    Interactive effects of solar radiation and inorganic nitrogen and phosphate on biofiltration capacity, biomass production, photosynthetic activity and the accumulation of bio-active compounds were studied in the red alga Gracilaria cornea grown in tanks with a seawater open-flow system during 35 days. Two light conditions were utilized: outdoor (O), full solar natural radiation, and indoor, inside of a greenhouse (G) where UV-B radiation was cut-off and part of the UV-A radiation was filtered. Two inorganic nitrogen and phosphate concentrations were used: High nutrients (HN; 100–200μM of NH4+ and 20μM of KHPO4) and Low nutrients (LN; 10–20μM of NH4+ and 1μM of KHPO4). Growth and biomass productivity were related to the daily integrated electron transport rate determined in situ, as an estimator of daily photosynthetic activity. Nitrogen uptake efficiency (NUE) was close to 100 % under LN, whereas under HN it ranged from 50 to 70 % in the first week of culture, decreasing to 10–15 % in the rest of the experimental period. Nitrogen uptake rate (NUR) ranged from 20 to 45 mmol N m−2 h−1 under HN, and 5 to 18 mmol N m−2 h−1 in LN treatments. Morphological and pigmentation changes were evident through the culture period. The thalli under HN were more reddish under the indoor treatments, than that in LN. The internal compounds increased throughout the experimental period. Mycosporine-like amino acids (MAAs) were accumulated under HN. N plays a photoprotective role due to both the increased photosynthesis and the MAA content. Maximal MAA productivity reached 113–253 mg MAAs m−2 d−1 under the O-HN treatment, the highest level reported until now in the bibliography. G. cornea could be used for bioremediation of high N content waters.     Antioxidant Activity and Phenolic Content of Marine Dissolved Organic Matter and Their Relation to Molecular Composition

    The potential of marine dissolved organic matter (DOM) for free radical scavenging has been extensively evaluated, however, the quantitative assessment of the antioxidant potential has been recently measured for the first time. The linkage of the DOM antioxidant potential to its molecular composition has not yet been examined. Following this line, this article takes a step forward by assessing, throughout a polarity-mediated fractionation, (1) the antioxidant capacity and phenolic content and (2) the molecular characterization of DOM in a more exhaustive manner. (3) The DOM antioxidant potential and phenolic content was linked to the molecular composition of DOM, which was molecularly characterized using ultrahigh resolution Fourier transform Ion Cyclotron Resonance mass spectrometry (FT-ICR MS). Antioxidant activity and phenolic content were quantified by the free radical 2,2’-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS⋅) and the Folin-Ciocalteu methods, respectively. We considered three types of different natural DOM samples: the deep North Pacific Ocean, the oligotrophic surface of the North Pacific Ocean and porewater from the sulfidic tidal flats of the Wadden Sea. Bulk porewater and its individual polarity fractions presented the highest antioxidant activity and phenolic content. DOM from the water column samples had lower antioxidant activity and phenolic content than porewater, but exceeded what it is commonly found in macroalgae, microalgae, fruits and vegetables with cosmeceutical purposes. Our values were similar to published values for terrestrial DOM. The variations in bioactivity were dependent on polarity and molecular composition. The high resolution and high mass accuracy used to determine the molecular composition of marine DOM and the chemometric and multistatistical analyses employed have allowed to distinguish molecular categories that are related to the bioactive potential. As a future perspective, we performed cytotoxicity tests with human cells and propose marine DOM as a natural ingredient for the development of cosmeceutical products

    Using pedigree information to monitor genetic variability of endangered populations: the Xalda sheep breed of Asturias as an example

    The aim of this work is to highlight the need of monitoring small populations to conserve their genetic variability by using a set of parameters to characterize both the structure of populations and management practices. As a representative example we analyse the pedigree information of the endangered Xalda sheep breed of Asturias. The herdbook of Xalda sheep included a total of 805 animals and 62 herds. The number of founders was 329. Nowadays, there are 562 live animals and 26 active herds. The breed is in risk of losing genetic diversity because of the abusive use of certain individuals as parents. The effective number of founder animals is 81.1. The effective number of founder herds is 9.9. The average value of inbreeding in the whole Xalda population was 1.5%. The average relatedness (AR) coefficient reached 1.8% in the whole pedigree. The genetic representation of the lines of founders is unbalanced. Inbreeding trends and effective size do not provide realistic information concerning the risk of loss of diversity as a result of the shallowness of the genealogical information. We suggest the monitoring of the breed using AR to unbalance the genetic contributions of specific individuals, equalizing the genetic representation of the founders and lines in the population. In addition, AR can suggest the introduction of new, under-represented animals in herds showing high average AR values relative to the population. Our results can be useful to improve the development of conservation initiatives involving open herdbooks to avoid the risk of loss of genetic diversity caused by incorrect management practices

    Assessment of Time and Frequency Domain Entropies to Detect Sleep Apnoea in Heart Rate Variability Recordings from Men and Women

    Producción CientíficaHeart rate variability (HRV) provides useful information about heart dynamics both under healthy and pathological conditions. Entropy measures have shown their utility to characterize these dynamics. In this paper, we assess the ability of spectral entropy (SE) and multiscale entropy (MsE) to characterize the sleep apnoea-hypopnea syndrome (SAHS) in HRV recordings from 188 subjects. Additionally, we evaluate eventual differences in these analyses depending on the gender. We found that the SE computed from the very low frequency band and the low frequency band showed ability to characterize SAHS regardless the gender; and that MsE features may be able to distinguish gender specificities. SE and MsE showed complementarity to detect SAHS, since several features from both analyses were automatically selected by the forward-selection backward-elimination algorithm. Finally, SAHS was modelled through logistic regression (LR) by using optimum sets of selected features. Modelling SAHS by genders reached significant higher performance than doing it in a jointly way. The highest diagnostic ability was reached by modelling SAHS in women. The LR classifier achieved 85.2% accuracy (Acc) and 0.951 area under the ROC curve (AROC). LR for men reached 77.6% Acc and 0.895 AROC, whereas LR for the whole set reached 72.3% Acc and 0.885 AROC.     Stereoselective Alkylation of Chiral Titanium(IV) Enolates with tert-Butyl Peresters

    [EN]Here, we present a new stereoselective alkylation of titanium(IV) enolates of chiral N-acyl oxazolidinones with tert-butyl peresters from Calpha-branched aliphatic carboxylic acids, which proceeds through the decarboxylation of the peresters and the subsequent formation of alkyl radicals to produce the alkylated adducts with an excellent diastereoselectivity. Theoretical calculations account for the observed reactivity and the outstanding stereocontrol.     Compuestos perfluorados en equinodermos marinos: metodología analítica para su determinación y monitorización

    Los disruptores endocrinos están en el punto de mira de estudios ambientales y ecotoxicológicos. Entre ellos destacan los compuestos perfluorados, tanto por su amplio uso industrial y doméstico como por su elevada actividad estrogénica. Se descargan al medio ambiente a través de efluentes industriales y de estaciones depuradoras de aguas residuales afectando principalmente a la biota donde pueden bioacumularse. En este trabajo se propone un método analítico para la determinación de seis compuestos perfluorados en organismos marinos (Holothuria tubulosa) y se lleva a cabo un programa de monitorización ambiental para el seguimiento de estos compuestos. El análisis de las muestras se realizó mediante extracción con disolventes, limpieza del extracto por extracción en fase sólida dispersiva y posterior determinación mediante cromatografía de líquidos con detección de espectrometría de masas en tándem. Las recuperaciones obtenidas se situaron en el rango de 84 a 101 % y límites de cuantificación inferior a 0,03 ng/g (peso seco (ps)). Todas las muestras dieron positivas en el análisis de los contaminantes con concentraciones entre 667 ng/g (ps) para el ácido perfluorooctanoico y 0,81 ng/g (ps) para el ácido perfluoropentanoico. Además, se observaron concentraciones más elevadas para los compuestos perfluorados de mayor cadena fluorocarbonada.In recent years endocrine disruptors have come into the spotlight of environmental and ecotoxicological studies. Among them, perfluorinated compounds stand out, both, for their wide industrial and domestic use and for their elevated estrogenic activity, showing adverse effects at trace level. They are released into the environment through industrial waste and wastewater discharges affecting to marine organisms, where they can accumulate. To contribute to this goal, this work proposes an analytical method for the simultaneous determination of six perfluorinated compounds in marine organisms (Holothuria tubulosa) and an environmental monitoring program is carried out in these organisms. The sample treatment involve steps of solvent extraction and clean-up of the extracts with dispersive sorbents prior to liquid chromatography–tandem mass spectrometry analysis. Recoveries between 84 and 101 %, precision (RSD < 9 %) and limits of quantification below 0.03 ng/g dry weight (d.w.) were achieved. All tested samples were positive in the analysis of contaminants with concentrations between 667 ng/g (dw) for perflorooctanoic acid (PFOA) and 0.81 ng/g (dw) for perfloropentanoic acid (PFPeA). In general, perfluorinated compounds of larger fluorocarbonated chain were quantified at higher concentration levels than those of shorter one.Plan Propio de la Universidad de Sevilla Proyecto: 2017/0000096
