    Donor CD47 controls T cell alloresponses and is required for tolerance induction following hepatocyte allotransplantation

    CD47-deficient hepatocyte transplantation induces rapid innate immune cell activation and subsequent associated graft loss in syngeneic recipients. However, the role of donor CD47 in regulation of T-cell alloresponses is poorly understood. We addressed this question by assessing OVA-specific immune responses in mice following hepatocyte transplantation from CD47-competent or -deficient OVAtransgenic donors. Compared to sham-operated controls, intrasplenic transplantation of CD47-deficient OVA+ hepatocytes significantly accelerated rejection of OVA+ skin grafted 7 days after hepatocyte transplantation. In contrast, mice receiving CD47-competent OVA+ hepatocytes showed prolonged and even indefinite survival of OVA+ skin allografts. T cells from mice receiving CD47-deficient, but not CD47-competent, OVA+ hepatocytes showed significantly enhanced responses to OVA+ stimulators compared to sham-operated controls. In contrast to the production of tolerogenic cytokines (IL-4 and IL-10) in the recipients of CD47-competent hepatocytes, mice receiving CD47-deficient hepatocytes showed elevated production of IFN-γ and IL-1α. Moreover, significant expansion of myeloid-derived suppressor cells was detected in the recipients of CD47-competent hepatocytes, which was required for tolerance induction in these mice. Thus, donor CD47 plays an important role in the control of T-cell alloresponses and tolerance induction following hepatocyte transplantation. Our data also suggest that intrasplenic hepatocyte transplantation may provide a means to induce allograft tolerance

    Pepper leaf disease recognition based on enhanced lightweight convolutional neural networks

    Pepper leaf disease identification based on convolutional neural networks (CNNs) is one of the interesting research areas. However, most existing CNN-based pepper leaf disease detection models are suboptimal in terms of accuracy and computing performance. In particular, it is challenging to apply CNNs on embedded portable devices due to a large amount of computation and memory consumption for leaf disease recognition in large fields. Therefore, this paper introduces an enhanced lightweight model based on GoogLeNet architecture. The initial step involves compressing the Inception structure to reduce model parameters, leading to a remarkable enhancement in recognition speed. Furthermore, the network incorporates the spatial pyramid pooling structure to seamlessly integrate local and global features. Subsequently, the proposed improved model has been trained on the real dataset of 9183 images, containing 6 types of pepper diseases. The cross-validation results show that the model accuracy is 97.87%, which is 6% higher than that of GoogLeNet based on Inception-V1 and Inception-V3. The memory requirement of the model is only 10.3 MB, which is reduced by 52.31%-86.69%, comparing to GoogLeNet. We have also compared the model with the existing CNN-based models including AlexNet, ResNet-50 and MobileNet-V2. The result shows that the average inference time of the proposed model decreases by 61.49%, 41.78% and 23.81%, respectively. The results show that the proposed enhanced model can significantly improve performance in terms of accuracy and computing efficiency, which has potential to improve productivity in the pepper farming industry

    Knowledge atlas of antibody-drug conjugates on CiteSpace and clinical trial visualization analysis

    ObjectiveAntibody-drugs conjugates (ADCs) are novel drugs with highly targeted and tumor-killing abilities and developing rapidly. This study aimed to evaluate drug discovery and clinical trials of and explore the hotspots and frontiers from 2012 to 2022 using bibliometric methods.MethodsPublications on ADCs were retrieved between 2012 and 2022 from Web of Science (WoS) and analyzed with CiteSpace 6.1.R2 software for the time, region, journals, institutions, etc. Clinical trials were downloaded from clinical trial.org and visualized with Excel software.ResultsA total of 696 publications were obtained and 187 drug trials were retrieved. Since 2012, research on ADCs has increased year by year. Since 2020, ADC-related research has increased dramatically, with the number of relevant annual publications exceeding 100 for the first time. The United States is the most authoritative and superior country and region in the field of ADCs. The University of Texas MD Anderson Cancer Center is the most authoritative institution in this field. Research on ADCs includes two clinical trials and one review, which are the most influential references. Clinical trials of ADCs are currently focused on phase I and phase II. Comprehensive statistics and analysis of the published literature and clinical trials in the field of ADCs, have shown that the most studied drug is brentuximab vedotin (BV), the most popular target is human epidermal growth factor receptor 2 (HER2), and breast cancer may become the main trend and hotspot for ADCs indications in recent years.ConclusionAntibody-drug conjugates have become the focus of targeted therapies in the field of oncology. The innovation of technology and combination application strategy will become the main trend and hotspots in the future

    Mapping age- and sex-specific HIV prevalence in adults in sub-Saharan Africa, 2000-2018

    BACKGROUND: Human immunodeficiency virus and acquired immune deficiency syndrome (HIV/AIDS) is still among the leading causes of disease burden and mortality in sub-Saharan Africa (SSA), and the world is not on track to meet targets set for ending the epidemic by the Joint United Nations Programme on HIV/AIDS (UNAIDS) and the United Nations Sustainable Development Goals (SDGs). Precise HIV burden information is critical for effective geographic and epidemiological targeting of prevention and treatment interventions. Age- and sex-specific HIV prevalence estimates are widely available at the national level, and region-wide local estimates were recently published for adults overall. We add further dimensionality to previous analyses by estimating HIV prevalence at local scales, stratified into sex-specific 5-year age groups for adults ages 15-59 years across SSA. METHODS: We analyzed data from 91 seroprevalence surveys and sentinel surveillance among antenatal care clinic (ANC) attendees using model-based geostatistical methods to produce estimates of HIV prevalence across 43 countries in SSA, from years 2000 to 2018, at a 5 × 5-km resolution and presented among second administrative level (typically districts or counties) units. RESULTS: We found substantial variation in HIV prevalence across localities, ages, and sexes that have been masked in earlier analyses. Within-country variation in prevalence in 2018 was a median 3.5 times greater across ages and sexes, compared to for all adults combined. We note large within-district prevalence differences between age groups: for men, 50% of districts displayed at least a 14-fold difference between age groups with the highest and lowest prevalence, and at least a 9-fold difference for women. Prevalence trends also varied over time; between 2000 and 2018, 70% of all districts saw a reduction in prevalence greater than five percentage points in at least one sex and age group. Meanwhile, over 30% of all districts saw at least a five percentage point prevalence increase in one or more sex and age group. CONCLUSIONS: As the HIV epidemic persists and evolves in SSA, geographic and demographic shifts in prevention and treatment efforts are necessary. These estimates offer epidemiologically informative detail to better guide more targeted interventions, vital for combating HIV in SSA

    Lithofacies types and reservoirs of Paleogene fine-grained sedimentary rocks Dongying Sag, Bohai Bay Basin, China

    By applying such technologies as detailed description of cores, identification of thin sections, X-ray diffraction analysis, and scanning electron microscopy, this paper establishes the lithofacies division scheme of the fine-grained sedimentary rocks in the Paleogene of the Dongying Sag, confirms the reservoir space types and characteristics of the major lithofacies, and analyzes the micro-structure characteristics of their pores. The research results show that: (1) the pore structure of the organic-rich laminated limestone facies is very complex, with multi-scale aperture. The micro-fractures connect the pores, and increase the porosity and permeability of reservoirs; (2) the pore structure of the laminated calcareous fine-grained mixed sedimentary facies with moderate organic content has not apparent multi-scale feature, but with better mesoporous opening property, poorer macroporous opening property, and ordinary connectivity of pores and fractures; (3) the reservoir space types of the organic-rich interbedded limestone facies are various, with stronger multi-scale aperture, better pore connectivity, apparent communication between pores and fractures, and the intercrystal pores and fractures in calcite recrystallization are the most important part of the macro-pores; (4) the reservoir space types of the organic-poor massive calcareous fine-grained mixed sedimentary facies are relatively simple, with undeveloped fractures, and poorer pore connectivity. By combining oil and gas generation potential, oil and gas flow ability, oil and gas reservoir properties and fracturing properties etc., the organic-rich laminated limestone and the organic-rich limy laminated fine-grained mixed sedimentary rocks are predicted as the favorable lithofacies.
