Probiotics and prebiotics in food and other products

V teoretické části práce se pojednává o současném stavu a výzkumu probiotik a prebiotik, jejich využitelnosti pro modulaci mikroflóry hostitele a jejich příznivých účincích na zdraví jedince. Dále je pojednáno o problematice účinnosti probiotických kmenů, která závisí na použité potravinové matrici a na řadě dalších faktorů. Experimentální je zaměřena na identifikaci vybraného bakteriálního kmene obsaženého v probiotickém výrobku. Bakteriální DNA byla z výrobku izolována metodou fenolové extrakce a pomocí magnetických nosičů s následnou analýzou získané DNA metodou polymerázové řetězové reakce (PCR).Theoretical part of this thesis focuses on present state and research of probiotics and prebiotics, their use for microflora modulation of host and their beneficial effects on the health of individuals. Furthermore thesis deals with efficiency of probiotics strains, which depends on the food matrix and other various factors. The experimental part focuses on the identification of chosen bacterial strain, which is contained in probiotics product. It is realized due the isolation of bacterial DNA by phenol extraction and by use of magnetic particles and subsequent analysis of obtained DNA by polymerase chain reaction (PCR).

Identification of microorganisms in cosmetic products with probiotics

Probiotické výrobky jsou nedílnou součástí současného trhu. Mezi výrobky obsahující přidané probiotické kultury se řadí také kosmetické výrobky. První část práce se zabývá testy na schopnost přežití bakterií v prostředí prezervačních látek, které jsou v kosmetických výrobcích obsaženy. Pro tyto testy byly použity sbírkové kmeny bakterií rodu Lactobacillus. Další část práce se zabývá izolací bakteriální DNA z probiotických kosmetických výrobků Ryor, Yoghurt of Bulgaria, FeminaMed a Lactovit Activit, a to v kvalitě vhodné pro amplifikaci v PCR. DNA byla izolována metodou fenolové extrakce a magnetickými nosiči. Přítomnost bakterií byla prokázána rodově a druhově specifickými PCR Lactobacillus. Byla provedena optimalizace druhově specifické PCR pro identifikaci Lactobacillus pentosus. Druhová identifikace byla v souladu s údaji deklarovanými výrobci.Probiotics products are an integral part of the current market. Products containing probiotics cultures are also cosmetic products. The first part of the study focuses on testing of bacterial survival abilities in the environment of preservatives presented in cosmetic products. Collection strains of genus Lactobacillus were used for these tests. Another part of the study focuses on isolation of bacterial DNA from probiotic cosmetic products Ryor, Yoghurt of Bulgaria, FeminaMed and Lactovit Activit in PCR-ready quality. DNA was isolated by fenol extraction and with magnetic particles. Presence of bacteria was proved by genus and species specific PCRs Lactobacillus. Species specific PCR for identification of Lactobacillus pentosus was optimalized. Species identification was in accord with data declared by producers.

Risk Factors and Detection of Lameness Using Infrared Thermography in Dairy Cows - A Review

Lameness in dairy cows is a worldwide problem, usually a consequence of hoof diseases. Hoof problems have a negative impact on animal health and welfare as well as the economy of the farm. Prevention and early diagnosis of lameness should prevent the development of the disease and consequent high costs of animal treatment. In this review, the most common causes of both infectious and noninfectious lesions are described. Susceptibility to lesions is primarily influenced by the quality of the horn. The quality of the horn is influenced by internal and external conditions such as hygiene, nutrition, hormonal changes during calving and lactation, the animal's age or genetic predisposition. The next part of this review summarizes the basic principles and possibilities of using infrared thermography in the early detection of lameness in dairy cows.O

Significant aspects of the national mergers in the Czech Republic

The paper deals with issues of national mergers carried out in the year 2015 and 2016. The development of the mergers is characterized by the fact that it is a multidisciplinary areas (tax, accounting and legal), in practice the most widespread method is Mergers through consolidation. The analysis is therefore focused just on this type of merger. It examines the development of mergers carried out in the Czech Republic as well as globally. In the global perspective,is processed the number of mergers and acquisitions, and the trends in company transformations are monitored. For the Czech Republic is analyzed the number of mergers, number of merging companies, including their legal form, the number of registered mergers by individual Regional Courts, the date of entries, determination of the appointed date of merger and date of completion of merger project. Consequently are presented features typical of the implementation of national mergers. The paper also examines the relationship between the number of mergers and GDP and PX index from 2002 to 2016.O

Possibilities of using plasticizers and superplasticizers in alkali-activated systems

Bakalářská práce se zabývá přípravou a vlastnostmi alkalicky aktivované vysokopecní strusky po přidání různých druhů plastifikátorů a superplastifikátorů v různém procentuelním zastoupení. Cílem této práce je nalézt vhodný plastifikátor nebo superplastifikátor, který by zlepšil jak zpracovatelnost, tak mechanické vlastnosti alkalicky aktivované strusky. Dále byl posouzen vliv těchto příměsí na pevnosti v tlaku a pevnosti v tahu za ohybu. Na závěr se pomocí infračervené spektrometrie studuje chování plastifikačních přísad v alkalickém prostředí.Bachelor thesis deals with the preparation and properties alkali activated blast furnace slag after addition of the different types of plasticizers and superplasticizers with the different weight percentage amount. The aim of this thesis is to find suitable plasticizers or superplasticizers which would improve both workability and mechanical properties of alkali activated blast furnace slag. The effect of these admixtures on compressive and flexural strength was determined. In conclusion the behaviour of plasticizing admixtures in alkaline environment through the infrared spectrometry is studied.

Lokální steroidogeneze v periferních tkáních a její regulace

Vrozené i adaptivní imunitní reakce jsou modulovány hormonálně, mimo jiné pomocí glukokortikoidů, a působením mikrobiomu, nicméně přesný mechanismus není zcela objasněn. Tato studie se zabývá vlivem mikrobiomu na de novo biogenezi a lokální regeneraci glukokortikoidů. Konkrétním předmětem analýzy je vliv komenzálů na expresi genů kódujících steroidogenní enzymy (Star, Cyp11a1, Hsd3b1, Cyp21a1, Cyp11b1) a enzym zajišťující regeneraci glukokortikoidů (Hsd11b1) v nadledvinách, tračníku, slezině a mesenteriálních lymfatických uzlinách. Studie byla provedena na konvenčních a bezmikrobních myších. Exprese všech 5 komponent de novo steroidogeneze byla pozorována pouze v nadledvinách a tračníku. V lymfatických orgánech byla pozorována především exprese Star, Cyp11a1 a Hsd3b1, naznačující pouze schopnost produkce progesteronu, nikoli jeho další konverzi na kortikosteron. Vlivem přítomného mikrobiomu došlo k poklesu exprese Star ve všech studovaných tkáních, exprese ostatních enzymů byla ovlivněna tkáňově specificky či ovlivněna nebyla. Konvenční myši vykazovaly zvýšenou expresi Hsd11b1 ve slezině, v ostatních orgánech exprese ovlivněna nebyla. Exprese Hsd11b1 nebyla ovlivněna ani in vitro stimulací Toll-like receptorů v imunitních buňkách izolovaných z mesenteriálních lymfatických uzlin. Z výsledků lze tedy...The innate and adaptive immune processes are modulated by hormones including glucocorticoids and by microbiota. The exact mechanisms underlying the microbial and hormonal contributions to this control are not completely clear. Present study is therefore focused to crosstalk between microbiota and de novo biogenesis or local regeneration of glucocorticoids. In particular, the study analysed the effect of commensal microbiota on expression of genes encoding steroidogenic enzymes (Star, Cyp11a1, Hsd3b1, Cyp21a1, Cyp11b1) and regeneration of glucocorticoids (Hsd11b1) in adrenal glands, colon, spleen and mesenteric lymph nodes using conventional and germ-free mice. The expression of all 5 components of steroidogenesis was identified only in the adrenal gland and colon, whereas the lymphoid organs expressed predominantly Star, Cyp11a1 and Hsd3b1 indicating the ability to produce only progesterone but not corticosterone. Microbiota decreased the expression of Star in all studied tissues but the expression of other genes was insensitive to microbiota or did not respond homogenously depending on the tissue and gene. Hsd11b1 expression was upregulated by microbiota in the spleen but not in other tissues. Physiotherapy in Patient with Morbus Kienböck

Bakalářská práce se zabývá fyzioterapií u pacienta s morbus Kienböck. Předmětem práce je zpracovat komplexní informace o tomto onemocnění a na základě kineziologického rozboru sestavit a následně uskutečnit rehabilitační plán a poté zhodnotit jeho efekt. V teoretické části se v rámci celkové problematiky pojednává o funkci ruky, jemné motorice a úchopu. Dále je zde probrána kineziologie, biomechanika a anatomie zápěstí včetně podrobné anatomie a mikrostruktury os lunatum. Druhou polovinu teoretické části tvoří souhrnné informace a poznatky zahrnující definici tohoto onemocnění, jeho epidemiologii a historii, dále etiologii a patogenezi, klinický obraz, diagnostiku a léčbu. V závěru je popsána pooperační péče a fyzioterapie. V kapitole Metodika jsou uvedeny vyšetřovací a terapeutické postupy využité v této práci. Speciální část obsahuje kazuistiku pacienta s morbus Kienböck. Je zde zpracován vstupní kineziologický rozbor, na jehož základě je stanoven rehabilitační plán. Dále jsou zde popsány jednotlivé terapeutické jednotky, během nichž byl proveden také kontrolní kineziologický rozbor. V kapitole Výsledky je uveden výstupní kineziologický rozbor a následně zhodnocen efekt terapie. Získané výsledky potvrzují pozitivní vliv terapie a jsou zpracovány formou tabulek a slovního hodnocení. V kapitole Diskuze je poté rozebírána problematika tohoto onemocnění. Závěr hodnotí splnění cílů a přínos práce.My bachelor's project deals with physiotherapy of patients with Kienböck's disease. The purpose of my study is to gather complex information about this disease, to assemble and realize specific physiotherapy plan on the basis of kinesiological analysis, and finally to evaluate its effect. The theoretical part discusses the function of hand within the general problematics, focusing on fine motor skills and grip. Further on, the kinesiology, biomechanics and anatomy of the wrist is discussed, including detailed anatomy and microstructure of os lunatum. The second part consists of summary of information and findings comprising the definition of the disease, its epidemiology and history, etiology and pathogenesis, clinical picture, diagnosis and therapy. In conclusion, postoperative care and physiotherapy is described. The diagnostic and therapeutic methods used in this project are described in the Methodology section. The special section contains case report of a patient with Kienböck's disease. His initial kinesiological analysis is processed, and specific physiotherapy plan created on its basis. Further on, individial therapeutic units are described, during which control kinesiological analysis was performed. In Results section, his final kinesiological analysis is presented and its therapeutic effect evaluated. Gained results confirm the positive effect of the therapy and are presented as tables and verbal evaluation. In the Discussion part the problematics of this disease is finally analyzed. The Conclusion evaluates fulfillment of the aims and contribution of the work
