    肾透明细胞癌差异表达基因的筛选及其核心基因相应miRNA和lncRNA预测分析

    【目的】筛选肾透明细胞癌(ccRCC)中差异表达基因并其相关miRNA和lncRNA,寻找可作为ccRCC诊断及治疗的生物标志物。【方法】通过从TCGA数据库和GEO数据库筛选ccRCC差异表达基因并取交集,对差异表达基因进行GO功能分析和KEGG通路富集分析,使用MCODE软件筛选出差异表达基因中的核心基因,并利用mirDIP数据库和STARBASE数据库对核心基因上游的miRNA和lncRNA进行预测。【结果】共筛选出427个差异表达基因,这些差异表达基因在GO功能分析上主要与催化活性、受体活性和细胞黏附分子活性等功能相关,KEGG信号通路富集结果则表明其主要与PPAR、Rap1以及细胞因子受体相互作用等信号通路相关。从这些差异表达基因中筛选出11个核心基因,并预测到134个相应miRNA,接着对5个与ccRCC总体生存率相关的miRNA在STARBASE中共预测到6个相应lncRNA。最后将lncRNA、miRNA、核心基因以及相应信号通路在CYTOSCAPE中构建出一个互作网络。【结论】利用生物信息学技术将TCGA数据库与GEO数据库结合起来,筛选出ccRCC中差异表达基因,并对其中的核心基因进行miRNA与lncRNA预测分析,为后续找到可用于ccRCC临床诊断的靶标与治疗的靶点提供了有力帮助。国家自然科学基金(81570748);;福州总医院杰出青年培养专项(2017Q05

    肾透明细胞癌Caki-1细胞系差异表达基因的生物信息学分析

    目的比较肾透明细胞癌Caki-1细胞系与正常肾上皮细胞系ASE-5063中的差异表达基因(DEGs),寻找潜在的肾透明细胞癌特异性分子标志物。方法利用GEO数据库自带的GEO2R在线分析工具分析基因芯片GSE78179,将筛选出的DEGs分别导入Metascape、STRING以及Cytoscape进行综合分析并筛选出核心基因。最后使用Fun Rich等软件对筛选出的核心基因进行GO和KEGG富集分析。结果共筛选出562个DEGs,其中上调基因345个,下调基因217个。进一步使用MCODE筛选出36个关键基因,GO功能分析发现这些基因与细胞粘附分子活性、趋化因子活性、细胞通讯和信号转导等密切相关;KEGG通路富集结果则表明差异基因主要集中在趋化因子信号通路、TNF信号通路以及NF-κB信号通路等多种与肿瘤相关的通路上。结论运用生物信息学方法筛选出肾透明细胞癌Caki-1细胞系中DEGs,其中数个核心基因广泛参与多种肿瘤的病理进程,但尚未在肾透明细胞癌有相关研究报道,提示其可能是治疗肾透明细胞癌的潜在靶点。国家自然科学基金(81570748);;福州总医院杰出青年培养专项(2017Q05

    要旨:平成29 年10 月から国保特定健診の結果、糖尿病性腎症重症化リスクが高い対象者を抽出し、重症化予防保健指導に取り組んでいる。しかし、本事業により新たに対象となった治療中の患者への保健指導については、医療との連携等に関する従事者のスキル向上が課題であった。そこで、平成30 年度に「生活習慣病予防対策ワーキング」が中心となり、課題の整理や医師等を交えた事例学習会、検討会を開催し、保健指導の力量形成に関する取組を展開することで、従事者が「事業をこなす」という意識から重症化予防の重要性を理解し、かかりつけ医等と連携した保健指導の実施により重症化予防の徹底に向け日々取り組んでいる。Article信州公衆衛生雑誌 14(1): 26-27(2019)journal articl

    The First-Year Experience or Freshman Seminar Program has been instituted in many Japanese universities and colleges. We have a course called "Basic Seminar" for all students incoming freshman. However, it sometimes becomes a major burden for teachers to help various types of students and arouse their interest in the age of "universalization". To arouse students\u27interest and to reduce our burden at the same time, we have tried efficient and effective use of ICT(Information and Communication Technology). In this paper, we, focusing on "Basic Seminar" as a program for active learning, propose the system designs for utilizing ICT in theseminar and show several cases of their applications

    脂多糖激活所致大鼠抑郁样行为及对海马神经细胞钾电流变化的影响

    采用细胞因子刺激剂脂多糖(lipopolysaccharide,LPS)为免疫激活手段,研究LPS诱导的免疫激活产生的抑郁样行为及对海马神经细胞电压依赖钾电流变化的影响。应用膜片钳技术对海马神经细胞钾电流进行全细胞记录,比较抑郁样行为大鼠与正常大鼠钾离子通道电流密度和激活特性的变化。结果发现,与生理盐水对照组相比,一次LPS注射后2hr,实验组动物产生抑郁样行为,同时急性观察的海马神经细胞的钾离子通道的电流密度呈现显著升高(p<0.01);而一次LPS注射后24hr,动物的抑郁样行为消失,且急性观察的海马神经细胞的钾离子通道与对照组相比较,其电流密度和激活曲线没有显著性变化。结论:LPS诱导的抑郁样行为,与LPS诱导的海马神经细胞电压依赖钾电流的上调在时程上同步,提示钾离子通道可能参与免疫激活所致的抑郁样行为

    Generation of a phase-conjugate wave in a saturable-dye-doped planar waveguide

    A phase-conjugate wave (PCW) by degenerate four-wave mixing (DFWM) in a nonlinear-optical waveguide was observed with an Ar-ion laser. The planar waveguide consists of a transparent poly-vinyl alcohol (PVA) layer partly doped with erythrosin B. The DFWM excitation was performed as follows: two pump waves impinged in the interaction region of the waveguide from the outside and a probe wave was introduced into the guiding layer by a prism coupler. The PCW reflectivity of 6.3 × 10⁻⁵ outside the prism coupler was obtained at a pump intensity of 1.2 W/cm² in the PVA film having a thickness of about 80μm and the product of a small-signal absorption coefficient and the film thickness of 0.97; the probe intensity was chosen to be 0.2 of the pump intensity. If the prism-outcoupling efficiency is assumed to be 100 percent, this corresponds to the PCW reflectivity of 4.2 × 10⁻⁴ inside the waveguide by taking into consideration the prism-coupling efficiency of 15 percent

    通过对鼓浪屿古榕树的普查,选择其健康诊断的生长指标和生理指标,包括当年生枝叶、果实生长状况、叶绿素荧光等,对不同生长势的古榕树进行分析,旨在探讨基于生长指标与生理指标相结合的古榕树的健康诊断方法,为古榕树的养护与复壮提供依据。结果表明,生长指标与生理指标能较一致地指示古榕树的健康状况。因此,可通过生长指标与生理指标相结合的方法诊断古榕树的健康状况。Based on the general investigation and analysis of the ancient trees of Ficus microcarpa at Gulangyu by use of the growth indices and the physiological indices,such as the growth status of annual branches,leaves and fruits and the chlorophyll fluorescence,the diagnosis of the ancient trees of Ficus microcarpa with different growth vigor was discussed to provide evidence for their maintenance and rejuvenation.The results indicated that the growth indices and the physiological indices could consistently instruct the health status of the ancient trees of Ficus microcarpa.Thus the diagnosis by the combination of the growth indices and the physiological indices could be applied to the ancient trees of Ficus microcarpa.鼓浪屿万石山国家级风景名胜区科技项目资助(GW2008

    人工呼吸器装着患者は,術後の創痛や気管チューブの痛みなど様々な痛みを感じているといわれている。しかし,当院ICU での痛みの評価は,看護師の知識や経験により大きく差が出る現状があった。そこで人工呼吸器装着患者の痛みの評価方法を統一するためBehavioral Pain Scale(以下BPS)を導入した。BPS 導入後は鎮痛剤の使用量が増加し,離床拡大につながったが,人工呼吸期間やせん妄期間の短縮には至らなかった。今後は人工呼吸期間やせん妄期間の短縮に向けての検証が必要である
