    Construction of suppression subtractive hybridization(SSH) library of copepod Pseudodiaptomous annandalei and its ferritin cDNA cloning and differential expression under nickel stress

    为探讨镍胁迫下桡足类的分子响应机制,以安氏伪镖水蚤为研究对象,利用抑制性消减杂交(SuPPrESSIOn SubTrACTIVE HybrIdIzATIOn,SSH)技术,构建了镍胁迫下安氏伪镖水蚤SSHCdnA文库,并随机挑取生长菌落140个克隆子,进行菌液PCr鉴定,计算文库重组率为98.6%,文库容量为1.12x106Cfu.将重组子测序,经blAST一致性搜索比对分析发现,有一重组片段含有铁蛋白保守结构域,该片段大小为859bP,连续编码170个氨基酸残基,gEn-bAnk登录号为Jk312601.半定量PCr证实,镍胁迫24H后,安氏伪镖水蚤中铁蛋白表达显著上调.本文库的成功构建及铁蛋白CdnA片段的获得为进一步研究镍胁迫下桡足类的分子响应机制奠定了基础.To study the molecular response mechanisms of copepod to nickel stress,a suppression subtractive hybridization(SSH) cDNA library of Pseudodiaptomous annandalei under nickel stress was constructed by using SSH technique,and a total of 140 clones were randomly picked from the growing colonies and identified by PCR.The recombinant rate of the library was 98.6%,and the volume of the library was 1.12×106 cfu.After the recombinant plasmids were sequenced,a partial cDNA fragment of ferritin was recognized based on BLAST searches in NCBI,with a size of 859 bp and continuously encoding 170 amino acid residues.The semi-quantitative PCR results showed that the ferritin cDNA under 24 h nickel stress was distinctly up-regulated.The successful construction of the SSH library and the obtaining of ferritin cDNA fragment would supply basis for the further study of the molecular response mechanisms of copepod to nickel stress.国家自然科学基金项目(40876060)资

    新型铕络合物用于HBsAg的时间分辨荧光免疫检测

    利用稳定的新型铕荧光络合物BHHCT与Eu3 + 标记羊抗人HBsAb和Eu3 + 标记BSA -SA ,建立定量测定血清HBsAb的Eu3 + -BHHCT -HBsAb -TRFIA法和Eu3 + -BHHCT -BSA -SA -TRFIA法。结果表明 ,这两种方法最低检出值分别为 0 .2ng/mL和 0 .0 5ng/mL ,标准曲线范围均为 0 - 10 0ng/mL ,批内变异系数CV均小于10 % ,后者回收率为 85 % - 115 %。用Eu3 + -BHHCT -BSA -SA -TRFIA法与ELISA法同时检测 118份血清样品 ,结果表明前者的检出率比后者

    选择塔里木河北源及干流区12个代表气象站1959-2004年的年降水系列资料,采用线性回归、平滑方法和Morlet小波功率谱等分析方法,对比分析了塔里木河北源与其干流两个不同区域降水变化趋势与多尺度特征.趋势分析表明:近46a来,两区域年降水都存在增加的趋势,但北源区增加的幅度要明显大于干流区;两个区域在1986年前后,降水增加更趋明显.多尺度分析显示,两区域年降水存在集中于2~8a尺度带上的显著周期.其中,北源区域年降水尺度较为分散,大部分站点年降水集中在2~4a尺度带上,准5a~准8a尺度上也有少量分布;干流区域则主要集中于准3a尺度上.另外,由于时间序列长度的限制和其它不确定因素的影响,两区域部分站点检测出的16a以上的年代际尺度不显著

    为探讨镍胁迫对桡足类影响的分子机制,以安氏伪镖水蚤(PSEudOdIAPTOMuS AnnAndAlEI)为研究对象,采用Cd-nA末端快速扩增技术(rACE)方法获得了安氏伪镖水蚤精氨酸激酶(ArgInInE kInASE,Ak)CdnA全序列,全长1 180bP,开放阅读框1 068bP,编码355个氨基酸残基,预测蛋白质分子质量为39.43ku.blAST分析结果显示,安氏伪镖水蚤AkCdnA与罗氏沼虾(MACrObrACHIuM rOSEnbErgII)、多齿新米虾(nEOCArIdInA dEnTICulATA)和光滑双脐螺(bI-OMPHAlArIA glAbrATA)Ak基因具有较高的同源性(77%~82%).荧光定量PCr结果显示:低质量浓度镍(10μg/l)对Ak转录水平呈现峰值效应,高质量浓度(40和500μg/l)镍则对Ak呈现抑制效应,表明Ak与安氏伪镖水蚤响应重金属胁迫密切相关.本研究为揭示镍胁迫对桡足类的影响机制提供理论依据.To study the molecular response mechanisms of copepod to nickel stress,the full-length cDNA of arginine kinase gene from the copepod Pseudodiaptomus annandalei was cloned using the strategy of rapid amplification of cDNA ends(RACE).The arginine kinase cDNA(Pa-AK) was 1 180 bp in length including an open reading frame of 1 068 bp continuously encoded 355 amino acid residues,and the GenBank accession number was JX624126.The molecular weight of the predicted protein was 39.43 ku.Pa-AK showed high homology(77%-82%) with AKs from Macrobrachium rosenbergii,Neocaridina denticulate and Biomphalaria glabrata.Further,the transcripts of Pa-AK under nickel stress were examined using the real-time quantitative PCR technique.The results show that:low concentration of nickel(10 μg/L) first induced transcripts of Pa-AK,at 12 h,the transcript level started to decreased;high concentrations(40 and 500 μg/L) of nickel inhibited transcripts of Pa-AK.The results showed that Pa-AK is closely related to the response to heavy metal stress in Pseudodiaptomus annandalei.The successfully cloning of AK cDNA in Pseudodiaptomus annandalei and the study of expression characterization of Pa-AK under nickel stress will facilitate the study of the molecular mechanisms of nickel stress on copepod from proteomics aspects.国家自然科学基金项目(41276132
