    A Class-E Inductive Powering Link with Backward Data Communications for Implantable Sensor Systems

    The design and implementation of a wireless power and data transfer system based on inductive coupling, having the potential to be used in numerous implantable bio-medical sensors and systems, is presented. The system consists of an external (primary) unit and an internal (secondary) unit. The external unit incorporates a high-efficiency switch-mode Class-E amplifier operating at ~200 kHz for driving the primary coil. The secondary unit consists of a parallel resonant coil followed by the power recovery circuitry. Means for backward data communication from the internal to the external unit over the same pair of coils has been realized using a simple FSK-based modulation scheme incorporated into the internal unit. FSK demodulation and associated filtering are integrated with the base inductive powering system. Prototype system test results indicate the inductive link efficiency can exceed 80% under optimum operating conditions with the overall power transfer efficiency of approximately 30%. The communication system is capable of transmitting up to 10kbps of data with the FSK carrier frequency (i.e., middle-frequency) being only 120kHz. Brain implants for paralysis treatments are increasingly providing sensory feedback via neural stimulation. Within the field of neuroscience, the perturbation of neuronal circuits wirelessly in untethered, freely-behaving animals is of particular importance. In implantable systems, power consumption is often the limiting factor in determining battery or power coil size, cost, and level of tissue heating, with stimulation circuitry typically dominating the power budget of the entire implant. Thus, there is strong motivation to improve the energy efficiency of implantable electrical stimulators. In this thesis, I present two examples of low-power tissue stimulators. The first type is a wireless, low-power neural stimulation system for use in freely behaving animals. The system consists of an external transmitter and a miniature, implantable wireless receiver-and-stimulator utilizing a custom integrated chip built in a standard 0.5 ptm CMOS process. Low power design permits 12 days of continuous experimentation from a 5 mAh battery, extended by an automatic sleep mode that reduces standby power consumption by 2.5x. To test this device, bipolar stimulating electrodes were implanted into the songbird motor nucleus HVC of zebra finches. Single-neuron recordings revealed that wireless stimulation of HVC led to a strong increase of spiking activity in its downstream target, the robust nucleus of the arcopallium (RA). When this device was used to deliver biphasic pulses of current randomly during singing, singing activity was prematurely terminated in all birds tested. The second stimulator I present is a novel, energy-efficient electrode stimulator with feedback current regulation. This stimulator uses inductive storage and recycling of energy based on a dynamic power supply to drive an electrode in an adiabatic fashion such that energy consumption is minimized. Since there are no explicit current sources or current limiters, wasteful energy dissipation across such elements is naturally avoided. The stimulator also utilizes a shunt current-sensor to monitor and regulate the current through the electrode via feedback, thus enabling flexible and safe stimulation. The dynamic power supply allows efficient transfer of energy both to and from the electrode, and is based on a DC-DC converter topology that is used in a bidirectional fashion. In an exemplary electrode implementation, I show how the stimulator combines the efficiency of voltage control and the safety and accuracy of current control in a single low-power integrated-circuit built in a standard 0.35 pm CMOS process. I also perform a theoretical analysis of the energy efficiency that is in accord with experimental measurements.     Wireless Transceivers for Implantable Microsystems.

    In this thesis, we present the first-ever fully integrated mm3 low-power biomedical transceiver with 1 meter of range that is powered by a mm2 thin-film battery. The transceiver is targeted for biomedical implants where size and energy constraints dictated by application make design challenging. Despite all the previous work in RFID tags, form factor of such radios is incompatible with mm3 biomedical implants. The proposed transceiver bridges this gap by providing a compact low-power solution that can run off small thin-film batteries and can be stacked with other system components in a 3D fashion. On the sensor-to-external side, we proposed a novel FSK architecture based on dual-resonator LC oscillators to mitigate unwanted overlap of two FSK tones’ phase noise spectrum. Due to inherent complexity of such systems, fourth order dual-resonator oscillators can exhibit instable operation. We mathematically modeled the instability and derive design conditions for stable oscillations. Through simulation and measurements, validity of derived models was confirmed. Together with other low-power system blocks, the transmitter was successfully implanted in live mouse and in-vivo measurements were performed to confirm successful transmission of vital signals through organic tissue. The integrated transmitter achieved a bit-error-rate of 10-6 at 10cm with 4.7nJ/bit energy consumption. On the external-to-sensor link, we proposed a new protocol to lower receiver peak power, which is highly limited due to small size of mm3 microsystem battery. In the proposed protocol, sending same data multiple times drastically relaxes jitter requirement on the sensor side at the cost of increased power consumption on the external side without increasing peak power radiated by the external unit. The receiver also uses a dual-coil LNA to improve range by 22% with only 11% area overhead. An asynchronous controller manages protocol timing and limits total monitoring current to 43nA. The fabricated receiver consumes 1.6nJ/bit at 40kbps while positioned 1m away from a 2W source.PhDElectrical EngineeringUniversity of Michigan, Horace H.     An implantable micro-system for neural prosthesis control and sensory feedback restoration in amputees

    In this work, the prototype of an electronic bi-directional interface between the Peripheral Nervous System (PNS) and a neuro-controlled hand prosthesis is presented. The system is composed of two Integrated Circuits (ICs): a standard CMOS device for neural recording and a High Voltage (HV) CMOS device for neural stimulation. The integrated circuits have been realized in two different 0.35μm CMOS processes available fromAustriaMicroSystem(AMS). The recoding IC incorporates 8 channels each including the analog front-end and the A/D conversion based on a sigma delta architecture. It has a total area of 16.8mm2 and exhibits an overall power consumption of 27.2mW. The neural stimulation IC is able to provide biphasic current pulses to stimulate 8 electrodes independently. A voltage booster generates a 17V voltage supply in order to guarantee the programmed stimulation current even in case of high impedances at the electrode-tissue interface in the order of tens of k­. The stimulation patterns, generated by a 5-bit current DAC, are programmable in terms of amplitude, frequency and pulse width. Due to the huge capacitors of the implemented voltage boosters, the stimulation IC has a wider area of 18.6mm2. In addition, a maximum power consumption of 29mW was measured. Successful in-vivo experiments with rats having a TIME electrode implanted in the sciatic nerve were carried out, showing the capability of recording neural signals in the tens of microvolts, with a global noise of 7μVrms , and to selectively elicit the tibial and plantarmuscles using different active sites of the electrode. In order to get a completely implantable interface, a biocompatible and biostable package was designed. It hosts the developed ICs with the minimal electronics required for their proper operation. The package consists of an alumina tube closed at both extremities by two ceramic caps hermetically sealed on it. Moreover, the two caps serve as substrate for the hermetic feedthroughs to enable the device powering and data exchange with the external digital controller implemented on a Field-Programmable Gate Array (FPGA) board. The package has an outer diameter of 7mm and a total length of 26mm. In addition, a humidity and temperature sensor was also included inside the package to allow future hermeticity and life-time estimation tests. Moreover, a wireless, wearable and non-invasive EEG recording system is proposed in order to improve the control over the artificial limb,by integrating the neural signals recorded from the PNS with those directly acquired from the brain. To first investigate the system requirements, a Component-Off-The-Shelf (COTS) device was designed. It includes a low-power 8- channel acquisition module and a Bluetooth (BT) transceiver to transmit the acquired data to a remote platform. It was designed with the aimof creating a cheap and user-friendly system that can be easily interfaced with the nowadays widely spread smartphones or tablets by means of a mobile-based application. The presented system, validated through in-vivo experiments, allows EEG signals recording at different sample rates and with a maximum bandwidth of 524Hz.     An optical telemetry system for wireless transmission of biomedical signals across the skin

The certified thesis is available in the Institute Archives and Special Collections.A technology base for optically-coupled systems was developed that permits in-vivo transmission of biomedical signals across the skin. By complete implantation of sensors and instrumentation electronics, problems with percutaneous connectors were eliminated. Optical power and signal transmission was accomplished with smaller and lighter implant structures than previously achieved with radio frequency (RF) coupling techniques. This is particularly valuable in the field of neuroprosthetics, because it may be possible to implant an optical telemeter directly on the surface of the brain to make mechanically stable connections to microelectrode arrays for neuroelectric recordings. Miniature optical power panels (2.5 mm x 2.5 mm) were developed from arrays of photodiodes. Infrared light of 880 nm wavelength was effective for delivering power across the skin. Panels composed of silicon photodiodes were 14% efficient at converting this light to electrical power, and GaAlAs panels were 41% efficient. Tissue heating experiments demonstrated the safety of optical power transmission. An LED was identified that was both electrically efficient (16%) and of appropriate wavelength (660 nm) for transmitting optical signals from the implant. Pulse period encoding was used for transmission of signals because it was robust and required less power than schemes with higher LED duty cycles. Specialized photodetector circuits were developed to receive pulse encoded data, and decoder circuits were built to reconstruct the transmitted signals. Two prototype single-channel neural waveform telemeters (approx. 10 Hz to 7 kHz bandwidth) were constructed and implanted in the visual cortex of rabbits. Both implants successfully transmitted neuroelectric signals. The first implant survived for four weeks before failing due to a flaw in the encapsulation, and the improved second prototype continues to function properly 28 months after implantation. Integrated circuits (ICs) were designed to record and transmit eight channels of neural waveforms. The first IC telemeter functioned properly, although the sensitivity was not as great as needed for the recording of neural waveforms. It required less than 50 mW of electrical power to operate. Efforts to improve this design introduced flaws in the next set of IC designs, so these problems were addressed, and a final set of designs was submitted for fabrication at the conclusion of this research project.by Bruce C.     Implantable Transducers for Neurokinesiological Research and Neural Prostheses

    The objective of this thesis was to develop a family of advanced electrical and mechanical interfaces to record activity of nerves and muscles during natural movements. These interfaces have applications in basic research and may eventually be refined for used in restoring voluntary control of movement in paralyzed persons. I) A muscle length gauge was designed that is based on piezoelectric crystals attached at the ends of a fluid filled extensible tubing. The in-vivo performance of these gauges was equal to previous length gauge designs. In addition, the ultrasound based design provided for the first time a direct muscle length calibration method. 2) An innovative nerve cuff closing technique was devised that does not reqmre suture closures. The new design uses interdigitated tubes to lock the opening and fix the lumen of a nerve cuff. The cuffs were tested in long-term mammalian implants and their performance matched or surpassed previous closure designs. The nerve cuff was further redesigned to include a more compliant cuff wall and wire electrodes. 3) Floating microelectrodes previously used for central nervous system recordings were adapted for chronic use in the peripheral nervous system. These electrodes proved disappointing in terms of signal quality and longevity. The reasons for failure are thought to be of both electrical and mechanical origin. 4) An innovative silicon micromachined peripheral single unit electrode was designed and tested. In the in-vivo tests, a limited number of recording sites successfully established short-term neural interfaces. However, the quality of the electrode performance, in terms of signal amplitude and ability to discriminate single unit potentials, was insufficient. 5) Using a finite difference model, a numerical simulation of static and dynamic electrical interactions between peripheral axons and microelectrode interfaces was derived. The model consisted of resistive and capacitive elements arranged in a 3-dimensional conductive universe (two spatial dimensions and time). Models of intrafascicular fine wire or silicon based electrodes were used to record simulated propagating action potentials. It was confirmed that electrode movement affected the recorded signal amplitude and that a dielectric layer on a silicon electrode accentuated the recorded potential field. A conducting back plane facing away from axon sources did not have a significant effect on the electrode recording properties. In conclusion, several novel implantable transducers were developed for use in neurokinesiological research. A numerical simulation of the axonal potentials recorded by intrafascicular electrodes helped interpret various shortcomings found in the in-vivo electrode performance. Although not attempted in the present thesis some of the developed technologies may have potential of transferring to clinical neural prostheses applications

    Enhancing selectivity of minimally invasive peripheral nerve interfaces using combined stimulation and high frequency block: from design to application

    The discovery of the excitable property of nerves was a fundamental step forward in our knowledge of the nervous system and our ability to interact with it. As the injection of charge into tissue can drive its artificial activation, devices have been conceived that can serve healthcare by substituting the input or output of the peripheral nervous system when damage or disease has rendered it inaccessible or its action pathological. Applications are far-ranging and transformational as can be attested by the success of neuroprosthetics such as the cochlear implant. However, the body’s immune response to invasive implants have prevented the use of more selective interfaces, leading to therapy side-effects and off-target activation. The inherent tradeoff between the selectivity and invasiveness of neural interfaces, and the consequences thereof, is still a defining problem for the field. More recently, continued research into how nervous tissue responds to stimulation has led to the discovery of High Frequency Alternating Current (HFAC) block as a stimulation method with inhibitory effects for nerve conduction. While leveraging the structure of the peripheral nervous system, this neuromodulation technique could be a key component in efforts to improve the selectivity-invasiveness tradeoff and provide more effective neuroprosthetic therapy while retaining the safety and reliability of minimally invasive neural interfaces. This thesis describes work investigating the use of HFAC block to improve the selectivity of peripheral nerve interfaces, towards applications such as bladder control or vagus nerve stimulation where selective peripheral nerve interfaces cannot be used, and yet there is an unmet need for more selectivity from stimulation-based therapy. An overview of the underlying neuroanatomy and electrophysiology of the peripheral nervous system combined with a review of existing electrode interfaces and electrochemistry will serve to inform the problem space. Original contributions are the design of a custom multi-channel stimulator able to combine conventional and high frequency stimulation, establishing a suitable experimental platform for ex-vivo electrophysiology of the rat sciatic nerve model for HFAC block, and exploratory experiments to determine the feasibility of using HFAC block in combination with conventional stimulation to enhance the selectivity of minimally-invasive peripheral nerve interfaces.Open Acces
