Functional Imaging of the Outer Retinal Complex using High Fidelity Imaging Retinal Densitometry
We describe a new technique, high fidelity Imaging Retinal Densitometry (IRD), which probes the functional integrity of the outer retinal complex. We demonstrate the ability of the technique to map visual pigment optical density and synthesis rates in eyes with and without macular disease. A multispectral retinal imaging device obtained precise measurements of retinal reflectance over space and time. Data obtained from healthy controls and 5 patients with intermediate AMD, before and after photopigment bleaching, were used to quantify visual pigment metrics. Heat maps were plotted to summarise the topography of rod and cone pigment kinetics and descriptive statistics conducted to highlight differences between those with and without AMD. Rod and cone visual pigment synthesis rates in those with AMD (v = 0.043 SD 0.019 min-1 and v = 0.119 SD 0.046 min-1, respectively) were approximately half those observed in healthy controls (v = 0.079 SD 0.024 min-1 for rods and v = 0.206 SD 0.069 min-1 for cones). By mapping visual pigment kinetics across the central retina, high fidelity IRD provides a unique insight into outer retinal complex function. This new technique will improve the phenotypic characterisation, diagnosis and treatment monitoring of various ocular pathologies, including AMD
Multispectral imaging of organ viability during uterine transplantation surgery in rabbits and sheep
Uterine transplantation surgery (UTx) has been proposed as a treatment for permanent absolute uterine factor infertility (AUFI) in the case of the congenital absence or surgical removal of the uterus. Successful surgical attachment of the organ and its associated vasculature is essential for the organ’s reperfusion and long-term viability. Spectral imaging techniques have demonstrated the potential for the measurement of hemodynamics in medical applications. These involve the measurement of reflectance spectra by acquiring images of the tissue in different wavebands. Measures of tissue constituents at each pixel can then be extracted from these spectra through modeling of the light–tissue interaction. A multispectral imaging (MSI) laparoscope was used in sheep and rabbit UTx models to study short- and long-term changes in oxygen saturation following surgery. The whole organ was imaged in the donor and recipient animals in parallel with point measurements from a pulse oximeter. Imaging results confirmed the re-establishment of adequate perfusion in the transplanted organ after surgery. Cornual oxygenation trends measured with MSI are consistent with pulse oximeter readings, showing decreased StO2 immediately after anastomosis of the blood vessels. Long-term results show recovery of StO2 to preoperative levels
Spectral unmixing of Multispectral Lidar signals
In this paper, we present a Bayesian approach for spectral unmixing of
multispectral Lidar (MSL) data associated with surface reflection from targeted
surfaces composed of several known materials. The problem addressed is the
estimation of the positions and area distribution of each material. In the
Bayesian framework, appropriate prior distributions are assigned to the unknown
model parameters and a Markov chain Monte Carlo method is used to sample the
resulting posterior distribution. The performance of the proposed algorithm is
evaluated using synthetic MSL signals, for which single and multi-layered
models are derived. To evaluate the expected estimation performance associated
with MSL signal analysis, a Cramer-Rao lower bound associated with model
considered is also derived, and compared with the experimental data. Both the
theoretical lower bound and the experimental analysis will be of primary
assistance in future instrument design
