因于体积膨胀所致.SEM研究表明30次充放电循环后薄膜锡负极出现龟裂现象.Tin thin film coated on Cu substrate as anode of lithium-ion battery was prepared by electroplating. Its structure and properties were characterized and studied by X-ray diffration, scanning electron microscopy, cyclic voltammetry, charging/discharging test and AC impedence method. XRD patterns indicate that the tin thin film exhibits a structure of tetragonal crystal. The first discharge and charge capacities of the tin thin film electrode were determined to be 709 and 561 mAh?g-1, respectively. Cyclic voltammetric results illustrated that multi-phase changes occurred during the lithiation and delithiation. Electrochemical impedance spectros- copy (EIS) results indicated that SEI film was begun to form on the surface of tin thin film electrode at 1.2 V, and then break down below 0.4 V because of large volume expansion. SEM investigations revealed that the tin thin film electrode appeared serious cracks after 30 charging and discharging cycles.国家重点基础研究和发展规划(973项目)(No.2002CB211804)资助项目
