    Study on estrogenic effects of 17α-ethynylestradiol on juvenile Pagrosomus major

    采用静态暴露方式研究17α-乙炔基雌二醇(EE2)对真鲷(PAgrOSOMuS MAJOr)幼鱼的雌激素效应。当真鲷幼鱼暴露于0.01、0.1、0.5μg/lEE2,42 d后,真鲷幼鱼的肥满度极显著下降;血浆中卵黄蛋白原(VTg)被诱导产生,肝胰脏指数(HSI)和血浆蛋白总量极显著升高;血浆雌二醇(E2)和睾酮(T)水平显著降低,其中血浆雌二醇受EE2的干扰更显著,降低的幅度更大。结果提示,真鲷幼鱼的肥满度、肝胰脏指数、血浆蛋白总量、血浆性激素水平等可作为评估EE2等环境内分泌干扰物毒性效应的生物指标;真鲷幼鱼血浆中的VTg具有作为生物标志物,用于监测海洋水体中EE等环境内分泌干扰物的潜力。The estrogenic effects of 17α-ethynylestradiol(EE2) on the juvenile Pagrosomus major were studied via a static exposure experiment.After exposed to EE2 at doses of 0.01,0.1,0.5 μg/L for 42 d,the condition factors were significantly decreased,and plasma vitellogenins were induced,hepatopancreas somatic index(HSI) and total plasma protein were significantly increased.The levels of plasma estradiol(E2) and testosterone(T) were significantly decreased,and E2 were more affected.The results showed that the condition factor,HSI,total plasma protein and plasma sex hormone levels of juvenile Pagrosomus major may be used a biological index for evaluating the toxic effects of environmental endocrine disruptors such as EE2.The vitellogenin in the plasma of juvenile Pagrosomus major is potential to be a biomarker for monitoring the marine environmental endocrine disruptors such as EE2.国家自然科学基金(20277030

    Short-term dynamics of nutrients influenced by upwelling in a small oligotrophic coastal ecosystem, Gan Bay, in the northwest Philippines

    We present a time-series analysis of nutrient and pCO(2) (partial pressure of CO2) levels in an oligotrophic coastal ecosystem (Gan Bay), which was likely to be influenced by upwelled subsurface water. Gan Bay is off Currimao Harbor, in the northwest Philippines and is located at the boundary of the South China Sea (SCS). This 42-h time-series observation was conducted in December 2006. In addition to continuous observations of dissolved oxygen (DO) and pCO(2), discrete samples were collected at a depth of 5 m every 3 h for measurements of nutrients, including soluble reactive phosphorus (SRP) and inorganic nitrogen (NO3- + NO2-) in order to examine their dynamics and possible physical and biological controls. We observed remarkably large short-term variations in the surface water, spanning a 10-fold change for SRP (32-330 nM) and from <0.3 mu M to 4.3 mu M for (NO3- + NO2-. DO also varied substantially from a lower end of 171 to 205 mu M O-2. Surface water pCO(2) changed from an equilibrium stage with the atmosphere (similar to 386 mu atm) to a stage where it was a significant source for the atmospheric CO2 (seawater pCO(2) similar to 469 mu atm). We found that the variation of nutrients was driven neither by tidal mixing nor by biological activities, as was suggested by the variations in the total bacterial abundance and chlorophyll a. Instead, our inverse T-S relationship suggested a two end-member mixing process during the observation period. The N:P ratio throughout the observation period was similar to 13.2, which is characteristic of SCS subsurface and deep waters. Moreover, pCO(2) was correlated inversely with the sea surface temperature. It is likely, therefore, that an upwelled subsurface cold water with high nutrients, low-temperature and high-pCO(2) existed. It should be noted that this upwelled cold water did not appear to impact the entire observation period (approximately 35 h of 42 h), which might suggest an extremely dynamic nature for this upwelled cold water mass.     Analysis of contents and sources of pesticides in cultured shellfish of Fujian coastal areas

    对福建省15个缢蛏和牡蛎样品中的有机氯农药(六六六、DDTs)、PCBs、及有机磷农药残留进行了监测,并对其污染来源进行了探讨。结果表明,有机磷农药敌敌畏、甲胺磷和有机氯农药滴滴涕的检出率较高,敌敌畏和甲胺磷的平均含量分别为0.80和2.58×10-9(湿重),滴滴涕平均质量分数为8.84×10-9(湿重)。六六六和多氯联苯未检出。福建省内的闽江口和泉州湾的污染程度相对较高,但贝类体内污染物的含量均在食用卫生标准的控制下。The concentrations of organophosphorous pesticides, HCHs, DDTs and PCBs in 15 cultured shellfish samples (Ostrea gigas Thumberg and Sinonovacula constricta) collected from Fujian coastal areas were determined. The possible sources of these pollutants were discussed. The results showed that the shellfish samples were polluted mostly by organophosphorous pesticides, dichlorvos and methamidophos, as well as organochlorine pesticides DDTs. The average concentrations of dichlorvos and methamidophos in the shellfish were 0.80 and 2.58×10~(-9)(ww), respectively, while DDTs were 8.84×10~(-9) (ww). HCHs and PCBs were not detected. The water column in the Minjiang Estuary and Quanzhou Bay were more polluted than other coastal areas in Fujian. However, the pollutants in the cultured shellfish were still controled

    目的通过对厦门市0~6岁儿童尿汞水平调查,了解厦门市儿童汞暴露水平。方法根据行政区划以及儿童人数分布从全市选取代表性的8个社区,进行分层随机抽样,共收集到1076例0~6岁儿童尿样。样本采用随意尿,每个儿童统一使用聚乙烯塑料瓶收集不少于5 M l尿样,尿样统一采用dMA-80自动测汞仪测定。结果儿童尿汞测定值呈偏态分布,因此使用几何平均数表示儿童尿汞的平均水平。男女儿童尿汞几何平均值分别为0.77μg/l和0.74μg/l,经非参数T检验比较,差异无统计学意义(P>0.05)。0~1岁、2~3岁和3~6岁3个年龄段的儿童尿汞几何平均值分别为0.74μg/l、0.75μg/l和0.78μg/l,经非参数T检验比较,差异无统计学意义(P>0.05)。对1076名儿童尿汞值分析得出尿汞的几何平均值为0.76μg/l,95%置信区间0.70~0.81μg/l。结论厦门市0~6岁儿童的尿汞水平的调查结果可以为国内沿海城市0~6岁儿童汞暴露水平的本底值研究提供参考。Objective To understand the mercury exposure level in 0-6 year-old children of Xiamen city through the survey.Methods A total of 1 076 healthy infants and young children aged 0-6 year-old were selected from 8 communities with stratified and cluster sampling method respectively.The spot urine samples of the children were collected( more than 5m L per child).The concentration of mercury was determined by DMA-80 automatic mercy instrument.Results The urine mercury values of the healthy children were in skewed distribution and the results were expressed in geometric mean.The urine mercury values for boys and girls are0.77μg / L and 0.74μg / L,respectively.There was no significant difference for the urine mercury values between boys and girls( P >0.05).Meanwhile,the urine mercury values for groups of 0-1 year-old,2-3 year-old,and 4-6 year-old were 0.74μg / L,0.75μg / L and 0.78μg / L,respectively.The urine mercury values among the three age groups had no significantly difference neither( P > 0.05).The upper 95 th percentile in 1076 urine samples was 0.85 μg / L,95% confidence interval was 0.70-0.81μg / L.Conclusion The average value of 0-6 year-old children's urine mercury level in Xiamen was close to those in the reports of two researches in Shenyang and Germany.Therefore,results will be helpful for the understanding of mercury exposure in 0-6 year-old children of Xiamen city

    采用腹腔注射17β-雌二醇(E2),使真鲷雌鱼在一星期内产生大量卵黄蛋白原(Vtg);利用阴离子交换高效液相色谱系统分离纯化E2诱导后的Vtg.Native-PAGE电泳结果表明,真鲷有两种形式的Vtg,分子量分别为570 kDa和360 kDa;SDS-PAGE电泳结果表明,它们由两个不同的亚基构成,分子量分别为180 kDa和115 kDa;磷、脂、糖蛋白分析证实,所获取的Vtg是一类含磷、脂、糖的大分子蛋白.同时考察了4-壬基酚(4-NP)对真鲷Vtg的诱导作用.Vitellogenin(Vtg) was largely induced in female Pagrosomus major in a short time by only one intraperitoneal injection of 17β-estradiol(E_2).With anion exchange high performance liquid chromatography(HPLC),the Vtg was effectively purified from the plasma of female Pagrosomus major injected with E_2.The results of Native-PAGE showed that the Vtg of Parosomus major had two forms,with molecular weights around 570 kDa and 360 kDa,respectively.With SDS-PAGE,the Vtg was reduced into two different monomers,with molecular weights approximately 180 kDa and 115 kDa,respectively.The analyses of phosphorus,lipid and carbohydrate showed the Vtg a phospholipoglycoprotein.The further study indicated that the Vtg in Pagrosomus major could also be induced by 4nonylphenol,this shows that 4-nonylphenol is a kind of environmental estrogens.国家自然科学基金(20277030)资

    利用弱阴离子交换色谱,从雌激素17β-雌二醇诱导的真鲷鱼血清中分离纯化出卵黄蛋白原粗组分,以超滤法进一步除去杂质蛋白,并脱盐浓缩,获得纯度较高的卵黄蛋白原(Vtg),两级纯化得到的Vtg经常规聚丙烯酰胺凝胶电泳(Native-PAGE)分析,证实真鲷鱼Vtg有三种形式,分子量分别为665kDa,635kDa和355kDa;变性聚丙烯酰胺凝胶电泳(SDS-PAGE)分析结果表明,它们由185kDa左右的蛋白亚基组成.With ultrafiltration and anion exchange liquid chromatography,a two-step method had been developed for purification of vitellogenin(Vtg) from the serum of Pagromus major induced by 17β-estradiol(E_(2)).The(results) showed that most impurities could be removed during the first step by weak anion exchange liquid(chromatography) and further clean-up through ultrafiltration.In Native-PAGE,the Vtg purified by the two steps showed three forms,with molecular weights approximately 665kDa,635kDa and 355kDa,respectively.The result of SDS-PAGE indicated that the molecular weight of Vtg monomers was about 185kDa.High resolution anion exchange high performance liquid chromatography was applied to analyze the Vtg purified from the two-step method,and approved the high purity.The purified Vtg could be used for further analysis.国家自然科学基金(20277030)资助项

    目的分析厦门市2010-2012年间1月龄(30±7)d婴儿的营养与健康状况及存在的健康问题,为进一步做好儿童保健工作提供科学依据。方法按照出生日期抽取厦门市2010-2012年3 241名1月龄婴儿的体检数据资料,采用纵向比较法分析统计。结果厦门地区2010-2012年1月龄婴儿中男婴超重、低体重、肥胖、消瘦、生长迟缓、发育迟缓率分别为20.3%、1.49%、0.74%、1.65%、2.72%和0.17%,而女婴则为16.14%、2.32%、0.74%、2.78%、2.32%、0.46%。各年度男女婴身长体重、bMI及喂养方式差异无统计学意义(P均>0.05)。结论厦门市2010-2012年1月龄男女婴营养与健康状况之间差异无统计学意义,但整体存在营养不良与营养过剩共存的问题,尤其是女婴消瘦的问题,应引起高度重视。Objective To analyse the status of 1month infants' nutrition and health during 2010-2012in Xiamen.Method The physical examination data of 3 241one month old infants in Xiamen during 2010-2012were used horizontal analysis.Results The rates of overweight,underweight,obesity,marasmus,growth retardation and developmental retardation of 1month boy infants were 20.3%,1.49%,0.74%,1.65%,2.72% and 0.17%,while the girls' were 16.14%, 2.32%,0.74%,2.78%,2.32%,and 0.46%in Xiamen during 2010-2012.Boys' and girls' length,weight and BMI and feeding status were the same among three years.Conclusions There are no significant differences between boys and girls 1 month infants from Xiamen in the nutritional and health status.And some problems,such as mal-nutrition and overweight for 1month infants,especially marasmus for girls,should be paid more attention in Xiamen.世界卫生组织项目(2011/191434-0;CHN-11-MCN-005192)~
