    Werkzeug zur Entscheidungsunterstützung von KMU bei der Planung von Netzwerken zur Erbringung industrieller Dienstleistungen

    Für Investitionsgüterhersteller ist das Geschäft mit industriellen Dienstleistungen eng mit dem Produktgeschäft verbunden: Die Summe der verkauften und im Betrieb befindlichen Produkte – die „installierte Basis“ – stellt die Grundlage für den Absatz produktbegleitender industrieller Dienstleistungen dar. Mit einem Exportanteil von etwa 70 % agieren deutsche Investitionsgüterhersteller stark international, d. h. die installierte Basis ist weltweit verteilt (vgl. VDMA 2004). Um einen möglichst hohen Anteil dieser installierten Basis mit Dienstleistungen zu bedienen, muss das Dienstleistungsgeschäft ebenfalls international erfolgen. Gleichzeitig wird der Anteil des Dienstleistungsgeschäfts am Gesamterfolg der Unternehmen wichtiger. Eine Konzentration auf heimische Märkte, die nur einen geringen Anteil der installierten Basis darstellen, würde also auch ein entsprechend begrenztes Ertragspotenzial bedeuten. Allerdings können weltweite Serviceeinsätze wie z. B. Montage, Störfallbehebung oder Instandhaltung mit sehr hohen Kosten verbunden sein und so die Rendite schmälern. Für die Anbieter industrieller Dienstleistungen stellt sich daher die Frage nach einer geeigneten Form der Internationalisierung. Während große Unternehmen und Konzerne häufig Direktinvestitionen tätigen, benötigen insbesondere kleine und mittlere Unternehmen (KMU) Lösungen mit geringerem unternehmerischen Risiko. Hier bietet sich die Kooperation im Netzwerk an. Diese Lösung erfordert im Vergleich zur Direktinvestition weniger vertragliche Bindungen und geringere Investitionen, gleichzeitig bietet sie jedoch die Chance, von der Erfahrung und den Kenntnissen der Partner über die lokalen Märkte zu profitieren und Ressourcen zusammenfassen zu können (vgl. KUTSCHKER, MÖßLANG 1996; GULATI ET AL. 2000; WARNECKE 2002; ZAHN, STANIK 2003)

    Anti-VEGF Therapie unter Entzündungsparametern: Auswirkungen auf Zellvitalität und Zytokinsekretion im retinalen Pigmentepithel

    Die altersabhängige Makuladegeneration ist die häufigste Ursache für Erblindung in den westlichen Industrieländern beim älteren Menschen. Durch degenerative Veränderungen, genetische Prädispositionen, Umwelteinflüsse und Entzündungsreaktionen kommt es einerseits zu einem Untergang des Retinalen Pigmentepithels (RPE), was eine Degeneration der Photorezeptoren in der Retina zur Folge hat. Andererseits kommt es im Rahmen der feuchten AMD zu choroidalen Neovaskularisationen (CNV). Dies zeigt sich klinisch in einer zentralen Sehverschlechterung und führt unbehandelt zur Erblindung. Die medikamentöse Therapie der CNV besteht in der intravitrealen Injektion der Vascular Endothelial Growth Factor (VEGF) -Inhibitoren Aflibercept und Bevacizumab. Um die Assoziation der Pathogenese der AMD mit Entzündungsreaktionen des RPE näher zu untersuchen, wurden in dieser in vitro Studie die Effekte einer Langzeitstimulation mit dem Toll-like-Rezeptor (TLR)-3 Agonisten Polyinosin-Polycytidylsäure (Poly I:C), dem TLR-4 Agonisten Lipopolysaccharid (LPS) und dem proinflammatorischen Zytokin Tumornekrosefaktor alpha (TNFα) auf die Vitalität und die Sekretion der Zytokine IL-1β, IL-6, IL-8 und TNFα primär porciner RPE Zellen untersucht. Zusätzlich wurde überprüft welchen Einfluss die Anti-VEGF Therapie mit Aflibercept oder Bevacizumab allein und in Kombination mit den Entzündungsmediatoren auf die Vitalität und die Zytokinsekretion des RPE hat. Für die in dieser Studie getesteten proinflammatorischen Substanzen konnte grundsätzlich gezeigt werden, dass jede der drei Substanzen je nach Behandlungszeitraum und konzentration toxisch für das RPE sein kann. TNFα und LPS verminderten nach einer kurzzeitigen (24 Stunden) und mittelfristigen (sieben Tage) Stimulation das Zellüberleben signifikant. Poly I:C verringerte stattdessen das Zellüberleben nach Mittel- und Langzeit-stimulation (vier Wochen). Die Stimulation mit Aflibercept und Bevacizumab führte zu allen Zeitpunkten zu einer signifikanten Abnahme des Zellüberlebens. Die Kombination von Aflibercept mit Poly I:C, LPS oder TNFα führte unter langfristiger Stimulation zu einem signifikant verminderten Zellüberleben gegenüber der Kontrolle und gegenüber einer Stimulation mit der jeweiligen proinflammatorischen Substanz und Aflibercept allein. Somit scheint die langfristige Kombination der Aflibercepttherapie mit einer viralen oder bakteriellen Infektion (TLR3/4 Aktivierung über Poly I:C/LPS), einer degenerativen Gewebsverletzung (TLR-3/-4 Aktivierung über nekrotische Zellbestandteile) oder einer immunologischen Inflammation (TNFα), das Zellüberleben des RPE zu reduzieren. In Bezug auf die Zytokinsekretion konnte eine differenzierte proinflammatorische Antwort des RPE beobachtet werden, die langfristig hauptsächlich von IL-8 und IL-6 und weniger von IL-1β und TNFα geprägt wird und unterschiedliche Effekte je nach Stimulus zeigt. In Bezug auf die Kombination von Aflibercept und der mittleren Stimulationsdosis TNFα konnte über vier Wochen eine signifikante Erhöhung der IL-8 Sekretion im Vergleich zu einer Stimulation mit TNFα oder Aflibercept allein festgestellt werden. Unter noch genauer zu prüfenden Voraussetzungen könnte daher eine antiinflammatorische Therapie als unterstützende Behandlung unter Anti-VEGF Therapie in der Behandlung der feuchten AMD von Interesse sein

    Increased HRD score in cisplatin resistant penile cancer cells

    Penile cancer is a rare disease in demand for new therapeutic options. Frequently used combination chemotherapy with 5 fluorouracil (5-FU) and cisplatin (CDDP) in patients with metastatic penile cancer mostly results in the development of acquired drug resistance. Availability of cell culture models with acquired resistance against standard therapy could help to understand molecular mechanisms underlying chemotherapy resistance and to identify candidate treatments for an efficient second line therapy. We generated a cell line from a humanpapilloma virus (HPV) negative penile squamous cell carcinoma (UKF-PEC-1). This cell line was subject to chronic exposure to chemotherapy with CDDP and / or 5-FU to induce acquired resistance in the newly established chemo-resistant sublines (PEC-1 CDDP , adapted to 2500 ng/ml CDDP; UKF-PEC-1 5-FU , adapted to 500 ng/ml 5- FU; UKF-PEC1 CDDP / 5-FU , adapted to 2500 ng/ml CDDP and 500 ng/ml 5 -FU). Afterwards cell line pellets were formalin-fixed, paraffin embedded and subject to sequencing as well as testing for homologous recombination deficiency (HRD). Additionally, exemplary immunohistochemical stainings for p53 and gammaH2AX were applied for verification purposes. Finally, UKF-PEC-1 CDDP , UKF-PEC-1 5-FU , UKF-PEC1 CDDP / 5-FU , and UKF-PEC-3 (an alternative penis cancer cell line) were tested for sensitivity to paclitaxel, docetaxel, olaparib, and rucaparib. The chemo-resistant sublines differed in their mutational landscapes. UKF-PEC-1 CDDP was characterized by an increased HRD score, which is supposed to be associated with increased PARP inhibitor and immune checkpoint inhibitor sensitivity in cancer. However, UKF-PEC-1 CDDP did not display sensitivity to PARP inhibitors. [Abstract copyright: © 2022. The Author(s).

    Pharmacological targeting of host chaperones protects from pertussis toxin in vitro and in vivo

    Whooping cough is caused by Bordetella pertussis that releases pertussis toxin (PT) which comprises enzyme A-subunit PTS1 and binding/transport B-subunit. After receptor-mediated endocytosis, PT reaches the endoplasmic reticulum from where unfolded PTS1 is transported to the cytosol. PTS1 ADP-ribosylates G-protein alpha -subunits resulting in increased cAMP signaling. Here, a role of target cell chaperones Hsp90, Hsp70, cyclophilins and FK506-binding proteins for cytosolic PTS1-uptake is demonstrated. PTS1 specifically and directly interacts with chaperones in vitro and in cells. Specific pharmacological chaperone inhibition protects CHO-K1, human primary airway basal cells and a fully differentiated airway epithelium from PT-intoxication by reducing intracellular PTS1-amounts without affecting cell binding or enzyme activity. PT is internalized by human airway epithelium secretory but not ciliated cells and leads to increase of apical surface liquid. Cyclophilin-inhibitors reduced leukocytosis in infant mouse model of pertussis, indicating their promising potential for developing novel therapeutic strategies against whooping cough

    Influence of female sex and fertile age on neuromyelitis optica spectrum disorders

    Background: Gender and age at onset are important epidemiological factors influencing prevalence, clinical presentation, and treatment response in autoimmune diseases. Objective: To evaluate the impact of female sex and fertile age on aquaporin-4-antibody (AQP4-ab) status, attack localization, and response to attack treatment in patients with neuromyelitis optica (NMO) and its spectrum disorders (neuromyelitis optica spectrum disorder (NMOSD)). Methods: Female-to-male ratios, diagnosis at last visit (NMO vs NMOSD), attack localization, attack treatment, and outcome were compared according to sex and age at disease or attack onset. Results: A total of 186 NMO/SD patients (82% female) were included. In AQP4-ab-positive patients, female predominance was most pronounced during fertile age (female-to-male ratio 23:1). Female patients were more likely to be positive for AQP4-abs (92% vs 55%;p40years. Conclusion: Our data suggest an influence of sex and age on susceptibility to AQP4-ab-positive NMO/SD. Genetic and hormonal factors might contribute to pathophysiology of NMO/SD
