Influence of solar activity upon light curves of comets P/Halley (1986 3) and P/Churyumov-Gerasimenko (1982 8)
It is shown that the comet P/Halley's (1986 3) total magnitudes correlate with changes in the solar activity indices and the solar wind velocity. A statistically reliable correlation between the outbursts of brightness and magnitude variations of the short-period comet Churyumov-Gerasimenko (1982 8) and the level of the solar activity has been discovered
A clinical case of idiopathic pulmonary arterial hypertension in patient with comorbid ischemic heart disease: the key stages of diagnosis
Despite the progress achieved in the development of new directions in the diagnosis and treatment of idiopathic pulmonary arterial hypertension, it remains one of the most severe and prognostically unfavorable diseases. The article presents a clinical case and an algorithm for diagnosis of this rare disease, in respect to the decisionmaking in clinical practice. The present case underscores the need to consider a broad differential diagnosis for marked dyspnea in mature patients, especially when the intensity of dyspnea is out of proportion to the severity of underlying heart or lung diseases, or when symptoms fail to subside as expected in response to conventional therapy. Mechanics and Mathematical Modeling of Class III Treatment with Orthodontic Appliances with a Movable Ramp
Treatment of class III is a current problem in orthodontics that requires constant improvement of its methods, development of new or modifications of known methods and techniques. We have developed and studied the modification of removable functionally-directing orthodontic appliances for treatment of Class III, which consists of a plastic base, vestibular arc, retaining clasps, ramp, which is connected with the base by means of two torsion springs. Its usage ensures a prolonged contact of ramp with the teeth. We studied two types of club-shaped springs (torsion springs): one spring, which create an amortization effect during the action of the ramp, but do not change its inclination angle and second one – spring that seek to increase the angle of the ramp inclination due to the disclosure of its curl
Novel antibodies against RCD-8 as a tool to study processing bodies
Aim. To develop the model system for processing bodies (PBs) state monitoring and accomplish it in the future as a possible read-out of mTOR activity in mammalian cells. Methods. In course of this study we raised polyclonal antibodies against one of the PBs scaffold proteins – RCD-8 and employed cell imaging technique. Results. It has been shown that the obtained antibodies recognize the intracellular structures, namely PBs. The detected protein co-localized with known marker of PBs – DCP1a, and partly with marker of SGs – CPEB. Conclusions. Based on changes of PBs number and size in cells after exposure to known inductors or inhibitors of PB formation we prove the specificity of generated antibodies and possibility of their application for studies on the processing bodies dynamics controlled by mTOR-dependent signalin
Development of monoclonal antibodies specific to ribosomal protein S6 kinase 2
Ribosomal protein S6 kinase 2 (S6K2) is a serine/threonine kinase that belongs to the family of AGC kinases, which includes PKB/Akt, PKC, PDK1, and SGK1. Mammalian cells express two isoforms of S6K, termed S6K1 and S6K2. Each of these has nuclear and cytoplasmic spicing variants, which originate from different initiation start codons. Nuclear isoforms of S6K1 and S6K2 are slightly longer, as they possess additional sequences at the N-terminus with nuclear localization signals. Biochemical and genetic studies implicated S6Ks in the regulation of cell size, growth, and energy metabolism. Deregulation of S6K signaling has been linked to various human pathologies, making them excellent targets for drug discovery. The aim of this study was to produce monoclonal antibodies directed at the N-terminal regulatory region of S6K2, which shows very low homology to S6K1 or other members of the AGC family. To achieve this goal, two S6K2 fragments covering 1-64aa and 14-64aa N-terminal sequences were expressed in bacteria as GST/6His fusion proteins. Affinity purified recombinant proteins were used as antigens for immunization, hybridoma screening, and analysis of generated clones. We produced a panel of S6K2-specific antibodies, which recognized recombinant S6K2 proteins in ELISA and Western blot analysis. Further analysis of selected clones revealed that three clones, termed B1, B2, and B4, specifically recognized not only recombinant, but also endogenous S6K2 in Western blot analysis of HEK293 cell lysates. Specificity of B2 clone has been confirmed in additional commonly used immunoassays, including immunoprecipitation and immunocytochemistry. These properties make B2 MAb particularly valuable for elucidating signal transduction pathways involving S6K2 signaling under physiological conditions and in human pathologies
