Efeitos a posteriori dos défices hídricos sobre a actividade fotossintética: mecanismos de fotoprotecção e sistema enzimático antioxidante

posterior rehidratação foi investigada em plantas envasadas de Helianthus annuus L. e Lupinus
albus L. A rega foi manipulada de modo a induzir diferentes intensidades de défice hídrico e
posterior rehidratação. Nas plantas de H. annuus foram analisadas comparativamente folhas
jovens e folhas maduras, tendo-se também avaliado nas folhas jovens o efeito da diminuição
temporária da intensidade luminosa nas plantas em défice hídrico e rehidratação. Foram
também avaliados comparativamente os efeitos dos défices hídricos e da rehidratação na
actividade fotossintética e no sistema antioxidante em folhas de H. annuus e L. albus, espécies
com estratégias diferentes perante o défice hídrico (H. annuus é tolerante ao défice hídrico e L.
albus evita o défice hídrico). Os resultados obtidos indicam que, apesar de as taxas de
fotossíntese terem diminuído em resposta ao défice hídrico, não ocorreu fotoinibição crónica
nem foi induzida senescência foliar nas folhas de H. annuus amostradas. Nestas plantas, em
situação de défice hídrico, ocorreram limitações metabólicas à actividade fotossintética, que
nas folhas maduras podem ter incluído a activação da reacção de Mehler e da fotorrespiração.
As folhas jovens revelaram possuir a capacidade de reforçar a sua protecção antioxidante em
resposta ao défice hídrico, através do aumento do teor foliar em carotenóides. No entanto, as
diferenças registadas entre as folhas jovens e as folhas maduras nas plantas em défice hídrico
não se repercutiram na sua capacidade de recuperação da actividade fotossintética após
rehidratação, que foi semelhante. Os dados obtidos mostraram que o aumento do ‘quenching’
não fotoquímico nem sempre está directamente relacionado com o aumento do índice de
desepoxidação do ciclo das xantofilas e confirmaram a relação de dependência entre ambos os
parâmetros e a densidade de fluxo quântico. Nas plantas em défice hídrico transferidas para
intensidade luminosa baixa, a ausência de stresse oxidativo pode ter implicado falhas na
sinalização interna das plantas, enquanto o estímulo aparente da exportação de fotoassimilados
durante a rehidratação pode ter conduzido à inibição do crescimento, pelo que não se
observaram vantagens na imposição do défice hídrico e da rehidratação a baixas densidades de
fluxo quântico. A manutenção de altos teores foliares de sacarose nas plantas rehidratadas sob
irradiância alta não evitou que estas não recuperassem completamente a actividade
fotossintética. O défice hídrico induziu senescência foliar em L. albus. Esta pode ter resultado
do stresse oxidativo induzido nas plantas em défice hídrico, aparentemente devido ao
desequilíbrio entre as actividades da dismutase do superóxido e da peroxidase do ascorbato.
Os resultados mostram que o défice hídrico pode, por si só, induzir respostas normalmente
associadas à ocorrência conjunta de défice hídrico e intensidade luminosa alta, tais como o
stresse oxidativo e o aumento no teor foliar em pigmentos antioxidantes
The feedback regulation of miRNA biogenesis in responce to water deficit in Medicago truncatula
Tese de mestrado, Biologia (Biologia Celular e Biotecnologia), 2009, Universidade de Lisboa, Faculdade de CiênciasOs microARNs (miARNs) são pequenas moléculas de ARN com um tamanho de 20 a 24 nucleótidos (nt) e apresentam-se como reguladores negativos da expressão génica. As proteínas Dicer-like 1 (DCL1) e Argonaute 1 (AGO1) estão envolvidas na via de produção de miARNs e seus mARNs são regulados por miR162 e miR168 respectivamente. O principal objectivo deste trabalho foi analisar como se processa esta regulação em feed-back negativo na planta modelo Medicago truncatula sujeita a condições de défice hídrico. As plantas foram sujeitas a progressiva supressão da irrigação e de acordo com a avaliação do conteúdo hídrico relativo (CHR) das folhas, foram divididas em quatro grupos: controlo, défice hídrico moderado, défice hídrico severo e recuperação. Primeiro, foi analisada a expressão de 4 genes descritos por serem induzidos em resposta ao défice hídrico - Early Responsive to Dehydration 1 (ERD1), Plasma membrane intrinsic protein 1;5 (PIP1;5), Dehydrin e L-Myo-inositol-1-phosphate Synthase (MI-1-PS) - para comparar com os valores do CHR das plantas. Todos aumentaram a sua expressão na parte aérea e nas raízes em condições de défice hídrico, apresentando uma relação temporal e inversa com o CHR das plantas, à excepção do MI-1-PS na parte aérea. Em segundo, a expressão de miR162 reduziu e de DCL1 aumentou com o défice hídrico. Desta forma concluímos que DCL1 está a ser regulado nas raízes por miR162 em M. truncatula sujeita a condições de falta de água. Em ultimo, foram identificados em M. Truncatula, 3 genes codificantes para DCLs e 9 para AGOs envolvidos outras vias de processamento de pequenos ARNs. Observámos que têm os níveis de expressão aumentados em situação de défice hídrico. A MtAGO7 está envolvida na produção dos denominados trans-acting small RNAs e a MtDCL3, MtAGO4b e MtAGO4c, estão envolvidas em processos de metilação do DNA e consequente silenciamento transcripcional de genes.The microRNAs (miRNAs) are small RNAs with 20-24 nucleotides that negatively regulate the transcripts of several genes. Dicer-like 1 (DCL1) and Argonaute 1 (AGO1) are two key proteins involved in the miRNA biogenesis and their mRNAs are feedback regulated by miR162 and miR168 respectively. The main goal of this work was to study this mechanism of regulation of miRNAs biogenesis in Medicago truncatula under different water deficit conditions. Plants were subjected to progressive water deprivation and according to the evaluation of their water status by the leaf relative water content (RWC) were divided in four groups: control, moderate water deficit, severe water deficit and recovery. First, we analyzed the expression of 4 water deficit-responsive genes - Early Responsive to Dehydration 1 (ERD1), Plasma membrane intrinsic protein 1;5 (PIP1;5), L-Myo-inositol-1-phosphate Synthase (MI-1-PS) and Dehydrin - to establish a relation with the RWC of the plants. We found that the expression levels of ERD1, PIP1;5 and Dehydrin genes in plant shoots and roots and MI-1-PS in roots were temporally and inversely correlated with the plants RWC parameters, confirming that progressive water deficit induces an increase in transcription of water deficit-responsive genes. Second, we showed that miR162 was down-regulated in M. truncatula in water deficit whereas its target, DCL1, was up-regulated. This indicates that miR162 is regulating the mRNA levels of DCL1 in M. truncatula plants under water deficit. Third, in a bioinformatic screen we found and characterized the protein sequence of 3 and 9 new DCLs and AGOs in M. truncatula. The expression analysis of some of these genes under water deficit, showed up-regulation of genes encoding proteins involved in small RNA silencing pathways like direction of chromatin methylation for silencing of gene transcription (DCL3, AGO4b and AGO4c) or the production of trans-acting small interfering RNAs derived from TAS3 gene transcripts (AGO7)
Fotossíntese e acumulação de ácido abcísico em Lupinus albus sujeito a stress hídrico
The objective of the present work was to assess whether the depression of photosynthetic activity in
droughted Lupinus albus L. plants was more closely associated with carbohydrate build-up or ABA
accumulation. With that purpose we have measured the concomitant drought-induced changes in photosynthetic
capacity and the concentrations of Rubisco, chlorophylls, non-structural carbohydrates and ABA in young and
old leaves of white lupin plants. Although Rubisco and chlorophylls contents did not decrease with water stress,
the photosynthetic capacity was decreased by soil drying, the decline in photosynthesis being more tighly related
with ABA accumulation than with sugar content. In contrast to droughted plants, ABA feeding to intact plants
resulted in a significant decrease in Rubisco and chlorophyll content. However, no increase in shoot ABA
content was detected in ABA-fed plants. Therefore this work is not conclusive as to the correlation between
photosynthetic capacity and ABA accumulation in droughted white lupin plants being causal or not
Photosynthetic responses of Lupinus albus to soil water fluctuations
Lupinus albus plants were grown in 3 dm3 pots in a semi-controled greenhouse. Two water regimes were
imposed: water deficit (S, 47% of soil capacity), and control (T, 86% of soil capacity). Plants water status was
monitored through foliar relative water content (TRA) and pre-dawn (ypd) and midday water potential (ymd). Gas
exchanges, chlorophyll a fluorescence and photosynthetic capacity (Amax) were measured. Pigments and soluble
protein were quantified and antioxidant system enzymes activity was determined. Plants under water deficit were
rehydrated and the same measurements and sampling were done again after 48 h (R1) and 72 h (R2). Albeit water
availability has diminished from 86% to 47% of soil capacity, there weren’t any remarkable changes on S plants.
R1 plants which haven’t shown any alterations neither in Amax nor in stomatal conductance (gH2O), exhibited a
significant decrease on net photosynthesis (A), reaching negative values, along with a raise in non-photochemical
quenching (NPQ). Presumably this raise should be related with an increase in the xanthophyll cycle deepoxidation
state. The significantly increase in Superoxide dismutase (SOD) specific activity in R1 and R2
plants might be related with the activation of Mehler-peroxidase reaction which provokes lumen DpH raising.
This can provide photosynthesis protection through VAZ cycle and may justify the NPQ increase
