Research on the Dimerization Sequence of Exo70 and Its Role in Breast Cancer Cell Metastasis
Exo70是胞外分泌复合体Exocyst的关键亚基,可通过参与Exocyst复合体或自身聚合促进细胞迁移。我们在研究中发现,Exo70自身形成二聚体的能力显著强于与Exocyst其它7个亚基的结合,提示Exo70二聚化的重要作用。进一步构建Exo70缺失突变体,并通过一系列免疫共沉淀实验确定了Exo70形成二聚体的关键位置是位于氨基酸31~35与505~509的两段同样的序列SLEKS。进而利用这两个序列的缺失突变体,通过transwell迁移实验,证实这两段SLEKS序列在Exo70二聚化及乳腺癌细胞迁移过程中的关键作用。以上结果为Exo70独立于Exocyst复合体功能之外的新功能研究提供了依据,也为乳腺癌及其转移的治疗提供潜在的靶点。Exo70,the key subunit of Exocyst protein complex,promotes cell metastasisby self-dimerization or by involving in the Exocyst complex formation.In this research,we found that Exo70 was dramatically more capable of forming self-dimerization than binding to the other subunits of exocyst complex,and aindicative of the important roles of its self-dimerization.Then we constructed a set of truncation mutants of Exo70 in order to determine the core sequence for its dimerization.With co-immun oprecipitation analysis,we identified two identical protein sequences"SLEKS"located at amino acid 31~35 and 505~509 as critical sequencesfor the dimerization of Exo70.By "cell transwell"essay,we further found that the full length Exo70 significantly promotes breast cancer metastasis,while the two truncation mutants of Exo70(Δ31~35 and Δ505~509) don't process such ability,indicating that these two "SLEKS"sequences play pivotal role in breast cancer cell metastasis and invasion.Together,these results provide fundamental basis for researches on novel functions of Exo70 besides its well-known role in Exocyst complex formation and provide potential drug target for treating breast cancer and its metastasis.国家自然科学基金青年项目(31300970);; 福州大学科研启动项目(510402)共同资
Assisted Selecting Parents of Hybrid Combination Based on SSR Polymorphism
Corresponding author, zjhym@ public.xm.f j.cn[中文文摘]本研究利用 6 0对SSR引物对籼稻品种佳辐占与 6个候选亲本品种进行相互之间的多态性分析。结果表明 :佳辐占与明恢 6 3之间的多态性程度最高 ,达到 2 2 7% ;其次是与明恢 86之间的多态性程度 ,达 19 6 % ,而与其他品种的多态性程度较低。多态性程度高的品种间的杂交组合 ,其杂种优势明显高于多态性程度低的品种间的组合。初步表明 ,根据SSR多态性程度的高低选配亲本 ,其配制杂交组合可以获得较高的杂种优势。[英文文摘]In this research, the authors attempt to select the parents for hybrid combinat ion by using SSR polymorphism. Indica rices, Jiafuzhan and other six candidate parents, were employed to be detected and analyzed DNA polymorphism by using sixty pairs of SSR primers. It showed that the degree of polymorphism between Jianfuzhan and Ming hui 63 was highest , it reaches 22.7%, and of that between Jianfuzhan and Minghui 86 ranks the second, it reaches 19.6% . it is lower of the degree of polymorphism between Jiafuzhan and other 4 variet ies. The heterosis of the combinat ion w as according to the degree of polymorphism between parents, the higher polymorphic degree betw een parent s the hig her heterosis the combinat ions. was feasible to select the parents of hybrid combinat ions based on SSR polymorphism for evaluat ing the heterosis of the hybrid combinations
Lithium battery internal resistance detection based on transition process analysis
以常用的锂电池模型为研究对象,通过对电池放电过程瞬间电流阻断产生的过渡过程的分析,提出了一种锂电池模型参数估算方法,进而实现对锂电池内阻的在线实时辨识。通过实验,获得了电池内阻在不同工况下的变化趋势。Based on used lithium battery model, by the analysis of transition process produced by current blocking-up in the process of battery discharge, a lithium battery model parameter estimation method was put forward.Online real-time identification of lithium battery internal resistance was realized.Through experiments, the trend of the internal resistance of the battery in different operating conditions was obtained.厦门市科技计划项目(3502Z20123013

Flora analysis of seed plants in Wuyuan forest birds nature reserve
在野外调查及查阅相关资料的基础上,对婺源森林鸟类自然保护区的种子植物资源进行区系分析。结果表明:(1)区内植物种类丰富多样,共有种子植物1 724种,隶属于194科732属。其中裸子植物8科13属16种,被子植物186科719属1 708种。(2)区内种子植物科属的组成较为分散,植物成分较为复杂。在科的组成结构上,少种科的数量最多,占54.64%。但包含植物种类最多的是较大科,占总种类的29.23%。在属的组成结构上,则主要以少种属和单种属为主,分别占比32.79%和55.87%。(3)区内种子植物区系成分复杂。热带分布类成分相对占优势,有297属,占总属数的40.57%。其次是亚洲分布类有196属,占26.78%。此外温带分布类含173属,占23.63%;世界分布类有66属,占9.02%。(4)将婺源种子植物与邻近的四个地区进行比较。5个地区中世界分布类和亚洲分布类所占比值大致相当,但热带类和温带类所占比值则有较大差异。热带分布类成分最多的婺源,占比值为40.57%,最少的黄山只有31.82%。温带分布类成分最多的黄山,占比值为29.31%,最少的婺源只有23.63%。The area types analysis of the seed plant was done based on field investigation and relevant information about Wuyuan forest bird nature reserve. The results showed that:(1) There are 1 724 species of seed plants in this region, belong to 194 families and 732 genera, including 8 families,13 genera, 16 species of gymnosperm and 186 families, 719 genera, 1 708 species of angiosperms.(2) The composition of the seed plants in this region is highly dispersed and very complex. In the composition of the families, the number of small families is the most, which rate is 54.64%. However, the major families contain the largest number of plant species, the rate is 29.23%. In the composition of the genus, species focus on the small genera and the single genus that accounted for 32.79% and 55.87%.(3) The flora of seed plants in this region is complex. There are 297 genera, 40.57% genera of the total, in the tropical distribution. Followed by the Asian distribution of 196 genera, accounting for 26.78%. In addition, the temperate distribution including 173 genera, 23.63% of the total and the world distribution including 66 genera, accounting for 9.02% of the total.(4) Comparison of other 4 regions in the same latitude of Wuyuan forest birds nature reserve. In the 5 regions, the proportion of the world distribution and the Asia distribution is roughly equal, but there is a large difference between the tropical distribution and temperate distribution. Wuyuan, which contain the highest ratio of tropical distribution, 40.57% of the total, and in the least Huangshan the ratio is only 31.82%. Temperate distribution has the highest ratio 29.31% of Huangshan, and the least 23.63% of Wuyuan.国家科技部科技基础性工作专项(2015FY110200
中国物理海洋学研究70年:发展历程、学术成就概览
本文概略评述新中国成立70年来物理海洋学各分支研究领域的发展历程和若干学术成就。中国物理海洋学研究起步于海浪、潮汐、近海环流与水团,以及以风暴潮为主的海洋气象灾害的研究。随着国力的增强,研究领域不断拓展,涌现了大量具有广泛影响力的研究成果,其中包括:提出了被国际广泛采用的"普遍风浪谱"和"涌浪谱",发展了第三代海浪数值模式;提出了"准调和分析方法"和"潮汐潮流永久预报"等潮汐潮流的分析和预报方法;发现并命名了"棉兰老潜流",揭示了东海黑潮的多核结构及其多尺度变异机理等,系统描述了太平洋西边界流系;提出了印度尼西亚贯穿流的南海分支(或称南海贯穿流);不断完善了中国近海陆架环流系统,在南海环流、黑潮及其分支、台湾暖流、闽浙沿岸流、黄海冷水团环流、黄海暖流、渤海环流,以及陆架波方面均取得了深刻的认识;从大气桥和海洋桥两个方面对太平洋–印度洋–大西洋洋际相互作用进行了系统的总结;发展了浅海水团的研究方法,基本摸清了中国近海水团的分布和消长特征与机制,在大洋和极地水团分布及运动研究方面也做出了重要贡献;阐明了南海中尺度涡的宏观特征和生成机制,揭示了中尺度涡的三维结构,定量评估了其全球物质与能量输运能力;基本摸清了中国近海海洋锋的空间分布和季节变化特征,提出了地形、正压不稳定和斜压不稳定等锋面动力学机制;构建了"南海内波潜标观测网",实现了对内波生成–演变–消亡全过程机理的系统认识;发展了湍流的剪切不稳定理论,提出了海流"边缘不稳定"的概念,开发了海洋湍流模式,提出了湍流混合参数化的新方法等;在海洋内部混合机制和能量来源方面取得了新的认识,并阐述了混合对海洋深层环流、营养物质输运等过程的影响;研发了全球浪–潮–流耦合模式,推出一系列海洋与气候模式;发展了可同化主要海洋观测数据的海洋数据同化系统和用于ENSO预报的耦合同化系统;建立了达到国际水准的非地转(水槽/水池)和地转(旋转平台)物理模型实验平台;发展了ENSO预报的误差分析方法,建立了海洋和气候系统年代际变化的理论体系,揭示了中深层海洋对全球气候变化的响应;初步建成了中国近海海洋观测网;持续开展南北极调查研究;建立了台风、风暴潮、巨浪和海啸的业务化预报系统,为中国气象减灾提供保障;突破了国外的海洋技术封锁,研发了万米水深的深水水听器和海洋光学特性系列测量仪器;建立了溢油、危险化学品漂移扩散等预测模型,为伴随海洋资源开发所带来的风险事故的应急处理和预警预报提供科学支撑。文中引用的大量学术成果文献(每位第一作者优选不超过3篇)显示,经过70年的发展,中国物理海洋学研究培养了一支实力雄厚的科研队伍,这是最宝贵的成果。这支队伍必将成为中国物理海洋学研究攀登新高峰的主力军
