    Investigation of lithium manganese oxides with large tunnel structure as electrode material in Li ion batteries

    锂锰氧化物材料是一类重要的锂离子电池电极材料。文中报道一种新型大隧道结构镁锰复合氧化物〔结构类型:钡镁锰矿(TOdOrOkITE)型〕的合成方法及其电化学性能表征。X-光衍射(Xrd)谱证实了合成物具有所预想的(3x3)隧道结构。在慢速循环伏安图中可发现该类电极材料分别在3.35V及2.45V(VS.lI)出现一对锂离子的脱出-嵌入峰。充放电实验结果表明:该类大隧道结构复合氧化物可作为一种3V锂离子电池的电极材料,当充放电电流密度为0.1MA/CM2时,材料的初次容量可达158MAg/H,经过4次充放电循环后其容量仍可保持在约130MAH/g。Lithium manganese oxide is one kind of the important electrode material of Li ion batteries.In this paper, investigations of a novel Li Mn oxide electrode material with large tunnel structure such as todorokite type electode material For Li ion batteries are carried out in the lab.It is demonstrated that the material synthesized has a todo rokite structure (i.e.3×3 tunnel)using X ray diFFraction(XRD) method.It is also observed that a couple of deintercalation/intercalation peaks of lithium ion appear at 3.35V and 2.46V (vs.Li) respectively in the slow rate cyclic voltammograms.In addition,it is Found that the material shows quite good cyclic perFormance,the initial capacity of the material is 158mAh/g,and its discharge capacity is still kept at about 130mAh/g aFter Four cycles at charging/discharging current density of 0.1mA/cm 2.福建省自然科学基金;杰出青年基

    SIRT7基因低表达胶质瘤细胞株代谢特征的NMR分析

    肿瘤是一种代谢疾病,癌基因表达对肿瘤细胞代谢的影响是目前肿瘤研究的热点之一.本文利用基于核磁共振氢谱(~1H NMR)的代谢组学方法对癌基因SIRT7低表达胶质瘤细胞株的代谢特征进行分析,寻找与SIRT7基因表达相关的特征性代谢物和代谢通路.分析结果表明,SIRT7基因低表达组与对照组细胞的代谢轮廓存在显著性差异,其细胞水溶性萃取物中有22种代谢物浓度发生明显变化.与对照组相比,SIRT7基因低表达胶质瘤细胞株中乳酸、甘氨酸、谷氨酸等12种代谢物浓度升高;缬氨酸、亮氨酸、赖氨酸等10种代谢物浓度降低.通路富集分析提示氨酰-tRNA生物合成、氨基酸代谢等代谢通路与SIRT7低表达密切相关.以上结果为进一步阐明癌基因SIRT7调控胶质瘤细胞代谢的作用机制提供了理论依据.厦门市科技计划社会发展项目(3502Z20184064

    抗人DR5单克隆抗体的制备、鉴定及活性分析

    目的制备抗人DR5单克隆抗体(mAb),鉴定其特性,并进行生物学活性分析。方法以纯化的可溶性DR5(sDR5)免疫Balb/c小鼠,杂交瘤技术制备抗人DR5mAb;运用ELISA、SDS-PAGE电泳方法测定抗DR5mAb与sDR5结合的特性;Ig亚类ELISA试剂盒鉴定抗DR5mAb亚类;间接ELISA法检测腹水mAb效价;流式细胞仪检测肿瘤细胞表面DR5的表达水平;流式细胞仪检测抗DR5单克隆抗体(mAb)诱导肿瘤细胞凋亡的功能。结果获得1株可分泌抗DR5mAb的杂交瘤细胞系R150。SDS-PAGE电泳检测证实,获得的R150可特异性地识别DR5;R150的Ig亚类为IgGI(λ型);腹水效价为1×106;通过流式细胞仪可敏感地检测到肿瘤细胞表面DR5的表达水平及R150诱导肿瘤细胞凋亡情况。结论获得1株可分泌抗DR5mAb的细胞系R150,抗体具有效价高、特异性强等特点并能有效诱导肿瘤细胞凋亡,具有较好的应用价值

    Generating High-concentration Solution of Reactive Oxygen Species by Strong-field Ionization Discharge

    为优化氧活性粒子(rOS)在水中的生成条件,并为rOS溶液生成装置提供优化系统参数的依据,研究了rOS质量浓度在水温、PH值、O2给气体积流量、rOS投加体积质量,以及系统气压(混溶压力)等因素作用下的变化规律。实验中,气态rOS在强电离条件下通过介质阻挡放电生成,以O3计,通过O3检测仪测定其浓度;水中rOS质量浓度采用dPd分光光度法测定,用CrS来表示。实验结果表明:水温、PH值与CrS呈极显著负相关(相关系数P0.05);CrS在水温分别为16℃与24.5℃之间、PH=6.5与PH=7.0之间、O2给气体积流量为2 l/MIn与3 l/MIn之间均为差异不显著(P>0.05),其余各水温、PH值、O2给气体积流量之间均为差异显著(PO2给气体积流量>系统气压与O2给气体积流量交互作用。To optimize the generating condition of reactive oxygen species(ROS) in solution, and to provide a reference for improving ROS preparation system, we investigated the effects of several parameters, including solution temperature, solution pH, O2 input, ROS dosages, and system pressure, on the concentration of obtained ROS solution.Gaseous ROS was generated in a strong-field ionization condition induced by under dielectric barrier discharge(DBD), and its dosage represented by O3 was measured by ozone monitor.ROS in solution was caught by DPD(N, N-diethyl-p-pHenylenediamine), which was measured by DPD spectrophotometry, and the concentration of ROS solution was denoted by CRS.According to the experiments, both water temperature and pH have significant negative correlation with CRS(relativefactor P0.05).The differences between CRS are insignificant(P>0.05) under conditions of solution temperature of 16 ℃, 20 ℃, 24.5 ℃, solution pH of 6.5 and 7.0, as well as O2 input of 2 L/min and 3 L/min(P>0.05), but they are highly significant(P<0.01) or significant(P<0.05) under other tested conditions.Moreover, CRS significantly increases with the decrease of miscibility pressure(P<0.01), and it is significantly affected by the interaction between miscibility pressure and O2 input under higher ROS dosages(P<0.01).Lower water temperature, lower pH, higher ROS dosages, and lower miscibility pressure are all beneficial to increasing the concentration of ROS(CRS), which is significantly affected by the change of several parameters including solution temperature in the lower range, pH around 7, O2 input in the range of larger amount, etc.On the condition of high ROS dosage input, CRS is influenced by miscibility pressure, O2 input, and the interaction between miscibility pressure and O2 input in a descending order.国家高技术研究发展计划(863计划)(2012AA062609); 国家科技支撑计划(2013BAC06B02); 国家杰出青年科学基金(61025001)~

    ERK干扰质粒的构建及对胃癌细胞株BGC823 DcR3表达的影响

    目的探讨胃癌发展的分子机制,通过构建ERK1/2shRNA真核表达载体,体外研究其对人胃癌细胞株BGC823ERK1/2蛋白及DcR3表达水平的影响。方法应用PRNAT-U6.1/Neo载体构建ERK1/2基因shRNA重组质粒,经脂质体法导入BGC823细胞中,分别设置对照组、干扰组和U0126抑制剂组。Westernblot法检测转染后BGC823细胞及抑制剂使用后ERK1/2蛋白的表达变化,荧光显微镜检测质粒自带GFP基因的表达情况确认转染效率,ELISA法检测各组细胞上清中DcR3分泌蛋白的表达特点。结果成功构建ERK1/2基因shRNA重组质粒。证明了ERK1/2蛋白的表达与DcR3的分泌水平在BGC823细胞株中呈正相关。结论 ERK1/2干扰质粒明显降低BGC823细胞的ERK1/2蛋白表达水平,ERK信号通路对DcR3的分泌具有重要调控作用,为其下游调控机制的研究奠定了基础

    肝癌发展中FasL及其受体Fas/DcR3的表达分析

    目的研究小鼠肿瘤发生早期肿瘤坏死因子超家族成员FasL及其受体的表达特点,及其与免疫调节相关分子表达的时空关系,探讨其在诱导肿瘤免疫耐受中的作用。方法建立小鼠实体瘤模型,采用RT-PCR方法检测肿瘤组织中可溶型FasL、Fas及DcR3、TGF-β、IL-10在肿瘤发生不同时相的表达,同时,应用ELISA方法定量检测血清中TGF-β、IL-10的表达。结果在早期的肿瘤组织中,Fas的表达先于DcR3,其后DcR3大量表达,而Fas的表达则下调直至丢失,DcR3、FasL同时表达并上调,TGF-β和IL-10也随肿瘤的表达而上调。TGF-β同DcR3的表达具有空间位置的一致性。FasL、DcR3、TGF-β和IL-10的表达水平同肿瘤的生长呈正相关。结论随着肿瘤不断的生长,肿瘤局部的免疫应答逐渐趋向于负调节,FasL、DcR3在诱导肿瘤免疫耐受的过程中可能发挥着重要作用

    Genomic organization and tissue-specific expression analysis of hepcidin-like genes from black porgy (Acanthopagrus schlegelii B.)

    【英文摘要】Hepcidin is an antimicrobial peptide and putative iron regulatory hormone previously described in mice and humans. Dozens of fish hepcidins have been isolated and characterized so far. Here we present seven hepcidin-like cDNA sequences named AS-hepc1–7, amplified from the normal commercially cultured fish (black porgy) by RACE–PCR. Sequence analysis reveals that these seven potential hepcidin peptides have highly conserved sequences with other known hepcidins, but they are different from each other in constitution and characteristics of predicted mature amino acids. Based on the study, it is deduced that AS-hepc1–7 represent different variants of a family of hepcidin genes in black porgy. To understand the organization of these hepcidin-like genes, we sequenced AS-hepc2 DNA, AS-hepc3 DNA, AS-hepc4 DNA, AS-hepc7 DNA and AS-hepc2 upstream region; and all of the four genomic DNAs consisted of two introns and three exons, the same organization as other reported hepcidins. The tissue-specific gene expression of hepcidins in normal black porgy was evaluated using RT–PCR and dot blot approaches. RT–PCR showed that transcripts of hepcidin-like mRNAs were present in each tested tissue of normal juvenile black porgy, including liver, spleen, kidney, heart, brain, stomach, intestine, gill, skin and blood, but abundant hepcidin-like mRNA transcripts were only detected in the liver, kidney, spleen, intestine and stomach by dot blot assay. In addition, using dot blot and Northern blot approach, a significant increase of hepcidin mRNA transcription was observed in the liver within 48 h after immersion in a suspension of live bacteria, which suggested that the expression pattern of hepcidin-like genes in black porgy might be different in the liver from the other tissues as previously reported in several hepcidin studies
