    Functions and Mechanisms of the p38 MAP Kinase Pathway

One of the four sub-families, the p38 group MAP kinases, plays a vital role in numerous biological processes. The p38 signaling pathway can be activated in response to many physical, chemical,and biological stresses, including UV light, heat shock, osmotic shock, inflammatory cytokines, and growth factors.The p38 pathway also regulates various physiological processes such as cell differentiation, the cell cycle, and inflammation. This review focuses on the characteristics of each p38 member, the components of the p38 pathway, and the mechanisms and consequences of p38 activation. We also discuss the interplay between the p38 pathway and other signaling pathways

    水作为所有细胞和组织的重要组成部分,其pH和诸多生理活动密切相关.荧光分析具有可视化、高分辨率等特点,在生命体系的pH分析中得到广泛应用.近年来; , pH触发荧光成像技术在生物医学领域逐渐得到应用.本文简要介绍了一类酸度触发荧光、含分子内螺环的罗丹明衍生物的设计与功能化,及其生物成像应用.pH is critical for diverse physiological activities. Disruption of pH; homeostasis is pertinent to myriad pathological events. Particularly,; the organelle-specific pH within mammalian cells is elaborately; controlled and yet succumbs to pathological stress. Lysosomes are the; major intracellular acidic compartments, and the lysosomal acidity is; critical for diverse biological events. Aberrant lysosomal pH is; manifested a broad spectrum of diseases ranging from inflammation to; cancer metastasis, and to lysosomal storage diseases. As such, defining; pH changes in specific pathological settings would be of use for; biomedical investigations. Rhodamine-lactams, a group of nonfluorescent; rhodamine derivatives featuring intramolecular spirolactam, are poised; to proton triggered opening of the lactams to give fluorescent; rhodamines, and has been widely used for lysosomal imaging in cell; models.     地球周围的大气环境特性,随高度的上升,显著变化。对于本文关心的高度100km附近高超声速飞行器气动特性而言,稀薄大气密度涨落影响是一个重要的问题。按照大气层划分,高度100 km属于热层。热层的下界高度约86 km,上界高度与太阳活动有关,在太阳宁静期约为200 km,太阳活动期约500 km。热层下接中间层(高度50~86 km),上连电离层(高度80~1000 km),大气研究已知,中间层主要受重力波和湍流影响,电离层主要受太阳活动和地磁效应影响。高度100 km附近空域,处在热层下界,重力波、湍流、太阳活动和地磁效应等均有影响,理论上需要Navier-Stokes方程和Maxwell方程..

    Fault Localization Method Based on Program Execution Trace

    当程序在测试中发生错误时,将形成一条错误的程序执行路径,程序员将会花费很多精力去检测程序代码和定位最终的程序错误。提出一种基于单条程序执行路径的错误定位方法,该方法通过对程序进行反向执行,计算出多个最弱前置条件及其相对应的疑似错误语句集,并生成错误定位树,来辅助程序员进行快速错误定位。对西门子测试数据集进行的实验表明了该方法具有良好的效果。 After a failed test is encountered and the error trace is generated, a significant amount of effort is often required for programmers to manually examine the program code and localize the failure’s root cause. In this paper, we propose a fault localization method based on program execution trace. This method computes a set of possible error statements based on a combination of weakest pre-condition computation through program’s reverse execution. All possible error statements are organized in a fault localization tree to help developer identify the root cause. Our experiments on SIR datasets demonstrate that our method can not only efficiently compute the possible causes, but also provide sufficient information to help programmers quickly locate the root cause

    通信作者:[email protected]细胞连接蛋白是由多基因家族编码的一类结构相似、分子质量不同的蛋白质.在细胞膜上,每6个相同或不同的连接蛋白围绕中央孔排列形成一个连接子,相邻细胞膜上的连接子相互对接形成细胞间隙连接,细胞间隙连接是一种重要的通讯连接,它不仅是细胞间代谢偶联、冲动传导的结构基础,而且可以通过介导与细胞的迁移、分化、增生和器官形成有关的信号物质而在胚胎发育中起重要作用.为了进一步探讨细胞间隙连接在生物体中的作用,采用斑马鱼胚胎显微注射核酸酶I-SceI和质粒DNA的方法成功构建了绿色荧光蛋白GFP与细胞连接蛋白Connexin48.5融合表达的转基因荧光斑马鱼品系,并通过对荧光蛋白发光的观察在斑马鱼鱼体中进行连接蛋白Connexin48.5的定位.实验结果表明:核酸酶I-SceI介导下的斑马鱼转基因方法效率较高,可行性好;连接蛋白Connexin48.5在斑马鱼体内主要定位于眼球晶状体和脊索.所获得的Connexin48.5-GFP融合表达斑马鱼系,以及利用该品系进行的Connexin48.5定位研究对细胞连接蛋白在生物体内的功能研究将具有重要作用.[英文文摘]Connexins are a group of proteins coded by multi gene family.On the cell membrane,every 6 Connexins combine together to form a Connexon,and two Connexons of two different cells connect with each other to form a gap junction.Gap junction is a very important kind of communicctional connection between cells,which is not only the constructional basic of metabolic coupling and signal transduction betweens cells,but also the channeel of signal substance controlling cell migration,differentiation,proliferation and organogenesis.For a deeper study of gap junction function in organisms,we generated a transgenic zebrafish line that expresses Connexin 48.5-GFP fusion protein by co-injecting meganuclease I-Scel and plasmid DNA into zebrafish embryos.I-Scel mediate transgenesis showed high efficiency,which made it a better way to get transgenic animals than the traditional method.While the working mechanism of I-Scel remains unclear,in our opinion,I-Scel binds to its recognition site after the cleavage and promotes unclear localization of the transgene could be a good explanation.Using our transgenic zebrafish line,we found that Connexin 48.5 is highly expressed in lens and notochord in zebrafish.In addition to its expression in lens,which has been reported preciously,our new finding suggested that Connexin48.5 could have important roles  in notochord development or nervous system function.国家重点基础研究发展计划(973)项目(2009CB22200

    Circulating Specific IgG Immune Complexes Activating Complement in Pregnancy Induced Hypertension

    目的探讨免疫球蛋白 g 特异性激活补体类循环免疫复合物(Igg/C3-CIC)在妊高征(PIH)发病中的作用。方法采用捕捉法 ElISA 测定血清 Igg/C3-CIC 水平,单向免疫扩散法测定血清 Igg 水平,正常晚期妊娠34例(对照组),妊高征81例(研究组)。结果正常足月孕妇、轻度PIH、中度 PIH 及重度 PIH 孕妇血清 Igg/C3-CIC 浓度分别为0.284±0.18、0.303±0.16、0.483±0.286及0.462±0.206,血清 Igg 浓度分别为191.07±112.92 g/l、147.56±47.91 g/l、140.18±32.89 g/l 及125.4O±53.24 g/l。中度 PIH 和重度 PIH 孕妇血清 Igg/C3-CIC 浓度高于正常足月孕妇和轻度 PIH 者(P<0.01);不同程度 PIH 孕妇血清 Igg 浓度均高于正常足月孕妇者(P<0.05)。结论妊高征患者血清 Igg/C3-CIC 浓度升高及 Igg 浓度降低。Objective To determine circulating specific IgG immune complexes activating complement(IgG/C3-CIC)concentrations in maternal serum of normal term pregnant women and women with pregnancy induced hypertension(PIH).Methods We detect the serum level of IgG/ C3-CIC and IgG in 115 subjects,including 34 cases of normal pregnant women and 81 cases of PIH women.Results The serums concentrations of IgG/C3-CIC in normal term pregnant women, women with mild PHI,moderate PIH and serve PIH were 0.284±0.18,0.303±0.16,0.483±0.286 and 0.462±0.206,respectively.The serum concentration of IgG in the women were 191.07±112.92g/L,147.56±47.91 g/L,140.18±32.89 g/L and 125.40±53.24 g/L,respectively.The serum concentration of IgG/C3-CIC in patients with moderate PIH and severe PIH was higher than that in patients with mild PIH and normal term pregnant women(P<0.01).The serum concentration of IgG in patients with PIH was lower than that in normal term pregnant women,P<0.05.Conch- sions The serum concentration of IgG/C3-CIC increased and the serum concentration of IgG de- creased in patients with PIH
