Immunologie de l’œil

RésuméL’œil impose au système immunitaire des contraintes d’efficacité et d’innocuité particulièrement exigeantes et contradictoires. Interface avec le monde environnant, il constitue avec ses annexes un compartiment muqueux de l’organisme de petite dimension, particulièrement exposé, mais apparemment peu vulnérable grâce à une multiplicité de moyens de défense : flore microbienne proche de celle de la peau ; clignement des paupières, flux continu du film lacrymal et composition complexe et variable de ce dernier qui doit être agressif à l’égard des microorganismes, mais protecteur et nutritif pour la cornée et de plus optiquement favorable ; et enfin tissu lymphoïde associé aux muqueuses caractérisé là par la production abondante d’IgA1 et d’IgD et d’anticorps naturels. À la fois vitre et ultime barrière, la cornée possède tous les éléments nécessaires à une réponse efficace, mais par leur distribution ou leur état basal, ne les utilisera qu’en dernier recours. L’œil est en effet aussi un instrument d’optique prolongeant le système nerveux central ; les vaisseaux doivent donc être absents de l’axe optique et toute inflammation comporte le risque d’être délétère. Le « privilège immunitaire » résulte localement du contrôle étroit exercé sur les cellules lymphoïdes et réactions immunitaires par l’exclusion moléculaire due aux barrières hémato-oculaires, une ambiance moléculaire inhibitrice et diverses interactions cellulaires pouvant générer apoptose ou cellules régulatrices. De plus, ce contrôle peut s’accompagner d’une réponse systémique caractérisée par une diminution de l’hypersensibilité retardée, une facilitation de la production d’anticorps ne fixant pas le complément et la génération de cellules T régulatrices spécifiques d’antigènes, un phénomène nommé « déviation immunitaire associée à la chambre antérieure de l’œil ». Cependant, la relative inaccessibilité de l’œil sain au système immunitaire pourrait rendre les mécanismes de tolérance immunitaire périphérique moins efficaces d’où une vulnérabilité particulière aux états auto-immuns et des autoanticorps antirétine dans des syndromes paranéoplasiques. L’œil est ainsi armé pour réagir, mais sa réponse est étroitement contrôlée et orientée ; les associations entre pathologies et systèmes HLA ou du complément sont aussi des clefs pour mieux comprendre les mécanismes moléculaires à l’œuvre

Detection of HIV-1 infection in dried blood spots from a 12-year-old ABO bedside test card

Background and Objectives: We tested dried blood from an ABO bedside test card which had been stored at room temperature for 12 years, to prove that a patient with HIV-1 infection had been infected by blood transfusion. Materials and Methods: Immunoblots for HIV-1 antibodies and threefold PCRs with half-nested primers for the HIV-1 integrase gene were done with eluates from the dried blood spots. Results: HIV-1 antibodies and HIV-1 DNA could be detected in the sample from one unit of blood, but not from the two other units or from the recipient before transfusion. Conclusion: Further studies should be done on the validity of stored dried blood as an alternative to the storage of frozen donor serum for several years for `look-back' studies

Zusammenfassung: Die Infektion mit M.tuberculosis (Mtb) ist nach wie vor weit verbreitete, aber nur bei bestimmten Menschen wird aus der primären Infektion eine Erkrankung. Nur Patienten mit einer Immunschwäche oder einer reduzierten Immunität erkranken. Das sind pro Jahr weltweit ca. 8-10Mio. Menschen. Ein gutes Verständnis der Mtb-Immunität ist wichtig, wenn man Mtb verhindern, Immunmodulatoren für bestimmte Krankheiten einsetzen oder neue Impfstoffe auf der Grundlage des durch die Entschlüsselung der Genomstruktur von Mtb gewonnenen Wissens entwickeln wil

Requirement of Activation for Hepatitis B Virus Infection

Although _in vitro_ models of human hepatitis B virus replication are established, so far none could approximate infection efficiency as expected from _in vivo_ observations. Susceptibility for HBV infections has only been reported for primary hepatocytes of human, chimpanzee or Tupaia belangeri and the cell line HepaRG. Here we show that the insusceptible human hepatoma cell line HepG2 can be infected, when the virus was beforehand activated by passage over whole duck liver cell cultures. That suggests an activation step to be performed by specialized liver cells

Some characteristics of HIV, a retrovirus which is the causative agent of AIDS, and the immune system, are discussed in this paper to help one understand the mechanism and development of this devastating viral infection. The conclusion of this paper is that a suitable vaccine or drug, if and whenever it becomes available, is not the answer to this fatal disease. The only solution is to be found in educating the youth to implement Scriptural values in their life styles
