    Dynamic location problems with limited look-ahead

    Background Among the most frequently encountered mutations in dilated cardiomyopathy (DCM) are those in the lamin A/C (LMNA) gene. Our goal was to analyze the LMNA gene in patients with DCM and/or conduction disease referred to the cardiogenetics outpatient clinic and to evaluate the prevalence of LMNA mutations and their clinical expression. Methods and Results The LMNA gene was screened in 61 index patients. Eleven mutations (including 6 novel) were identified, mainly in the subgroup of familial DCM with cardiac conduction disease (3/10 index patients) and in patients with DCM and Emery-Dreifuss, Limb-Girdle, or unclassified forms of muscular dystrophy (7/8 index patients). In addition, a mutation was identified in 1 of 4 families with only cardiac conduction disease. We did not identify any large deletions or duplications.Genotype-phenotype relationships revealed a high rate of sudden death and cardiac transplants in carriers of the p.N 195K mutation. Our study confirmed that the p.R225X mutation leads to cardiac conduction disease with late or no development of DCM, underscoring the importance of this mutation in putative familial "lone conduction disease." Nearly one third of LMNA mutation carriers had experienced a thromboembolic event. Conclusions This study highlights the role of LMNA mutations in DCM and related disorders. A severe phenotype in p.N 195K mutation carriers and preferential cardiac conduction disease in p.R225X carriers was encountered.     Review of trends and targets of complex systems for power system optimization

    Optimization systems (OSs) allow operators of electrical power systems (PS) to optimally operate PSs and to also create optimal PS development plans. The inclusion of OSs in the PS is a big trend nowadays, and the demand for PS optimization tools and PS-OSs experts is growing. The aim of this review is to define the current dynamics and trends in PS optimization research and to present several papers that clearly and comprehensively describe PS OSs with characteristics corresponding to the identified current main trends in this research area. The current dynamics and trends of the research area were defined on the basis of the results of an analysis of the database of 255 PS-OS-presenting papers published from December 2015 to July 2019. Eleven main characteristics of the current PS OSs were identified. The results of the statistical analyses give four characteristics of PS OSs which are currently the most frequently presented in research papers: OSs for minimizing the price of electricity/OSs reducing PS operation costs, OSs for optimizing the operation of renewable energy sources, OSs for regulating the power consumption during the optimization process, and OSs for regulating the energy storage systems operation during the optimization process. Finally, individual identified characteristics of the current PS OSs are briefly described.     Interaction between intelligent agent strategies for real-time transportation planning

    In this paper we study the real-time scheduling of time-sensitive full truckload pickup-and-delivery jobs. The problem involves the allocation of jobs to a fixed set of vehicles which might belong to dfferent collaborating transportation agencies. A recently proposed solution methodology for this problem is the use of a multi-agent system where shipper agents other jobs through sequential auctions and vehicle agents bid on these jobs. In this paper we consider such a multi-agent system where both the vehicle agents and the shipper agents are using profit maximizing look-ahead strategies. Our main contribution is that we study the interrelation of these strategies and their impact on the system-wide logistical costs. From our simulation results, we conclude that the system-wide logistical costs (i) are always reduced by using the look-ahead policies instead of a myopic policy (10-20%) and (ii) the joint effect of two look-ahead policies is larger than the effect of an individual policy. To provide an indication of the savings that might be realized with a central solution methodology, we benchmark our results against an integer programming approach

    A dynamic day-ahead paratransit planning problem

    Abstract We consider a dynamic planning problem for the transport of elderly and disabled people. The focus is on a decision to make one day ahead: which requests to serve with own vehicles, and which ones to assign to subcontractors, under uncertainty of late requests which are gradually revealed during the day of operation. We call this problem the Dynamic Day-ahead Paratransit Planning problem. The developed model is a nonstandard two-stage recourse model in which ideas from stochastic programming and online optimization are combined: in the first stage clustered requests are assigned to vehicles, and in the dynamic second-stage problem an event-driven approach is used to cluster the late requests once they are revealed and subsequently assign them to vehicles. A genetic algorithm is used to solve the model. Computational results are presented for randomly generated data sets. Furthermore, a comparison is made to a similar problem we studied earlier in which the simplifying but unrealistic assumption has been made that all late requests are revealed at the beginning of the day of operation.

    Model Learning for Look-ahead Exploration in Continuous Control

    We propose an exploration method that incorporates look-ahead search over basic learnt skills and their dynamics, and use it for reinforcement learning (RL) of manipulation policies . Our skills are multi-goal policies learned in isolation in simpler environments using existing multigoal RL formulations, analogous to options or macroactions. Coarse skill dynamics, i.e., the state transition caused by a (complete) skill execution, are learnt and are unrolled forward during lookahead search. Policy search benefits from temporal abstraction during exploration, though itself operates over low-level primitive actions, and thus the resulting policies does not suffer from suboptimality and inflexibility caused by coarse skill chaining. We show that the proposed exploration strategy results in effective learning of complex manipulation policies faster than current state-of-the-art RL methods, and converges to better policies than methods that use options or parametrized skills as building blocks of the policy itself, as opposed to guiding exploration.     Active Sensing as Bayes-Optimal Sequential Decision Making

    Sensory inference under conditions of uncertainty is a major problem in both machine learning and computational neuroscience. An important but poorly understood aspect of sensory processing is the role of active sensing. Here, we present a Bayes-optimal inference and control framework for active sensing, C-DAC (Context-Dependent Active Controller). Unlike previously proposed algorithms that optimize abstract statistical objectives such as information maximization (Infomax) [Butko & Movellan, 2010] or one-step look-ahead accuracy [Najemnik & Geisler, 2005], our active sensing model directly minimizes a combination of behavioral costs, such as temporal delay, response error, and effort. We simulate these algorithms on a simple visual search task to illustrate scenarios in which context-sensitivity is particularly beneficial and optimization with respect to generic statistical objectives particularly inadequate. Motivated by the geometric properties of the C-DAC policy, we present both parametric and non-parametric approximations, which retain context-sensitivity while significantly reducing computational complexity. These approximations enable us to investigate the more complex problem involving peripheral vision, and we notice that the difference between C-DAC and statistical policies becomes even more evident in this scenario.Comment: Scheduled to appear in UAI 201

    Comparison of agent-based scheduling to look-ahead heuristics for real-time transportation problems

    We consider the real-time scheduling of full truckload transportation orders with time windows that arrive during schedule execution. Because a fast scheduling method is required, look-ahead heuristics are traditionally used to solve these kinds of problems. As an alternative, we introduce an agent-based approach where intelligent vehicle agents schedule their own routes. They interact with job agents, who strive for minimum transportation costs, using a Vickrey auction for each incoming order. This approach offers several advantages: it is fast, requires relatively little information and facilitates easy schedule adjustments in reaction to information updates. We compare the agent-based approach to more traditional hierarchical heuristics in an extensive simulation experiment. We find that a properly designed multiagent approach performs as good as or even better than traditional methods. Particularly, the multi-agent approach yields less empty miles and a more stable service level

    Design choices for agent-based control of AGVs in the dough making process

    In this paper we consider a multi-agent system (MAS) for the logistics control of Automatic Guided Vehicles (AGVs) that are used in the dough making process at an industrial bakery. Here, logistics control refers to constructing robust schedules for all transportation jobs. The paper discusses how alternative MAS designs can be developed and compared using cost, frequency of messages between agents, and computation time for evaluating control rules as performance indicators. Qualitative design guidelines turn out to be insufficient to select the best agent architecture. Therefore, we also use simulation to support decision making, where we use real-life data from the bakery to evaluate several alternative designs. We find that architectures in which line agents initiate allocation of transportation jobs, and AGV agents schedule multiple jobs in advance, perform best. We conclude by discussing the benefits of our MAS systems design approach for real-life applications
