The first chapter, Different techniques to detect G protein coupled receptor heteromers, describes state of the art techniques based in biophysical principles applied to detect oligomeric aggregates formed by G-protein coupled receptors in the cell membrane, and describes how the knowledge generated can be applied to design new compounds for the treatment of neurological and mental diseases. Obesity has become a worldwide problem and it is well known that causes and exacerbates many health problems by promoting profound changes in physiological functions. Chapter 2, Analysis of the role of diet in the appearance of neurodegenerative processes, reviews the consequences of these metabolic alterations while considering their effects in the development of Type 2 Diabetes Mellitus, and their role in the appearance of cognitive impairments such as the sporadic forms of Alzheimer鈥檚 disease. The development of new cultivars facing climate change is an issue of great interest for the agrochemical industry and can be approached in different ways. Chapter 3, Arabidopsis Thaliana A Model for the Study of Plant Speciation, reviews different aspects of the plant immune system and the different layers of the plant immune response and signaling. The emerging field in plant research that studies how soil microbiota influences plant basic mechanisms is also discussed. Chagas disease is endemic in Latin America, but recently and due to human migrations, it is becoming a global health problem. In chapter 4, Trypanosoma cruzi infection diagnosis: New insights, challenges and perspectives, a group of experts from several institutions describe the different techniques that can be used for the serological diagnosis of the infection and the characterization of Trypanosoma cruzi, discuss the advantages and drawbacks of each method and propose improvements that would entail important savings for health institutions. Chapter 5, Nutrients, Control of Gene Expression and Metabolic Homeostasis, focuses on the molecular mechanisms that control metabolism by means of regulating gene expression in response to dietary inputs, to design new therapeutic strategies based on nutritional interventions against metabolic diseases. In this context, involvement of FGF21 hormone in the regulation of lipid metabolism during amino acid starvation is described, thus reinforcing its important role as an endocrine factor in coordinating energy homeostasis under a variety of nutritional conditions. This raises the possibility of dietary modulation of circulating levels of FGF21 as an alternative approach to its pharmacological administration. Biodegradable polymeric nanoparticles encapsulating neuroprotective drugs have enormous potential to treat neurodegenerative diseases, including Alzheimer麓s disease and glaucoma. Recent advances in the field are described in chapter 6, Polymeric nanoparticles for the treatment of neurodegenerative diseases: Alzheimer鈥檚 disease and glaucoma, specifically the preparation of engineered polymeric nanoparticles with attached peptides or antibodies to increase their bioavailability, favoring their transport through the blood brain barrier and the blood retinal barrier, avoiding at the same time possible drug adverse and toxic effects. The last chapter, Pentacyclic triterpenes in table olives: Determination of their composition and bioavailability by LC-M, is a review on the pentacyclic triterpenes contained in table olives, natural compounds of enormous interest due to their beneficial effects on human health, including hepatoprotective, anti-diabetic, antiviral, cardioprotective and antitumor activities. Authors describe a selective and sensitive LC鈥揗S method for the simultaneous determination of the main triterpenic compounds present in Olea europaea L. This opens the possibility to bioavailability studies after consumption of different foods, or administration of plants widely used in traditional medicine, with the aim of studying in depth the beneficial effects of these compounds in human beings. We hope that this new volume will attract the interest of all the scientific community, especially those working in the fields of pharmaceutical, medical, biological, chemical and food sciences

    A principled approach to measuring the IoT ecosystem

    Internet of Things (IoT) devices combine network connectivity, cheap hardware, and actuation to provide new ways to interface with the world. In spite of this growth, little work has been done to measure the network properties of IoT devices. Such measurements can help to inform systems designers and security researchers of IoT networking behavior in practice to guide future research. Unfortunately, properly measuring the IoT ecosystem is not trivial. Devices may have different capabilities and behaviors, which require both active measurements and passive observation to quantify. Furthermore, the IoT devices that are connected to the public Internet may vary from those connected inside home networks, requiring both an external and internal vantage point to draw measurements from. In this thesis, we demonstrate how IoT measurements drawn from a single vantage point or mesaurement technique lead to a biased view of the network services in the IoT ecosystem. To do this, we conduct several real-world IoT measurements, drawn from both inside and outside home networks using active and passive monitoring. First, we leverage active scanning and passive observation in understanding the Mirai botnet---chiefly, we report on the devices it infected, the command and control infrastructure behind the botnet, and how the malware evolved over time. We then conduct active measurements from inside 16M home networks spanning 83M devices from 11~geographic regions to survey the IoT devices installed around the world. We demonstrate how these measurements can uncover the device types that are most at risk and the vendors who manufacture the weakest devices. We compare our measurements with passive external observation by detecting compromised scanning behavior from smart homes. We find that while passive external observation can drive insight about compromised networks, it offers little by way of concrete device attribution. We next compare our results from active external scanning with active internal scanning and show how relying solely on external scanning for IoT measurements under-reports security important IoT protocols, potentially skewing the services investigated by the security community. Finally, we conduct passive measurements of 275~smart home networks to investigate IoT behavior. We find that IoT device behavior varies by type and devices regularly communicate over a myriad of bespoke ports, in many cases to speak standard protocols (e.g., HTTP). Finally, we observe that devices regularly offer active services (e.g., Telnet, rpcbind) that are rarely, if ever, used in actual communication, demonstrating the need for both active and passive measurements to properly compare device capabilities and behaviors. Our results highlight the need for a confluence of measurement perspectives to comprehensively understand IoT ecosystem. We conclude with recommendations for future measurements of IoT devices as well as directions for the systems and security community informed by our work

    Evaluating FAIR Digital Object and Linked Data as distributed object systems

    FAIR Digital Object (FDO) is an emerging concept that is highlighted by European Open Science Cloud (EOSC) as a potential candidate for building a ecosystem of machine-actionable research outputs. In this work we systematically evaluate FDO and its implementations as a global distributed object system, by using five different conceptual frameworks that cover interoperability, middleware, FAIR principles, EOSC requirements and FDO guidelines themself. We compare the FDO approach with established Linked Data practices and the existing Web architecture, and provide a brief history of the Semantic Web while discussing why these technologies may have been difficult to adopt for FDO purposes. We conclude with recommendations for both Linked Data and FDO communities to further their adaptation and alignment.Comment: 40 pages, submitted to PeerJ C

    Application Adaptive Bandwidth Management Using Real-Time Network Monitoring.

    Application adaptive bandwidth management is a strategy for ensuring secure and reliable network operation in the presence of undesirable applications competing for a network鈥檚 crucial bandwidth, covert channels of communication via non-standard traffic on well-known ports, and coordinated Denial of Service attacks. The study undertaken here explored the classification, analysis and management of the network traffic on the basis of ports and protocols used, type of applications, traffic direction and flow rates on the East Tennessee State University鈥檚 campus-wide network. Bandwidth measurements over a nine-month period indicated bandwidth abuse of less than 0.0001% of total network bandwidth. The conclusion suggests the use of the defense-in-depth approach in conjunction with the KHYATI (Knowledge, Host hardening, Yauld monitoring, Analysis, Tools and Implementation) paradigm to ensure effective information assurance

    Transcriptomic and proteomic analysis of lycopene-overproducing Escherichia coli strains

The certified thesis is available in the Institute Archives and Special Collections.Includes bibliographical references.Systems biology represents a powerful method to describe and manipulate phenotypes of interest by incorporating biological information from various levels of cellular organization. Such an approach is illustrated from a library of both rationally-directed and combinatorial gene knockout strains of E. coli recombinantly producing the small molecule lycopene. Global genomic and proteomic expression changes associated with increased lycopene production of mutant E. coli constructs were discovered using whole-genome DNA microarrays and a novel LC-MS technique, respectively. While most genes and proteins showed few expression changes, key differences were identified, including targets distal to the non-mevalonate and precursorsupplying pathways. Based upon the expression data sets, it was hypothesized that the following may be associated with lycopene overproduction: histidine biosynthesis (hisH); the quinone pool (wrbA); acid resistance (ydeO and gadE); the glyoxylate pathway (iclR); NADPH redox balance (pntB); growth rate reduction; and membrane composition. In the pre-engineered background strain, deleting pntB (~20-25%) and ydeO (~30%) each led to moderately increased production; overexpressing wrbA led to 50-100% more production at 8 hours and 5-15% more production at later time points; deleting iclR caused small production increases (~5-10%); and supplementing media with histidine caused the parental and mutant strains to have similar production.(cont.) From these observations, several themes emerged. First, reduced cellular growth and energy conservation appear to be important tradeoffs for increasing lycopene production. Second, reducing overflow metabolism to acetate and corresponding acid stress as well as providing a gluconeogenic flux to increase lycopene precursors appeared beneficial. Next, NADPH availability and balance seemed to be critical production factors. The sS factor is known to affect lycopene accumulation, and it was observed to have far-reaching effects on both the transcriptomic and proteomic data sets. While expression changes were not strictly additive between the five mutant strains examined in comparison to the pre-engineered background strain, a number of these common factors appear to be responsible for the high lycopeneproduction phenotype. This work serves as an important example of incorporating multiple layers of complementary biological information to define a basis for an observed phenotype, demonstrating a powerful paradigm for realizing production increases via systems metabolic engineering.by Brian E.
