PHOTOSENSITIZATION of SULFONATED ALUMINUM PHTHALOCYANINE (AIPCS) ON HUMAN HEPATOMA SMMC7721 CELL AND ITS MECHANISM
目的探讨铝酞箐光敏剂对人肝癌细胞SMMC┐7721的作用及其机制。方法以碘钨灯为光源,采用台盼兰拒染法观察铝酞箐对人肝癌细胞的光敏作用。结果随着铝酞箐浓度的增加,对细胞光敏作用增强;避光组对细胞无毒害作用。羟自由基(·OH)清除剂l┐组氨酸可明显抑制铝酞箐对癌细胞的光敏作用。结论铝酞箐是一种理想的治癌光敏剂,其光敏作用与·OH有关。Objective:To investigate the effect and mechanism of photosensitization of sulfonated aluminum phthalocyanine(AIPCS) on human hepatoma SMMC7721 cell.Methods:Using iodinetungsten lamp as a light source,the cytotoxicity of AIPCS on cancer cell was estimated by trypan blue exclusion method.Results:The role of photosensitized cytotoxicity increased with the concentration of AIPCS, while AIPCS alone without light has no any toxicity to hepatoma cells.The Lhistidine, one hydroxyl radical scavenger, could inhibit the phototsensitization of AIPCS obviously.Conclusion:AIPCS is an ideal photosensitizer for cancer therapy and the photocytotoxicity of AIPCS is due to hydroxyl radical
沙棘提取物对癌细胞DNA、蛋白质合成及体内血浆中的cAMP含量的影响
采用同位素示踪技术和蛋白质结合法研究了沙棘汁和沙棘油的抗癌机理,结果显示:沙棘汁对L7712白血病、S180肉瘤、腹水型肝癌等细胞的DNA、蛋白质合成均无抑制作用。沙棘油浓度在0.005mg/L~1.0mg/L时对L7712白血病细胞DNA合成有抑制作用,且此时抑制率与浓度呈负相关,沙棘汁ip注射剂量为1/5LD50时,沙棘汁对小鼠血浆cAMP含量略有提高
Antitumour activities of natural dinene compounds
采用MTT快速比色法,以长春碱(VInblASTInE,Vlb)为阳性对照,研究了36种从中草药中分离提取的天然二萜类化合物对三种不同癌细胞(肝癌SMMC-7721、肺腺癌l(342)、胃腺癌MCC80-3)生长的抑制活性。结果表明:6种二萜类化合物抗肿瘤活性强度与其化学结构密切相关;其中E(18)、E(21)和E(13)的抑瘤活性强度比长春碱强,顺序为:E(18)~E(21)>E(13)>Vlb>E(19)>V8>E(12);酯酰化取代基是影响其抗肿瘤活性强度的主要官能团。Using the MTT colorimetric assay, we studied acntitumour activities of 6 natural dinene compounds on three diFFerent tumour cell lines vs well-known antitumour drug, vinblastine (VLB).The results is that antitumour activities of the 6 dinene compounds are related with their chemical structures; the rank of their antitumour activities is; E18~E21 >E13>VLB>E19>V8>E12, acethyl moiety on the 6 dinene compounds is the Funcational group
抗氧化剂Isoverbascoside对MGC80-3细胞抗氧化酶活性和癌基因表达的影响
以苔酚蓝拒染法、核黄素-NBT还原法、DTNB还原法及RNA斑点杂交分析法分别对经不同浓度Isoverbascoside(Isov)处理的MGC80-3 细胞的生长速率、II期抗氧化酶(超氧化物歧化酶、谷胱甘肽过氧化物酶和过氧化氢酶)活性和N-ras、C-m yc及p53 基因表达作了检测.结果显示:经Isov处理后,细胞生长明显受抑;II期抗氧化酶活性较处理前明显上升;癌基因N-ras、C-m yc的表达较处理前下降,而抑癌基因p53 表达上升. 表明Isov 的抑癌作用与诱导II期抗氧化酶活性升高和癌基因表达改变有
Antitumour eFFects of phenylpropanoid glycosides
采用抗癌药筛选新方法-MTT法,研究了6种从中草药马先蒿(PEdICulArIS)中分离提取的苯丙素甙化合物(PHEnylPrOPAnOIdglyCOSIdES,PPg)对3种不同组织癌细胞生长的抑制活性。结果表明:6种PPg的抗肿瘤活性强度与其化学结构密切相关;它们的抑癌活性强度顺序为:ISOVErbASCOSIdE,VErbASCOSIdE,ECHInACOSIdE,PEdICulArIOSIdEA>CISTAnOSIdEd>PErMETHylVErbASCOSIdE;即分子中酚羟基越多,抗肿瘤活性愈强,当酚羟基完全被甲基化后,则对癌细胞生长无抑制作用。Using the MTT colorimetric assay,we studied antitumour eFFects of 6 phenyl-propanoid glycosides(PPG) From Pedicularis on three diFFerent tumour cell lines, The antitumoureFFects of 6 PPG were related to their chemical structure,Their antitumour eFFects ranked asisoverbascoside, verbascoside,echinacoside, pedicularioside A >cistanoside D>permethyl verbas-coside.It was shown that the more number of phenolic hydroxyl groups,the stronger PPG’s anti-tumour eFFects.The phenolic hydroxyl groups were methoxylated,and PPG couldn’t inhibit thegrowth of tumour cells.国家自然科学基金及福建省自然科学基

天然产物知母宁对氧自由基的清除作用李忌,李明(厦门大学抗癌研究中心,深圳999企业集团医药研究院)大量研究表明,氧自由基在炎症反应,自身免疫病,辐射损伤,心血管疾病及衰老,吞噬杀菌等生理过程中起极其重要的作用[1~3],并发现某些氧自由基清除剂具有抗...The scavenging eFFects of chinonin(chi),extracted From Anemarrhema asphodeloides,on oxygen Free radicals were studied.Chi was Found to signiFicantly scavenge the hydroxyl radical (OH) produced by Fenton reaction(Fe2+ +H2O2) (EC50=26 μmol/L) and inhibltory the rate of OH generation (IC50=10.5 μmol/L).the inhibitory the eFFects of Chi were signiFicantly more potent than those of mannitol, a speciFic.OH scavenger, with EC50 of 20.5mmol/L and IC50 of 8.6 mmol/L respectively.Chi also showed certain scavenging eFFect on superoxide anion (O) produced by NBT/PMS/NADH system.(IC50=45.6 μmol/L, which is as strong as 9U of SOD,a speciFic O scavenger).The results suggested that the anti-inFlammation,anti-radiation and anti-aging activities of Chi may be, at least partially, related to its scavenging eFFects on oxygen Free radicals.国家自然科学基
