Influence of short-term simvastatin administration on parameters of autonomic nervous system activity and blood pressure in hypercholesterolemic patients with or without hypertension
Background The beneficial effects of statins are probably
not exclusively related to lipid lowering activity. Therefore,
we investigated the effect of simvastatin on blood pressure
and heart rate variability (HRV), which can be used for
studying autonomous nervous system activity.
Material and methods In a prospective, placebo-controlled,
crossover trial, we studied 9 males and 11 females aged
55.2 ± 9.7 years with hypercholesterolemia (TC > 220
mg/dl), and normal blood pressure or essential hypertension
stage 1–2. After 4 weeks of dietary modifications and
placebo administration all subjects were given 20 mg of
simvastatin for 4 weeks, followed by a placebo for another
4 weeks. Office and 24-h blood pressure (ABPM), the biochemical
parameters of neuroendocrine function and HRV
were evaluated. Time-domain and frequency-domain variables
were studied such as: SDNN, r-MSSD, pNN50, TP,
HF, LF and LF/HF ratio.
Results All subjects had significant decrease in total and
LDL-cholesterol as well as triglycerides. However, four
weeks of simvastatin was not associated with a significant
change in office or ambulatory blood pressure. Treatment
did not affect basal or stimulated plasma levels of
catecholamines, aldosterone, cortisol, neuropeptide Y,
renin activity. The differences in HRV between groups did not reach statistical significance. No significant correlation
between changes in serum lipid levels and change in cardiac
autonomic indices was found.
Conclusions This short, prospective, cross-over study has
not shown significant effects of simvastatin on blood pressure,
biochemical parameters of neuroendocrine function
and heart rate variability (HRV) parameters in patients
with hypercholesterolemia with or without hypertension
Elevated Neuropeptide Y Plasma Concentration in Non-dippers With Essential Hypertension
Wstęp Brak nocnego obniżenia ciśnienia tętniczego prowadzi do zwiększonej częstości powikłań narządowych nadciśnienia tętniczego, a jego przyczyny pozostają niejasne. Celem pracy była ocena zależności między biochemicznymi wykładnikami
aktywności współczulnej a rytmem dobowym ciśnienia u chorych z pierwotnym nadciśnieniem tętniczym.
Materiał i metody Do badania zakwalifikowano 68 chorych z nadciśnieniem tętniczym w średnim wieku 40 ± 1 lat (20 K, 48 M). Do grupy kontrolnej włączono 25 zdrowych ochotników w średnim wieku 38 ± 1 lat (8 K i 17 M). U badanych wykonywano
24-godzinny pomiar ciśnienia (SpaceLabs 90207) oraz oznaczano we krwi stężenie katecholamin i neuropeptydu Y. Całodobowo prowadzono zbiórkę moczu w celu oznaczenia
wydalania katecholamin.
Wyniki U chorych z nadciśnieniem tętniczym stężenie neuropeptydu Y było
wyższe niż u osób zdrowych. Stężenie katecholamin w surowicy i ich wydalanie z moczem były w obu grupach podobne. Chorych z nadciśnieniem podzielono na dwie grupy zależnie od obecności - dippers (46 chorych) - lub braku nocnego obniżenia ciśnienia - non-dippers (22 chorych). Stężenie neuropeptydu Y było istotnie wyższe w grupie non-dippers niż w grupie dippers (10,6 ± 1 vs. 8,6 ± 1 fmol/ml, p < 0,05). Nie stwierdzono między grupami różnic w zakresie stężenia katecholamin w surowicy i katecholamin wydalanych z moczem. W obu grupach wydalanie katecholamin w czasie nocy było istotnie niższe aniżeli w ciągu dnia. Nocne i dzienne wydalanie katecholamin było podobne w grupie dippers i non-dippers.
Wnioski Uzyskane wyniki wskazują, że neuropeptyd Y może wpływać na zaburzenie rytmu dobowego ciśnienia tętniczego u chorych z nadciśnieniem i brakiem nocnego obniżenia ciśnienia. Prawdopodobnie katecholaminy nie wpływają na dobowy rytm ciśnienia tętniczego.Background The aim of the study was to investigate the relationship between biochemical indices of sympathetic activity and diurnal blood pressure rhythm in patients with essential hypertension.
Material and methods The study included 68 patients with mild-to-moderate essential hypertension (EH) (mean age 40 ± 1 yrs; 20 F, 48 M). The control group consisted of 25 healthy normotensive volunteers (C) (mean age 38 ± 1 yrs; 8 F, 17 M). Twenty-four hour ambulatory blood pressure monitoring was obtained using the SpaceLabs 90207. Blood samples for NPY and catecholamines were taken. Urine for catecholamines determination was collected.
Results In patients with EH NPY, plasma concentration was significantly higher as compared to C. Plasma catecholamine and urine catecholamine excretion was similar in patients with EH and in C. Patients with EH were divided into two groups: dippers (46 pts) and non-dippers (22 pts). In EH, the NPY plasma concentration was significantly higher in non-dippers than in dippers (10,6 ± 1 vs. 8,6 ± 1 fmol/mL, respectively, p < 0,05). No differences in plasma catecholamine and urine catecholamine excretion were found between dippers and non-dippers. Urine catecholamine excretion during nighttime was significantly lower as compared to day-time excretion in dippers and non-dippers. No differences between the groups were found with regard to day-time and night-time catecholamine excretion.
Conclusion NPY may play a role in diurnal blood pressure rhythm disturbances in non-dippers with EH. Catecholamines appear not to influence non-dipper phenomenon
Structural Modifications of Nature-Inspired Indoloquinolines: A Mini Review of Their Potential Antiproliferative Activity
Cryptolepine, neocryptolepine and isocryptolepine are naturally occurring indoloquinoline alkaloids with various spectrum of biological properties. Structural modification is an extremely effective means to improve their bioactivities. This review enumerates several neocryptolepine and isocryptolepine analogues with potent antiproliferative activity against MV4-11 (leukemia), A549 (lung cancer), HCT116 (colon cancer) cell lines in vitro. Its activity towards normal mouse fibroblasts BALB/3T3 was also evaluated. Furthermore, structure activity relationships (SAR) are briefly discussed. The anticancer screening of neocryptolepine derivatives was performed in order to determine their cytotoxic and growth inhibitory activities across the JFCR39 cancer cell line panel
The Effect of Maternal Depression on Infant Attachment: A Systematic Review
Aims and objectives: The aim of this systematic review was to summarize the key findings of
empirical studies assessing the influence of maternal depression on child attachment security measured
before 24 months after birth. Method: The study followed the Preferred Reporting Items for Systematic
Reviews and Meta-Analyses (PRISMA) statement guidelines. A literature search was conducted
on the EBSCO (Academic Search Complete; Health Source: Nursing/Academic Edition; MEDLINE;
PsycARTICLES) and PubMed databases, with infant attachment AND depression as search terms with
Boolean operators. Study design or sample size did not affect inclusion. After screening, 29 of the
1510 unique publications originally identified were included in the review. Results: The studies reveal
an equivocal association between maternal depression and child attachment security. Our findings
indicate that depression had a significant influence on the attachment style almost only when
diagnosed by structured interview: Depression measured by self-descriptive questionnaires was
unrelated to attachment style. Furthermore, postpartum depression was found to be significant only
when measured up to six months after childbirth. Conclusion: The relationship between maternal
depression and infant attachment is both complex and dynamic, and the possible negative effects of
depression might be compensated by maternal involvement in childcare. Therefore, further studies
in this area should employ a reliable methodology for diagnosing depression and a suitable time point
for measuring it; they should also adopt a multifactorial and prospective approach. It is important to
note that breastfeeding/formula feeding was omitted as a factor in the majority of studies
Combined anticancer therapy with imidazoacridinone analogue C-1305 and paclitaxel in human lung and colon cancer xenografts : modulation of tumour angiogenesis
The acridanone derivative 5-dimethylaminopropylamino-8-hydroxytriazoloacridinone (C-1305) has been described as a potent inhibitor of cancer cell growth. Its mechanism of action in in vitro conditions was attributed, among others, to its ability to bind and stabilize the microtubule network and subsequently exhibit its tumour-suppressive effects in synergy with paclitaxel (PTX). Therefore, the objective of the present study was to analyse the effects of the combined treatment of C-1305 and PTX in vivo. In addition, considering the results of previous genomic analyses, particular attention was given to the effects of this treatment on tumour angiogenesis. Treatment with C-1305 revealed antitumor effect in A549 lung cancer cells, and combined treatment with PTX showed tendency to anticancer activity in HCT116 colon cancer xenografts. It also improved tumour blood perfusion in both tumour models. The plasma level of CCL2 was increased and that of PDGF was decreased after combined treatment with C-1305 and PTX. The experimental results showed that the levels of FGF1, TGF-beta and Ang-4 decreased, whereas the levels of ERK1/2 and Akt phosphorylation increased in HCT116 tumour tissue following combined treatment with both drugs. The results of in vitro capillary-like structure formation assay demonstrated the inhibiting effect of C-1305 on this process. Although previous in vitro and in vivo studies suggested a positive effect of C-1305 on cancer cells, combined treatment of HCT116 human colon and A549 lung cancer cells with both PTX and C-1305 in vivo showed that the antitumor activity was restricted and associated with the modulation of tumour angiogenesis
Is There a Relationship between Leptin Concentration, Sympathetic Nervous System and Left Ventricular Mass Index in Patients with Essential Hypertension According to Gender and Body Mass Index?
Wstęp Celem niniejszej pracy była ocena współzależności między stężeniem leptyny a układem współczulnym, ciśnieniem tętniczym i masą lewej komory serca u chorych z nadciśnieniem tętniczym pierwotnym w zależności od płci i wskaźnika
masy ciała (BMI).
Materiał i metody Badaniami objęto 46 chorych (21K, 25M; śr. wiek 44,0 ± 8,4 roku), z nadciśnieniem tętniczym pierwotnym. Grupę kontrolną stanowiło 37 ochotników (19K, 18M; śr. wiek 40,8 ± 9,8 roku). Badanych podzielono na grupy
w zależności od płci i BMI (poniżej i powyżej 25).
U wszystkich badanych oznaczano stężenie leptyny (RIA), noradrenaliny (NA) i adrenaliny (A) (HPLC) oraz NPY (RIA) we krwi; mierzono odsetek tkanki tłuszczowej i wykonywano badanie echokardiograficzne.
Wyniki U chorych z nadciśnieniem tętniczym pierwotnym stężenie leptyny
było podobne jak u osób bez nadciśnienia w porównywalnych grupach kontrolnych, było natomiast znamiennie wyższe u kobiet i u osób z nadwagą. Stężenie NA było istotnie wyższe u chorych z nadwagą. W badanych grupach nie wykazano różnic w stężeniu NPY, bez względu na płeć i BMI. Wskaźnik masy lewej komory serca (LVMI) był znamiennie wyższy u kobiet z nadciśnieniem tętniczym pierwotnym w porównaniu z grupą kontrolną, niezależnie od BMI. Wieloczynnikowa analiza regresji wykazała w całej grupie badanych (n = 83) związek między leptyną a płcią, BMI, wiekiem, ciśnieniem skurczowym i NA (r2 = 0,28, p < 0,0001), a u kobiet i mężczyzn z nadciśnieniem tętniczym pierwotnym - związek między leptyną a odsetkiem tłuszczu
i NA (r2 = 0,34, p < 0,001). U chorych mężczyzn z nadwagą analiza regresji wykazała związek między leptyną a LVMI i ciśnieniem rozkurczowym (DBP), natomiast u chorych kobiet z nadwagą - z wiekiem, odsetkiem tłuszczu i LVMI.
Wnioski Uzyskane wyniki mogą sugerować udział leptyny w regulacji ciśnienia tętniczego poprzez układ współczulny, jak również w patogenezie przerostu masy lewej komory serca.Background The aim of the study was to evaluate the relationship between plasma leptin, sympathetic nervous system, blood pressure and left ventricular mass index in patients with essential hypertension (EH) according to gender and BMI.

Results In patients with EH plasma leptin concentration was comparable to controls, but values were significantly higher in women and in overweight patients.
Plasma NA concentration was significantly elevated in overweight EH patients. No difference in NPY concentration was found in none of compared groups. LVMI was significantly higher in women with EH in comparison with controls, independently on body mass index. Multiple regression analysis indicated the relation between
leptin and gender, BMI, age, systolic blood pressure and NA (r2 = 0,28, p < 0,0001) in all investigated subjects, as well as between leptin and body fat % and NA in EH patients (r2 = 0,34, p < 0,001). Multiple regression analysis
indicated also the relation between leptin and LVMI, A and diastolic blood pressure (r2 = 0,54, p < 0,05) in overweight EH men as well as between leptin and age, body fat % and LVMI in overweight EH women (r2 = 0,69, p < 0,05).
Conclusion This study suggests that leptin may contribute to regulation of blood pressure through sympathetic nervous system and may have implication in pathogenesis of left ventricular hypertrophy
Improved HDAC Inhibition, Stronger Cytotoxic Effect and Higher Selectivity against Leukemias and Lymphomas of Novel, Tricyclic Vorinostat Analogues
Histone deacetylase (HDAC) inhibitors are a class of drugs used in the cancer treatment. Here, we developed a library of 19 analogues of Vorinostat, an HDAC inhibitor used in lymphomas treatment. In Vorinostat, we replaced the hydrophobic phenyl group with various tricyclic ‘caps’ possessing a central, eight-membered, heterocyclic ring, and investigated the HDAC activity and cytotoxic effect on the cancer and normal cell lines. We found that 3 out of the 19 compounds, based on dibenzo[b,f]azocin-6(5H)-one, 11,12-dihydrodibenzo[b,f]azocin- 6(5H)-one, and benzo[b]naphtho[2,3-f][1,5]diazocine-6,14(5H,13H)-dione scaffolds, showed better HDACs inhibition than the referenced Vorinostat. In leukemic cell line MV4-11 and in the lymphoma cell line Daudi, three compounds showed lower IC50 values than Vorinostat. These compounds had higher activity and selectivity against MV4-11 and Daudi cell lines than reference Vorinostat. We also observed a strong correlation between HDACs inhibition and the cytotoxic effect. Cell lines derived from solid tumours: A549 (lung carcinoma) and MCF-7 (breast adenocarcinoma) as well as reference BALB/3T3 (normal murine fibroblasts) were less susceptible to compounds tested. Developed derivatives show improved properties than Vorinostat, thus they could be considered as possible agents for leukemia and lymphoma treatment

międzynarodowych zespołów badawczych. Ich wiedza i kompetencje wpływają na
pozycję uczelni w międzynarodowym środowisku naukowym.
W Politechnice Krakowskiej oraz w środowisku akademickim szczególną pozycję
zajmuje Wydział Architektury. Jego historię od 1945 roku, a więc od czasu utworzenia
uczelni, tworzą wybitni naukowcy i twórcy. Uczestniczą oni aktywnie w rozwoju
dyscypliny architektura i urbanistyka, a jednocześnie są zarówno uznanymi teoretykami,
jak i twórcami cenionych dzieł w zakresie architektury, urbanistyki oraz założeń
krajobrazowych.
Do grona tych osób niewątpliwie należał prof. Wojciech Kosiński. Z Wydziałem
Architektury był związany prawie 60 lat. Tu ukończył studia, w 1967 roku pod promotorstwem
prof. Włodzimierza Gruszczyńskiego obronił dyplom i rozpoczął pracę.
Cała Jego kariera naukowa związana była z Wydziałem Architektury – tu w 1976
roku uzyskał stopień doktora (promotor – prof. Witold Cęckiewicz), w roku 2001
doktora habilitowanego, a w 2012 roku tytuł naukowy profesora. Był znakomitą postacią,
dydaktykiem lubianym przez studentów i uznanym w kraju i za granicą, autorem
wielu publikacji naukowych. Był autorem lub współautorem licznych realizacji
architektonicznych, laureatem wielu prestiżowych konkursów architektonicznych,
znakomitym rysownikiem."(...
Problem decyzyjności w zakresie doboru kolorystyki architektury - teoria i praktyka. Analiza na wybranych przykładach z Polski i ze Słowacji
The colours of external architecture are crucial to the reception of urbanized space. An appropriate concept is necessary to maintain order and harmony of colours. Decision-making in this respect was analysed, based on Polish and Slovak legal acts. The formal and actual status was compared through analyses of local spatial development plans and site inspections. For a broader perspective, examples from some of the countries are presented. Using a survey, public knowledge of the current legal situation related to colour decision-making was investigated alongside preferences as to who should manage this issue. It was also investigated how space users assessed the external colours of buildings in the areas under study.Kolorystyka zewnętrznej architektury ma kluczowe znaczenie w odbiorze przestrzeni zurbanizowanej. Dla zachowania ładu oraz harmonii barw niezbędna jest odpowiednia koncepcja. Przeanalizowano kwestię decyzyjności w tym zakresie, opierając się na aktach prawnych polskich i słowackich. Stan formalny z rzeczywistym porównano za pomocą analiz miejscowych planów zagospodarowania przestrzennego oraz wizji lokalnych. Dla szerszej perspektywy przedstawiono przykłady z różnych krajów. Poprzez ankietyzację zweryfikowano wiedzę społeczną dotyczącą obecnej sytuacji prawnej związanej z decyzyjnością kolorystyczną, a także preferencje w kwestii tego, kto powinien tą kwestią zarządzać. Sprawdzono także, jak użytkownicy przestrzeni oceniają zewnętrzne barwy obiektów na badanych obszarach
