    Swelling-induced bending and pumping in homogeneous thin sheets

    We realize steady curved shapes from homogeneous hydrogel flat structures which are in contact with two environments at different chemical conditions. We numerically investigate the behaviour of beam-like and plate-like structures during the transient state, which realize osmotic pumps. Through numerical experiments, we determine the relationship between the difference in the chemical potentials at the top and bottom of a beam and the curvature of the bent beam as well as the Gaussian curvature of a spherical cap morphed from a flat plate. We also propose an approximate modeling of both the beam and the plate, to evaluate explicitly that relationship and show the good agreement between those formulas and the outcomes of the numerical simulations

    This text presents theories that give alternatives to the substantialist theory of being, passing through Kant, Zemach, Williams, Frege, Lawe, and has its focus on Lorenz Puntel's categorical theory. In the search for the absolutely universal (or primordial) dimension of being, Puntel finds in the meta-(...) particle the correspondence to what can be considered as the absolutely universal being. This stems from his logical-semantic reflection, which starts from the theoretical operator "is it the case", which is the universal meta-(...) particle before it. In fact, the absolutely universal particle, or being primordial, is presupposed by the theoretical operator, and here it is intended to deal with Puntel articulate and reach this conclusion, as it is also argued here that for Puntel such an absolutely universal particle does not have the predicate “exist” or “be determined”.O presente texto apresenta teorias que dão alternativas à teoria substancialista do ser, passando por Kant, Zemach, Williams, Frege, Lawe, e tem seu foco na teoria categorial de Lorenz Puntel. Na procura da dimensão absolutamente universal (ou primordial) do ser, Puntel encontra na partícula meta-(...) a correspondência ao que se pode considerar como o ser absolutamente universal.     Immune-mediated loss of transgene expression from virally transduced brain cells is irreversible, mediated by IFNγ, perforin, and TNFα, and due to the elimination of transduced cells

    The adaptive immune response to viral vectors reduces vector-mediated transgene expression from the brain. It is unknown, however, whether this loss is caused by functional downregulation of transgene expression or death of transduced cells. Herein, we demonstrate that during the elimination of transgene expression, the brain becomes infiltrated with CD4 and CD8 T cells and that these T cells are necessary for transgene elimination. Further, the loss of transgene-expressing brain cells fails to occur in the absence of IFNγ, perforin, and TNFα receptor. Two methods to induce severe immune suppression in immunized animals also fail to restitute transgene expression, demonstrating the irreversibility of this process. The need for cytotoxic molecules and the irreversibility of the reduction in transgene expression suggested to us that elimination of transduced cells is responsible for the loss of transgene expression. A new experimental paradigm that discriminates between downregulation of transgene expression and the elimination of transduced cells demonstrates that transduced cells are lost from the brain upon the induction of a specific antiviral immune response.     O outro do “quadro referencial teórico” – uma resposta a Lorenz Puntel

    O artigo tem o objetivo de oferecer uma resposta às críticas que Puntel fez a Levinas em seu livro Ser e Deus. Iniciamos nossa abordagem com uma contextualização do pensamento punteliano. Dado esse passo inaugural, propomos uma análise de seu exame crítico à obra de Levinas em dois tempos, seguindo a ordem de seus argumentos em Ser e Deus. Primeiramente, analisamos as críticas de Puntel às “assunções fundamentais” da filosofia levinasiana, sobre teoria/conhecimento, metafísica e Ser. Depois disso, analisamos suas críticas à “concepção de transcendência e Deus” em Levinas. O título deste artigo é uma provocação a partir da pergunta: “Qual é o lugar do absolutamente Outro no quadro referencial teórico de Puntel?”. Tal como ocorre na ontoteologia e na ontologia fundamental, o absolutamente Outro não encontra lugar no quadro referencial teórico de Puntel.   Palavras-chave: Ontoteologia, Ética, Alteridade, Levinas, Puntel

    Modeling water penetration at dam-foundation joint

    When fracture occurs in a concrete dam, the crack mouth is typically exposed to water. Very often this phenomenon occurs at the dam-foundation joint and is driven also by the fluid pressure inside the crack. Since the joint is the weakest point in the structure, this evolutionary process determines the load bearing capacity of the dam. In this paper the cracked joint is analyzed through the cohesive model proposed by Cocchetti et al. [Cocchetti G, Maier G, Shen X. Piecewise linear models for interfaces and mixed mode cohesive cracks. J Engng Mech (ASCE) 2002;3:279-98.], which takes into account the coupled degradation of normal and tangential strength. The water pressure inside the crack, which reduces fracture energy and increases the driving forces, is analyzed through the model proposed by Reich et al. [Reich W, Bruhwiler E, Slowik V, Saouma VE. Some numerical results are presented which refer to the benchmark problem proposed in 1999 by the International Commission On Large Dams. During the evolutionary process the horizontal dam crest displacement has been found to be a monotonic increasing function of the external load multiplier. As the fictitious process zone moves from the upstream to the downstream edge a transition occurs in the path of crack formation: the initial phase is dominated by the opening displacement, on the contrary afterwards the shear displacement dominates.     Development of a nitrogen recommendation tool for corn considering static and dynamic variables

    Many soil and weather variables can affect the economical optimum nitrogen (N) rate (EONR) for maize. We classified 54 potential factors as dynamic (change rapidly over time, e.g. soil water) and static (change slowly over time, e.g. soil organic matter) and explored their relative importance on EONR and yield prediction by analyzing a dataset with 51 N trials from Central-West region of Argentina. Across trials, the average EONR was 113 ± 83 kg N ha−1 and the average optimum yield was 12.3 ± 2.2 Mg ha−1, which is roughly 50% higher than the current N rates used and yields obtained by maize producers in that region. Dynamic factors alone explained 50% of the variability in the EONR whereas static factors explained only 20%. Best EONR predictions resulted by combining one static variable (soil depth) together with four dynamic variables (number of days with precipitation\u3e20 mm, residue amount, soil nitrate at planting, and heat stress around silking). The resulting EONR model had a mean absolute error of 39 kg N ha−1 and an adjusted R2 of 0.61. Interestingly, the yield of the previous crop was not an important factor explaining EONR variability. Regression models for yield at optimum and at zero N fertilization rate as well as regression models to be used as forecasting tools at maize planting time were developed and discussed. The proposed regression models are driven by few easy to measure variables filling the gap between simple (minimum to no inputs) and complex EONR prediction tools such as simulation models. In view of increasing data availability, our proposed models can be further improved and deployed across environments. Includes supplemental figures and table.     Study of the Efficacy, Biodistribution, and Safety Profile of Therapeutic Gutless Adenovirus Vectors as a Prelude to a Phase I Clinical Trial for Glioblastoma

    Glioblastoma multiforme (GBM) is the most common and most aggressive primary brain tumor in humans. Systemic immunity against gene therapy vectors has been shown to hamper therapeutic efficacy; however, helper-dependent high-capacity adenovirus (HC-Ad) vectors elicit sustained transgene expression, even in the presence of systemic anti-adenoviral immunity. We engineered HC-Ads encoding the conditional cytotoxic herpes simplex type 1 thymidine kinase (TK) and the immunostimulatory cytokine fms-like tyrosine kinase ligand 3 (Flt3L). Flt3L expression is under the control of the regulatable Tet-ON system. In anticipation of a phase I clinical trial for GBM, we assessed the therapeutic efficacy, biodistribution, and clinical and neurotoxicity with escalating doses of HC-Ad-TetOn-Flt3L + HC-Ad-TK in rats. Intratumoral administration of these therapeutic HC-Ads in rats bearing large intracranial GBMs led to long-term survival in ~70% of the animals and development of antiglioma immunological memory without signs of neuropathology or systemic toxicity. Systemic anti-adenoviral immunity did not affect therapeutic efficacy. These data support the idea that it would be useful to develop HC-Ad vectors further as a therapeutic gene-delivery platform to implement GBM phase I clinical trials
