Evaluation of Highway-pilot Function Based on FMEA Safety Analysis
Cybersecurity is becoming more and more relevant. Autonomous vehicles handle large amounts of data and can connect to more and more existing devices, smartphones, tablets, or even other cars and systems. This poses the risk of unauthorized access to data. Theoretically cars have separate computer units, operate in isolation, and are not connected, so there is less possibility to be attacked. However if the vehicles are interconnected, hackers can have easier access to personal data. They can get information about the location of the car owner, their typical trips, and, for example, allow an intruder to know when the tracked person is not at home. Furthermore it can also be happened that the vehicle operation is maliciously disturbed, which can result in a security risk for the passengers. In extreme cases, computer terrorist attacks can also be prepared - large-scale interventions on roads can lead to chaos across a region or country. In accordance with the introduced threats, it is a crucial objective of this research to indicate specific methods, which can help the industry to evaluate and prepare for these kinds of attacks in a proper way
Kenyérbúza környezeti adaptációjának molekuláris-genetikai vizsgálata
Az adaptációs képesség meghatározásában a vernalizációs igényért, a nappalhossz érzékenységért és a cirkadián ritmusért felelős géncsoportok fő szerepet játszanak.
Az egyedfejlődési gének aktivitása határozott napi ciklikusságot mutat, azonban ennek mértéke és mintázata jelentősen függ a környezeti hőmérséklet változásától, a genotípustól, illetve e két faktor kölcsönhatásától is. A cirkadián gének napi expressziós ciklusa az általunk alkalmazott hőmérsékleti kezeléstől és genotípustól függetlenül minden esetben határozott volt. Az egyes genotípusokban hőmérsékleti kezelés hatására jelentősen változhat a génaktivitás csúcsértéke. A két kezelés hatására a VRN1 gén expressziós értéke nem változott számottevően, míg a VRN2 és a VRN3 géneké 18°C-os körülmények között jelentős mértékben nőtt. A PPD1 gén aktivitásának napi csúcsértéke 18°C-on mindhárom fajta esetében megemelkedett, ami a kései kalászolású fajták esetében még inkább kifejezőbbé vált. A korai fajta esetében a CCA1 gén csúcsértéke közel kétszeresére emelkedett 18°C-os kezelés hatására, míg a TOC1 gén esetében ez a jelenség üvegházi körülmények között volt megfigyelhető. PRR95 gén aktivitása 18°C-on több mint kétszeresére emelkedett. A LUX gén esetében a késői fajták 18°C-on közel háromszoros génexpressziós maximumértékekkel rendelkeznek
Hosszú- és rövidnappalos megvilágítás hatása a kenyérbúza egyedfejlődési dinamikájára
A kenyérbúza esetében a domináns nappalhossz-érzéketlen alléltípusok korai virágzást eredményeznek rövid nappalon, míg a nappalhossz-érzékeny alléltípust hordozó genotípusok kalászolása vontatottá válik, illetve teljes mértékben el is maradhat. A nappalhossz érzékelése hatással van a búza intenzív szárnövekedési fázisának kezdeti idejére és hosszára is, amely alapvető szerepet játszik az adaptációs képesség meghatározásában. A vizsgált nappalhossz-érzéketlen és érzékeny csoportok átlagmagasságuk alapján nem mutattak jelentős különbséget a kezelések hatására (46-50 cm), azonban növekedési dinamikájukat megfigyelve szignifikáns eltérés volt tapasztalható rövid nappalos megvilágítás mellett. Hosszú nappalos körülmények között mind a nappalhossz-érzéketlen, mind a nappalhossz-érzékeny allélt hordozó genotípusok intenzív szárnövekedésük kezdeti szakaszában, illetve az intenzív szárnövekedésük hosszában is hasonló átlagértékeket mutattak (a regressziós egyenes meredekségének értéke: 0,025 és 0,024, illetve 26 nap és 29 nap). Ezzel szemben rövid nappalon a két csoport között ezek az átlagértékek 0,01 és 0,008, illetve 39 nap és 57 nap volt. A nappalhossz-érzéketlen genotípusok rövid nappalon megfigyelt intenzív szárnövekedés alatti átlagos fejlődési dinamikája kétszer akkora volt (a regressziós egyenes meredekségének értéke: 0,04), mint a nappalhossz-érzékeny fajtáknál (0,02). A két eltérő nappalhossz-érzékenységű csoport egyedfejlődési paraméterei között is jelentős eltéréseket mutattunk ki. A nappalhossz-érzéketlen fajták intenzív szárnövekedése átlagosan még az első szárcsomó megjelenése előtt két nappal elkezdődött hosszú nappalos körülmények között, míg rövid nappalon ez az érték 3 nap volt. Ezzel szemben a nappalhossz-érzékeny csoportnál ez az aránypár 2 nap és 11 nap volt
Photometric and radial-velocity time-series of RR Lyrae stars in M3: analysis of single-mode variables
We present the first simultaneous photometric and spectroscopic investigation of a large set of RR Lyrae variables in a globular cluster. The radial-velocity data presented comprise the largest sample of RVs of RR Lyrae stars ever obtained. The target is M3; BVI_C time-series of 111 and b flux data of further 64 RRab stars, and RV data of 79 RR Lyrae stars are published. Blazhko modulation of the light curves of 47 percent of the RRab stars are detected. The mean value of the center-of-mass velocities of RR Lyrae stars is −146.8 km/s with 4.52 km/s standard deviation, which is in vgood agreement with the results obtained for the red giants of the cluster. The Φ21 RV
phase difference of the RV curves of RRab stars is found to be uniformly constant both for the M3 and for Galactic field RRab stars; no period or metallicity dependence of the Φ21 RV is detected. The Baade-Wesselink distances of 26 non-Blazhko variables with the best phase-coverage radial-velocity curves are determined; the corresponding distance of the cluster, 10480±210 pc, agrees with the previous literature information. A quadratic formula for the Apuls−AV relation of RRab stars is given, which is valid for both OoI and OoII variables. We also show that the (V − I)0 of RRab stars measured at light minimum is period dependent, there is at least 0.1 mag difference between the colours at minimum light of the shortest- and longest-period variables
A convenient method to pre-screen candidate guide RNAs for CRISPR/Cas9 gene editing by NHEJ-mediated integration of a 'self-cleaving' GFP-expression plasmid.
The efficacies of guide RNAs (gRNAs), the short RNA molecules
that bind to and determine the sequence specificity of the
Streptococcus pyogenes Cas9 nuclease, to mediate DNA cleavage
vary dramatically. Thus, the selection of appropriate target
sites, and hence spacer sequence, is critical for most
applications. Here, we describe a simple, unparalleled method
for experimentally pre-testing the efficiencies of various
gRNAs targeting a gene. The method explores NHEJ-cloning,
genomic integration of a GFP-expressing plasmid without
homologous arms and linearized in-cell. The use of 'self-
cleaving' GFP-plasmids containing universal gRNAs and
corresponding targets alleviates cloning burdens when this
method is applied. These universal gRNAs mediate efficient
plasmid cleavage and are designed to avoid genomic targets in
several model species. The method combines the advantages of
the straightforward FACS detection provided by applying
fluorescent reporter systems and of the PCR-based approaches
being capable of testing targets in their genomic context,
without necessitating any extra cloning steps. Additionally,
we show that NHEJ-cloning can also be used in mammalian cells
for targeted integration of donor plasmids up to 10 kb in
size, with up to 30% efficiency, without any selection or
enrichment
A Sequence of Phase Transformations and Phases in NiCoFeCrGa High Entropy Alloy
The present investigation is directed to phase transitions in the equimolar NiCoFeCrGa high entropy alloy, which is a mixture of face-centered cubic (FCC) and body-centered cubic (BCC) crystalline phases. The microstructure of the samples was investigated by using scanning electron microscopy (SEM), time-of-flight secondary ion mass spectroscopy (TOF-SIMS), transmission electron microscopy-based energy-dispersive spectroscopy (EDS) and electron energy loss spectroscopy (EELS), as well as X-ray diffraction (XRD) measurements. Based on the phases observed in different temperature ranges, a sequence of the phase transitions can be established, showing that in a realistic process, when freely cooling the sample with the furnace from high to room temperature, a microstructure having spinodal-like decomposition can also be expected. The elemental mapping and magnetic behaviors of this decomposed structure are also studied
