    The effect of post-exercise drink macronutrient content on appetite and energy intake

    Carbohydrate and protein ingestion post-exercise are known to facilitate muscle glycogen resynthesis and protein synthesis, respectively, but the effects of post-exercise nutrient intake on subsequent appetite are unknown. This study aimed to investigate whether protein induced satiety that has been reported at rest was still evident when pre-loads were consumed in a post-exercise context. Using a randomized, double blind, crossover design, 12 unrestrained healthy males completed 30 min of continuous cycling exercise at ~60% VO2peak, followed by five, 3 min intervals at ~85% VO2peak. Ten min post-exercise, subjects consumed 500 ml of either a low energy placebo (15 kJ) (PLA); a 6% whey protein isolate drink (528 kJ) (PRO); or a 6% sucrose drink (528 kJ) (CHO). Sixty min after drink ingestion, a homogenous ad-libitum pasta lunch was provided and energy intake at this lunch was quantified. Subjective appetite ratings were measured at various stages of the protocol. Energy consumed at the ad-libitum lunch was lower after PRO (5831 ± 960 kJ) than PLA (6406 ± 492 kJ) (P0.315). Considering the post-exercise drink, total energy intake was not different between trials (P=0.383). There were no differences between trials for any of the subjective appetite ratings. The results demonstrate that where post-exercise liquid protein ingestion may enhance the adaptive response of skeletal muscle, and this may be possible without affecting gross energy intake relative to consuming a low energy drink

    Organic compound-mineral interactions: using flash pyrolysis to monitor the adsorption of fatty acids on calcite

    Fatty acids are near ubiquitous organic compounds in living organisms in the Earth’s biosphere. Following death of an organism in the marine environment its fatty acids may survive descent to the sea bed where they can be juxtaposed with minerals. The aim of this study was to investigate the interaction of fatty acids with the common marine mineral calcite. Adsorption of tetradecanoic acid (C14) on calcite results in a sigmoidal or “s” isotherm. Flash pyrolysis experiments were conducted on samples of fatty acid adsorbed onto calcite and were compared with similar experiments on pure fatty acid and on salts of a fatty acid. Flash pyrolysis of pure tetradecanoic acid generated unsaturated and saturated hydrocarbons and a series of unsaturated and saturated low molecular weight fatty acids. Flash pyrolysis of free tetradecanoic acid salt produced saturated and unsaturated hydrocarbons, an aldehyde and a homologous series of saturated and unsaturated ketones, one of which was a symmetrical mid chain ketone (14-heptacosanone). Flash pyrolysis data from adsorbed tetradecanoic acid samples suggested that adsorption is analogous to the formation of the calcium salt of tetradecanoic acid. A key characteristic of the flash pyrolysis products of adsorbed fatty acids and fatty acid salts was the production of ketones with higher molecular weights than the starting fatty acids. Ketonisation was not observed from the flash pyrolysis of pure acid which implied the catalytic significance of the calcite mineral surface. The abundance of hydrocarbons relative to ketones in the pyrolysates negatively correlated with the proportion of fatty acids adsorbed to the surface of calcite. The ability to use flash pyrolysis to diagnose the nature of fatty acid interactions with mineral surfaces provides a valuable tool for monitoring the fate of these important lipids at the Earth’s surface as they pass into the geosphere and are subjected to diagenetic processes

    Single-cell lineage tracing in the mammary gland reveals stochastic clonal dispersion of stem/progenitor cell progeny.

    The mammary gland undergoes cycles of growth and regeneration throughout reproductive life, a process that requires mammary stem cells (MaSCs). Whilst recent genetic fate-mapping studies using lineage-specific promoters have provided valuable insights into the mammary epithelial hierarchy, the true differentiation potential of adult MaSCs remains unclear. To address this, herein we utilize a stochastic genetic-labelling strategy to indelibly mark a single cell and its progeny in situ, combined with tissue clearing and 3D imaging. Using this approach, clones arising from a single parent cell could be visualized in their entirety. We reveal that clonal progeny contribute exclusively to either luminal or basal lineages and are distributed sporadically to branching ducts or alveoli. Quantitative analyses suggest that pools of unipotent stem/progenitor cells contribute to adult mammary gland development. Our results highlight the utility of tracing a single cell and reveal that progeny of a single proliferative MaSC/progenitor are dispersed throughout the epithelium.This work was supported by a grant from the Medical Research Council programme grant no.     Teaching, research or balanced? An exploration of the experiences of biomedical scientists working in UK medical schools

    Driven by demand for high standards in university education, efforts have been made in the UK to address the perceived imbalance between teaching and research. However, teaching is still perceived by many as having less credibility and is attributed less importance. The purpose of our research was to explore how distinct types of academic job profiles (‘research’ or ‘education’ focused, or ‘balanced’) impact on biomedical scientists' perceptions of the lecturer role. Specifically, we investigated the experiences of biomedical scientists in ‘post-1990’ medical schools, which are known for their commitment to excellence in both research and education. We conducted 22 face-to-face, semi-structured interviews with biomedical scientists in five schools. Focusing on experiences of work, the interviews covered: ‘motivations’, ‘role expectations’, ‘teaching’, ‘research’ and ‘career’. The recorded qualitative data were transcribed and then analysed thematically. Our results, offering an insight into the working lives of biomedical scientists in medical education, suggest that in settings with a dual emphasis on education and research, individuals on ‘balanced’ contracts can experience a strong pull between research and teaching. In addition to posing significant challenges with respect to workload management, this can impact profoundly on professional identity. In contrast to the balanced role, ‘research’ or ‘education’ focused roles appear to have clearer requirements, leading to higher employee satisfaction. We conclude that to assist the educational mission of Higher Education, attention should be paid to balanced contracts, to (a) ensure employee support, (b) mitigate against negative perceptions of teaching, and ultimately, (c) guard against staff attrition

    Article image contrast, image pre-processing, and T₁ mapping affect MRI radiomic feature repeatability in patients with colorectal cancer liver metastases

    Imaging biomarkers require technical, biological, and clinical validation to be translated into robust tools in research or clinical settings. This study contributes to the technical validation of radiomic features from magnetic resonance imaging (MRI) by evaluating the repeatability of features from four MR sequences: pre-contrast T_{1}- and T_{2}-weighted images, pre-contrast quantitative T_{1} maps (qT_{1}), and contrast-enhanced T_{1} weighted images. Fifty-one patients with colorectal cancer liver metastases were scanned twice, up to 7 days apart. Repeatability was quantified using the intraclass correlation coefficient (ICC) and repeatability coefficient (RC), and the impact of non-Gaussian feature distributions and image normalisation was evaluated. Most radiomic features had non-Gaussian distributions, but Box–Cox transformations enabled ICCs and RCs to be calculated appropriately for an average of 97% of features across sequences. ICCs ranged from 0.30 to 0.99, with volume and other shape features tending to be most repeatable; volume ICC > 0.98 for all sequences. 19% of features from non-normalised images exhibited significantly different ICCs in pair-wise sequence comparisons. Normalisation tended to increase ICCs for pre-contrast T_{1}- and T_{2}-weighted images, and decrease ICCs for qT_{1} maps. RCs tended to vary more between sequences than ICCs, showing that evaluations of feature performance depend on the chosen metric. This work suggests that feature-specific repeatability, from specific combinations of MR sequence and pre-processing steps, should be evaluated to select robust radiomic features as biomarkers in specific studies. In addition, as different repeatability metrics can provide different insights into a specific feature, consideration of the appropriate metric should be taken in a study-specific context

    Methodological bias in cluster randomised trials

    Background: Cluster randomised trials can be susceptible to a range of methodological problems. These problems are not commonly recognised by many researchers. In this paper we discuss the issues that can lead to bias in cluster trials. Methods: We used a sample of cluster randomised trials from a recent review and from a systematic review of hip protectors. We compared the mean age of participants between intervention groups in a sample of 'good' cluster trials with a sample of potentially biased trials. We also compared the effect sizes, in a funnel plot, between hip protector trials that used individual randomisation compared with those that used cluster randomisation. Results: There is a tendency for cluster trials, with evidence methodological biases, to also show an age imbalance between treatment groups. In a funnel plot we show that all cluster trials show a large positive effect of hip protectors whilst individually randomised trials show a range of positive and negative effects, suggesting that cluster trials may be producing a biased estimate of effect. Conclusion: Methodological biases in the design and execution of cluster randomised trials is frequent. Some of these biases associated with the use of cluster designs can be avoided through careful attention to the design of cluster trials. Firstly, if possible, individual allocation should be used. Secondly, if cluster allocation is required, then ideally participants should be identified before random allocation of the clusters. Third, if prior identification is not possible, then an independent recruiter should be used to recruit participants

    Optimising NDWI supraglacial pond classification on Himalayan debris-covered glaciers

    The ability of medium-resolution (10–30 m) satellite imagery to delineate the size and persistence of ponds on debris-covered glaciers is a topic of recent interest as it has become apparent through the use of fine-resolution products that smaller ponds have often been neglected. In this study, we performed a quantitative analysis of pond detection using a normalised difference water index (NDWI) applied to several widely used satellite sensors, which offer multispectral information at high radiometric precision. These data include: RapidEye (5 m spatial resolution), Sentinel-2 Multispectral Instrument (MSI) (10–20 m), and Landsat 8 Operational Land Imager (OLI) multispectral imagery (30 m). We demonstrate a method to derive an optimum NDWI value for pond classification using a subset reference dataset of 285 ponds classified using fine-resolution (0.5 m) imagery. We then applied the optimised NDWI (NDWI-O) to the remaining images to assess pond classification accuracy against a broader reference dataset of 898 ponds. NDWI values calculated using Sentinel-2 imagery showed the best spectral contrast between water and surrounding debris cover, and the strongest relationship with pixel water content (R2 = 0.56), followed by the RapidEye NDWI (R2 = 0.45). We conclude that RapidEye and Sentinel-2 imagery is best suited for accurate pond classification using a multispectral classification approach, which is important for quantifying their role in glacier ablation, meltwater regulation, and lake development. By comparison, the impact of using coarse-resolution Landsat 8 imagery to characterise surface water dynamics is minimised when applied to large glacier lakes, where the area-to-perimeter ratio is greater
