    Preparation of Blood-Deficient Model and Research of Angelica Polysaccharide on Enriching Blood in Chickens

    In this study cyclophosphamide was used to prepare the blood-deficient model. The red blood cell count and hemoglobin content were measured. The experimental chickens presented the symptoms of blood-deficient syndrome, dullness, shrinkinginto oneself, broken winded, loose feather, waxy eyelid, and pale tongue. At the same time, red blood cell count and hemoglobin content decreased significantly. Angelica polysaccharide as the effective component of Angelica Sinensis could significantly increase the red blood cell count and the hemoglobin content of blood-deficient chickens. The results indicated that cyclophosphamide could significantly reduce the red blood count and hemoglobin content, and make the ideal blood-deficient model successfully. Angelica polysaccharide had the function of enriching blood in different ways. On the one hand Angelica polysaccharide enriched he blood directly, increased the number of RBC and hemoglobin; on the other hand it regulated the hematopoietic factors, enriched the blood indirectly

    Quercetin and Bornyl Acetate Regulate T-Lymphocyte Subsets and INF-γ/IL-4 Ratio In Utero in Pregnant Mice

    The objective of this study is to investigate the antiabortive effects of Quercetin and Bornvl Acetate and their immunological modulation at maternal-fetal interface. Lipopolysaccharide (LPS) was injected via tail vein to induce abortion in mice which received Quercetin and Bornvl Acetate at days 4–7 of gestation. Uterine CD4+/CD8+ T lymphocytes and IFN-γ/IL-4 of each group (n = 10) were detected by immunohistochemistry and enzyme-linked immunosorbent assay, respectively. The ratio of CD4+/CD8+ increased significantly (P < .01) in the uterus of LPS-induced abortion mice. In the Quercetin and Bornvl Acetate pretreated mice followed by LPS administration, the ratio of CD4+/CD8+ dropped to 0.562 ± 0.021, lower than that of LPS-abortion group (P < .01). The mean value of IFN-γ/IL-4 in LPS-treated mice was 0.310 ± 0.066, higher than that of Quercetin and Bornyl Acetate group. The results indicate that Quercetin and Bornyl Acetate have an antiabortive effect through modulation of immunological balance at maternal-fetal interface

    IL28B is associated with outcomes of chronic HBV infection

    Purpose The role of IL28B gene variants and expression in hepatitis B virus (HBV) infections are not well understood. Here, we evaluated whether IL28B gene expression and rs12979860 variations are associated with HBV outcomes. Materials and Methods IL28B genetic variations (rs12979860) were genotyped by pyrosequencing of DNA samples from 137 individuals with chronic HBV infection [50 inactive carriers (IC), 34 chronic hepatitis B (CHB), 27 cirrhosis, 26 hepatocellular carcinoma (HCC)], and 19 healthy controls. IL28A/B mRNA expression in peripheral blood mononuclear cells was determined by qRT-PCR, and serum IL28B protein was measured by ELISA. Results Patients with IL28B C/C genotype had greater IL28A/B mRNA expression and higher IL28B protein levels than C/T patients. Within the various disease stages, compared to IC and healthy controls, IL28B expression was reduced in the CHB, cirrhosis, and HCC cohorts (CHB vs. IC, p=0.02; cirrhosis vs. IC, p=0.01; HCC vs. IC, p=0.001; CHB vs. controls, p&#60;0.01; cirrhosis vs. controls, p&#60;0.01; HCC vs. controls, p&#60;0.01). When stratified with respect to serum HBV markers in the IC and CHB cohorts, IL28B mRNA and protein levels were higher in HBeAg-positive than negative individuals (p=0.01). Logistic regression analysis revealed that factors associated with high IL28B protein levels were C/C versus C/T genotype [p=0.016, odds ratio (OR)=0.25, 95% confidence interval (CI)=0.08-0.78], high alanine aminotransferase values (p&#60;0.001, OR=8.02, 95% CI=2.64-24.4), and the IC stage of HBV infection (p&#60;0.001). Conclusion Our data suggest that IL28B genetic variations may play an important role in long-term development of disease in chronic HBV infections.</p

    ADJUVANT EFFECTS OF SIJUNZI DECOCTION IN CHICKENS ORALLY VACCINATED WITH ATTENUATED NEWCASTLE-DISEASE VACCINE

    Many Chinese Herbal medicines (CHMs) and their components have been reported to enhance immunity. In this study, the capacity for the Chinese herbal medicine, oral administration Sijunzi Decoction (SJZD) in stimulating Newcastle disease virus(NDV) immunity in chickens was examined. Serum was sampled on days 20,30,40,50 and 60 and tissues were collected on days 20, 40 and 60, respectively. The immune responses were determined by means of hemagglutination inhibition test, immunohistochemistry examination and semi-quantitative RT–PCR. The results showed that SJZD could increase the antibody titers and the area coefficient of IgA secreting cells, promote the expression of IL-2 mRNA in the whole immune period and IFN-γ mRNA was increased in the initial stage. The SJZD used was safe with no adverse effects on chicken weight or survival, providing evidence for the use of SJZD as an oral adjuvant

    High Frequency of CD4+CXCR5+ TFH Cells in Patients with Immune-Active Chronic Hepatitis B

    BACKGROUND: T follicular helper (TFH) cells are a special subpopulation of T helper cells and can regulate humoral immune responses. This study examined whether the frequency of CD4(+)CXCR5(+) TFH cells could be associated with active immunity in chronic hepatitis B (CHB) patients. METHODOLOGY AND FINDINGS: The frequencies of peripheral blood CD4(+)CXCR5(+) TFH cells, inducible T cell costimulator (ICOS), and/or programmed death 1 (PD-1) positive CD4(+)CXCR5(+) TFH cells in immune-active (IA), immune-tolerant (IT) CHB, and healthy controls (HC) were characterized by flow cytometry analysis. The effect of adevofir dipivoxil treatment on the frequency of CD4(+)CXCR5(+) TFH cells, the concentrations of serum IL-2, IFN-γ, TNF-α, IL-4, IL-6, IL-10, IL-21, ALT, AST, HBsAg, HBsAb, HBeAg, HBeAb and HBV loads in IA patients were determined. The potential association of the frequency of CD4(+)CXCR5(+) TFH cells with clinical measures was analyzed. In addition, the frequency of splenic and liver CD4(+)CXCR5(+) TFH cells in HBV-transgenic mice was examined. We found that the frequency of CD4(+)CXCR5(+) TFH cells in IA patients was significantly higher than that of IT patients and HC, and the percentages of CD4(+)CXCR5(+) TFH in IA patients were positively correlated with AST. Furthermore, the percentages of ICOS(+), PD-1(+), and ICOS(+)PD-1(+) in CD4(+)CXCR5(+) TFH cells in CHB patients were significantly higher than that of HC. Treatment with adefovir dipivoxil reduced the frequency of CD4(+)CXCR5(+) TFH, PD-1(+)CD4(+)CXCR5(+) TFH cells and the concentrations of HBsAg and HBeAg, but increased the concentrations of HBsAb, HBeAb, IL-2 and IFN-γ in IA patients. Moreover, the frequency of splenic and liver CD4(+)CXCR5(+) TFH cells in HBV-transgenic mice was higher than that of wild-type controls. CONCLUSIONS: These data indicate that CD4(+)CXCR5(+) TFH cells may participate in the HBV-related immune responses and that high frequency of CD4(+)CXCR5(+) TFH cells may be a biomarker for the evaluation of active immune stage of CHB patients

    IL28B Genetic Variation Is Associated with Spontaneous Clearance of Hepatitis C Virus, Treatment Response, Serum IL-28B Levels in Chinese Population

    &lt;p&gt;&lt;b&gt;Background:&lt;/b&gt; The interleukin-28B gene (IL28B) locus has been associated with host resistance to hepatitis C virus (HCV) infection and response to PEG-IFN/RBV treatment in western populations. This study was to determine whether this gene variant is also associated with spontaneous clearance of HCV infection, treatment response and IL-28B protein production in Chinese patients.&lt;/p&gt; &lt;p&gt;&lt;b&gt;Methods:&lt;/b&gt; We genotyped IL28B genetic variations (rs12980275, rs8103142, rs8099917 and rs12979860) by pyrosequencing DNA samples from cohorts consisting of 529 subjects with persistent HCV infection, 196 subjects who cleared the infection, 171 healthy individuals and 235 chronic HCV patients underwent IFN/RBV treatment. The expression of IL-28B were measured by ELISA and RT-PCR.&lt;/p&gt; &lt;p&gt;&lt;b&gt;Results:&lt;/b&gt; We found that the four IL28B variants were in complete linkage disequilibrium (r2 = 0.97–0.98). The rs12979860 CC genotype was strongly associated with spontaneously HCV clearance and successful IFN/RBV treatment compared to the CT/TT. IL-28B levels in persistent HCV patients were significantly lower than subjects who spontaneously resolved HCV and healthy controls and were also associated with high levels of ALT (alanine aminotransferase) and AST (aspartate aminotransferase). IL-28B levels were also significantly lower in individuals carrying T alleles than CC homozygous.&lt;/p&gt; &lt;p&gt;&lt;b&gt;Conclusions:&lt;/b&gt; Thus, the rs12979860-CC variant upstream of IL28B gene is associated with spontaneous clearance of HCV, susceptible to IFN/RBV treatment and increased IL-28B levels in this Chinese population.&lt;/p&gt

    Effects of Huangqi Maxingshigan decoction on infectious laryngotracheitis in chickens

    This study was conducted to investigate the effectiveness and possible mechanisms of an herbal prescription, <em>Huangqi Maxingshigan</em> decoction (R<em>adix Astragali, Herba Ephedrae, Almond, Gypsum Fibrosum, Radix Glycytthizae</em>) on infectious laryngotracheitis (ILT) in chickens. One hundred and sixty one-day-old chicks were randomly allocated into four groups, housed in isolated cages of 40 birds each: groupⅠwas Huangqi Maxingshigan decoction treatment group, group II as <em>Ding Chuan San</em> control group, group III as the model group, and group IV as the blank control group. Except for the blank control group, other groups were challenged with infectious laryngotracheitis virus via the intratracheal route on age day 52. Birds in group I were administered <em>Huangqi Maxingshigan </em>decoction at a concentration of 0.4 mL, or with <em>Ding Chuan San</em> in group II for comparison at 48 h post virus challenge (day 54) when they showed clinical signs, 2 times daily for 5 days consecutively. Then, the superoxide dismutases (SOD) activity and malondialdehyde (MDA) contents in serum, the expression of IFN-γ, IL-4 mRNA in spleen, the sIgA contents in tracheal fluid, the sIgA secreting cells in trachea were determined on day 5, day 15, day 25 post infection. The results showed that <em>Huangqi Maxingshigan</em> decoction could correct Th2-dominant Th1/th2 imbalance through up-regulation of IFN-γ, and down-regulation of IL-4 at the transcriptional level, enhance cell-mediated immunity, ease inflammatory responses caused by ILTV. The results also showed that <em>Huangqi Maxingshigan </em>decoction provide an important antioxidant defense in the process of anti-ILT. Furthermore, it could induce the production of sIgA to enhance mucosal immunity

    Bisphenol A Exposure during Pregnancy Alters the Mortality and Levels of Reproductive Hormones and Genes in Offspring Mice

    The present study investigated the reproductive toxicity of bisphenol A (BPA) exposure to the mother on the offspring mice. BPA was given to pregnant mice at 50 mg/kg, 500 mg/kg, and 2500 mg/kg BW BPA daily by gavage during the whole gestation period. The offspring mice were sacrificed at 8 weeks of age. Results showed that exposure of BPA to the mother increased the mortality (P<0.05). Maternal exposure of BPA reduced the levels of T (♂) and FSH (♀) (P<0.01) and elevated E2 (♀) level in the adult offspring (P<0.01). BPA exposure caused testicular damage as shown by less Leydig cells and ovarian injury as shown by more vacuoles and less corpus granules in the adult offspring mice. Immunohistochemistry revealed that maternal exposure of BPA increased Bax and decreased Bcl-2 at the protein levels in testicular and ovary tissues in the offspring mice. BPA significantly reduced the expression of StAR in male offspring (P<0.05). Interestingly, the mRNA levels of Cyp11a were significantly decreased in 50 mg/kg groups and were increased in 500 mg/kg group in the males.
