    Molecular identification of original plants of Fritillariae cirrhosae bulbus, a Tradtional Chinese Cedicine (TCM) using plant DNA barcoding

    Background: DNA barcoding is a widely used tool that enables rapid and accurate identification of species based on standardized DNA regions.Materials and Methods: In this study, potential DNA barcodes, namely three plastid regions (rbcL, trnH-psbA and matK) and one nuclear ribosomal internal transcribed spacer (ITS) were adopted for species identification of original plants of Fritillariae Cirrhosae Bulbus.Results: The rbcL and trnH-psbA regions showed better success rate of PCR amplification and DNA sequencing, as well as superior discriminatory ability. On the contrary, ITS region did not possess effective genetic variation and matK was faced with low success rate of sequencing. Combination of multi-loci sequences could improve identification ability of DNA barcoding. The trnH-psbA + rbcL could discriminate 25% - 100% species based on the Blast, Tree-Building and Distance methods.Conclusion: The potential DNA barcodes could not completely solving species identification of botanic origins of Fritillariae Cirrhosae Bulbus.     Roles of circulating soluble interleukin (IL)-6 receptor and IL-6 receptor expression on CD4+ T cells in patients with chronic hepatitis B

    SummaryObjectivesThe objective of this study was to investigate the potential clinical roles of circulating soluble interleukin (IL)-6 receptor (sIL-6R) and IL-6R expression on CD4+ T cells (CD4+ IL-6R+ T cells) in chronic hepatitis B (CHB) patients.MethodsOne hundred and thirty-three subjects, including 72 CHB patients, 27 asymptomatic carriers, eight acute hepatitis B (AHB) patients, and 26 healthy donors were included in this study. Plasma IL-6 and sIL-6R levels were measured by enzyme-linked immunosorbent assay (ELISA); the frequency of CD4+ IL-6R+ T cells was detected by flow cytometry analysis.ResultsOur data showed a significant increase in plasma sIL-6R levels and the frequency of CD4+ IL-6R+ T cells in peripheral blood in CHB patients compared to asymptomatic carriers and healthy controls (both p<0.05). The elevated prevalence of CD4+ IL-6R+ T cells was positively associated with increased serum alanine aminotransferase levels in CHB patients (r = 0.316, p = 0.007), but was not correlated with serum hepatitis B virus (HBV) DNA load. Moreover, CHB patients with an HBV DNA load >1.0×106 copies/ml had a lower level of plasma sIL-6R than those with an HBV DNA load <1.0×106 copies/ml.ConclusionsCirculating sIL-6R and CD4+ IL-6R+ T cells were increased in CHB patients. Elevated plasma sIL-6R is probably associated with HBV elimination, and CD4+ IL-6R+ T cells in peripheral blood might contribute to the pathogenesis of liver injury in CHB patients

    Rab7 GTPase controls lipid metabolic signaling in myeloid-derived suppressor cells

    Lysosomal acid lipase (LAL) is a critical neutral lipid metabolic enzyme that regulates metabolic reprogramming in myeloid-derived suppressor cells (MDSCs) through over-activation of mammalian target of rapamycin (mTOR). Affymetrix GeneChip microarray analysis of MDSCs from LAL deficient mouse (lal-/-) revealed upregulation of Rab7 GTPase protein, which belongs to a superfamily of small-molecular-weight GTPase known to regulate intracellular membrane trafficking from early to late endosomes and lysosomes. Here, the physical protein-protein interaction between Rab7 GTPase and mTOR has been detected by co-immunoprecipitation in the cell extract of wild type HD1A and lal-/- MDSC-like HD1B myeloid cell lines. The GST pull down assay using the recombinant GST-Rab7 GTPase fusion protein showed that Rab7 GTPase interacts with the mTOR N-terminal heat repeat domain. Rab7 GTPase siRNA knocking down reversed the altered lysosome/mTOR distribution and expression levels in HD1B cells. Rab7 GTPase siRNA knocking down in isolated bone marrow lal-/- MDSCs or HD1B cells not only reduced over-activation of mTOR and its downstream effector S6, but also decreased glucose consumption, decreased ROS over-production, and increased healthy mitochondria by membrane potential measurement. Inhibition of Rab7 GTPase led to reduced lal-/- MDSCs differentiation from bone marrow Lin- progenitor cells, reduced lal-/- MDSCs trans-endothelial migration, and reversed lal-/- MDSCs suppression of T cell proliferation. Furthermore, inhibition of Rab7 GTPase reduced lal-/- MDSCs ability to stimulate tumor cell proliferation in vitro, tumor growth in vivo, and tumor invasion. Together, these results showed that Rab7 GTPase is critically involved in MDSCs homeostasis and pathogenic functions

    The First and Second Order Asymptotics of Covert Communication over AWGN Channels

    This paper investigates the asymptotics of the maximal throughput of communication over AWGN channels by nn channel uses under a covert constraint in terms of an upper bound δ\delta of Kullback-Leibler divergence (KL divergence). It is shown that the first and second order asymptotics of the maximal throughput are nδlog⁡e\sqrt{n\delta \log e} and (2)1/2(nδ)1/4(log⁡e)3/4⋅Q−1(ϵ)(2)^{1/2}(n\delta)^{1/4}(\log e)^{3/4}\cdot Q^{-1}(\epsilon), respectively. The technique we use in the achievability is quasi-ε\varepsilon-neighborhood notion from information geometry. We prove that if the generating distribution of the codebook is close to Dirac measure in the weak sense, then the corresponding output distribution at the adversary satisfies covert constraint in terms of most common divergences. This helps link the local differential geometry of the distribution of noise with covert constraint. For the converse, the optimality of Gaussian distribution for minimizing KL divergence under second order moment constraint is extended from dimension 11 to dimension nn. It helps to establish the upper bound on the average power of the code to satisfy the covert constraint, which further leads to the direct converse bound in terms of covert metric

    Fluent: Round-efficient Secure Aggregation for Private Federated Learning

    Federated learning (FL) facilitates collaborative training of machine learning models among a large number of clients while safeguarding the privacy of their local datasets. However, FL remains susceptible to vulnerabilities such as privacy inference and inversion attacks. Single-server secure aggregation schemes were proposed to address these threats. Nonetheless, they encounter practical constraints due to their round and communication complexities. This work introduces Fluent, a round and communication-efficient secure aggregation scheme for private FL. Fluent has several improvements compared to state-of-the-art solutions like Bell et al. (CCS 2020) and Ma et al. (SP 2023): (1) it eliminates frequent handshakes and secret sharing operations by efficiently reusing the shares across multiple training iterations without leaking any private information; (2) it accomplishes both the consistency check and gradient unmasking in one logical step, thereby reducing another round of communication. With these innovations, Fluent achieves the fewest communication rounds (i.e., two in the collection phase) in the malicious server setting, in contrast to at least three rounds in existing schemes. This significantly minimizes the latency for geographically distributed clients; (3) Fluent also introduces Fluent-Dynamic with a participant selection algorithm and an alternative secret sharing scheme. This can facilitate dynamic client joining and enhance the system flexibility and scalability. We implemented Fluent and compared it with existing solutions. Experimental results show that Fluent improves the computational cost by at least 75% and communication overhead by at least 25% for normal clients. Fluent also reduces the communication overhead for the server at the expense of a marginal increase in computational cost

    In vitro regeneration of ‘Feizixiao’ litchi (Litchi chinensis Sonn.)

    A simple efficient in vitro plant regeneration system was developed by indirect somatic embryogenesis of ‘Feizixiao’ litchi (Litchi chinensis Sonn.). Pollen in the anther of monocytes was used to induce callus. Two auxins (naphthalene acetic acid [NAA] and 2,4-dichloriphenoxyacetic acid [2,4-D]), and two cytokines (kinetin [KT] and 6-benzyladenine [BA]) were tested to explore their influence on callus induction. MS medium supplemented with 2.22 μM BA, 2.69 μM NAA, 13.57 μM 2,4-D, and 0.4 g/L LH (lactalbumin hydrolysate) showed the highest callus induction frequency. The callus obtained from anther was subcultured in MS medium containing 4.52 μM 2,4-D to obtain synchronized friable embryogenic callus. Different developmental stages of SEs were obtained from the callus on MS medium containing 6% (w/v) sucrose and different PGRs (plant growth regulators). On MS medium containing 6% (w/v) sucrose and supplemented with 0.54 μM NAA, 23.23 μM KT, 0.4 g/L LH, 0.56 μM inositol, and 10% (w/v) CW (coconut water), a higher number of SEs (globular, heart, torpedo and cotyledonary embryos) was achieved than on other media. Plantlets were established onto half-strength MS medium containing 1.44 μM GA3 (gibberellic acid) followed by successful acclimatization in the greenhouse. With flow cytometry and chromosome counting, ploidy analysis of regenerated plants revealed that the regenerated plantlets were all diploid. This study is the first report on somatic embryogenesis of ‘Feizixiao litchi’, providing an opportunity to improve the cultivar by biotechnology methods.Keywords: litchi (Litchi chinensis Sonn.), anther culture, callus, regeneration, somatic embryogenesi

    Melatonin Supplementation, a Strategy to Prevent Neurological Diseases through Maintaining Integrity of Blood Brain Barrier in Old People

    Blood brain barrier (BBB) plays a crucial role in maintaining homeostasis of microenvironment that is essential to neural function of the central nervous system (CNS). When facing various extrinsic or intrinsic stimuli, BBB is damaged which is an early event in pathogenesis of a variety of neurological diseases in old patients including acute and chronic cerebral ischemia, Alzheimer’s disease and etc. Treatments that could maintain the integrity of BBB may prevent neurological diseases following various stimuli. Old people often face a common stress of sepsis, during which lipopolysaccharide (LPS) is released into circulation and the integrity of BBB is damaged. Of note, there is a significant decrease of melatonin level in old people and animal. Melatonin has been shown to preserves BBB integrity and permeability via a variety of pathways: inhibition of matrix metalloproteinase-9 (MMP-9), inhibition of NADPH oxidase-2, and impact on silent information regulator 1 (SIRT1) and nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome. More important, a recent study showed that melatonin supplementation alleviates LPS-induced BBB damage in old mice through activating AMP-activated protein kinase (AMPK) and inhibiting gp91phox, suggesting that melatonin supplementation may help prevent neurological diseases through maintaining the integrity of BBB in old people

    Elevated IL-6 Receptor Expression on CD4+ T Cells contributes to the increased Th17 Responses in patients with Chronic Hepatitis B

    <p>Abstract</p> <p>Background</p> <p>Increased numbers of Interleukin-17-producing CD4<sup>+ </sup>T cells (Th17) have been found in association with hepatitis B virus (HBV)-induced liver injury. However, the mechanism underlying the increase of Th17 responses in patients with HBV infection remains unclear. In this study, we investigate the possible regulatory mechanisms of increased Th17 responses in patients with chronic hepatitis B(CHB).</p> <p>Methods</p> <p>Th17 response and IL-6R expression on CD4<sup>+ </sup>T cells in peripheral blood samples were determined by flow cytometry. Cytokines TGF-β, IL-1β, IL-6 and IL-17 in plasma and/or supernatant samples were determined by ELISA and the IL-17 and IL-6R mRNA levels were quantified by quantitative real-time reverse polymerase chain reaction.</p> <p>Results</p> <p>All these data indicated that the frequency of periphery Th17 cells is significantly correlated with the percentage of CD4<b><sup>+ </sup></b>T cells expressing IL-6R in CHB patients. CD4<sup>+ </sup>T cells from patients with CHB, but not those from healthy donors, produced higher levels of IL-17 and had more IL-6R expression upon stimulation with the HBV core antigen (HBcAg) in vitro. The PMA/ionomycin and HBcAg -stimulated up-regulation of IL-17 production by CD4<sup>+ </sup>T cells could be reversed by a neutralizing antibody against IL-6R.</p> <p>Conclusion</p> <p>we showed that enhancement of IL-6R expression on CD4<sup>+ </sup>T cells upon HBV infection contributes to increased Th17 response in patients with CHB.</p
