    用高效液相色谱法测定一种中成药"小儿热速清"口服液中的绿原酸、黄芩苷和葛根素含量。采用Zorbax SB-C18色谱柱分离,以不同比例混合的甲醇和乙酸-水(1+99)溶液为流动相,进行梯度洗脱,在327nm波长处测定绿原酸、黄芩苷的含量,在250nm波长处测定葛根素的含量。绿原酸质量浓度在3.0~30.0mg.L-1范围内呈线性关系;黄芩苷质量浓度在2.7~27.0mg.L-1范围内呈线性关系;葛根素质量浓度在3.1~31.0mg.L-1范围内呈线性关系。方法的平均回收率为99.5%,99.8%,99.3%,相对标准偏差(n=6)分别为0.88%,0.84%,1.0%。The effective components,i.e.chlorogenic acid,baicalin and puerarin,in a TCM preparation,named "xiao-er resuqing" oral liquor,were determined by HPLC.The Zorbax SB-C18 chromatographic column was used as the stationary phase and mixtures of methanol and CH3COOH-H2O(1+99) solution with various proportions were used in the gradient elution.UV detections at 327 nm(for chlorogenic aicd and baicalin) and at 250 nm(for puerarin) were adopted in the determinations.Linear relationships between values of peak area and mass concentration of the 3 compounds were found in the following ranges of ① 3.0-30.0 mg·L-1,② 2.7-27.0 mg·L-1 and ③ 3.1-31.0 mg·L-1 respectively.Values of average recovery obtained were 99.5%(for chlorogenic acid),99.8%(for baicalin) and 99.3%(for puerarin).Values of RSD′s(n=6) found for the 3 compounds were 0.88%,0.84% and 1.0% respectively

    建立了1种同时测定右旋糖酐发酵液中蔗糖、果糖、葡萄糖等3种糖分含量的高效液相色谱定量分析方法。发酵液用流动相乙腈水溶液定容预处理,使其中的产物右旋糖酐析出以消除其对糖分测定的干扰。将预处理液离心,取上清液,在选定的色谱条件下:分离柱为OSMOSIL Sugar-D(4.6 mm I.D.×250 mm,0.46μm)柱,柱温为35℃,80%(V/V)乙腈水溶液为流动相,流速为1.0 m L/min,进样量为20μL,示差折光检测器,温度为40℃,实现了发酵液中蔗糖、果糖、葡萄糖的理想分离。在蔗糖质量浓度为0.1~15.0 g/L,果糖和葡萄糖质量浓度为0.5~25 g/L范围内,对糖分质量浓度与其峰面积进行线性回归分析,其相关系数在0.9994~0.9999之间,平均加标回收率为95.20%~104.00%,相对标准偏差在1.8%~3.2%之间。该方法可直接同时测定右旋糖酐发酵液中蔗糖、果糖、葡萄糖等3种糖分的质量浓度,为右旋糖酐发酵过程提供了一种操作简便、快速、可靠的监测方法。Abstract: A high performance liquid chromatographic method for simultaneous determination of sucrose, fructose and glucose in dextran fermentation broth was set up. The fermentation broth was pretreated by joining mobile phase of acetonitrile-water with metered volume to precipitating dextran for Eliminating its interference. The pretreated fermentation broth was eentrifugalized and take the its supernatant to determinate. The sucrose, fructose and glucose in dextran fermentation broth was well separated by the selected conditions, which were on an OSMOSIL Sugar-D(4.6 mm I.D.×250 mm, 0.46 μm)column,which was held at a constant temperature of 35 ℃, using a mobile phase composed of acetonitrile and water (80:20, V/V) at a flow rate of 1.0 mL/min, injection volume 20 μL, refractive index detector (RID) temperature was set as 40 ℃. The developed calibration curves displayed good linearity with the correlation coefficient were between 0.9994~0.9999 over a sucrose mass concentration range of 0.1~15.0 g/L, fructose and glucose mass concentration range of 0.5~25 g/L. Average spike recoveries for 5 replicate determinations of three saccharides were found in a range of 95.2%~104.0%, with a relative standard deviation (RSD) between 1.8%~3.2%. The results demonstrated that the mass concentration of sucrose, fructose and glucose in dextran fermentation broth can be simultaneously calculated.
