    Efficacy of colposcopy technique with Pap smear and histology in screening of cervical lesions

    Background: In India the most common genital cancer among women is carcinoma of the cervix (80%). The objective of the study is analysis of efficacy of colposcopy technique with Pap smear and histology in screening of cervical lesions.Methods: Patients above 18 years of age with varied parity and socioeconomic status attending out-patient clinics were studied for correlation between cytology, colposcopy and colposcopically directed biopsy for a one year period.Results: In cytology and colposcopic directed biopsy sensitivity is 81%, specificity is 95%, false positive is 4.9%, false negative is 18% and accuracy is 92%. In colposcopy and directed biopsy sensitivity is 94%, specificity is 91%, false positive is 7.4%, false negative is 5.8% and accuracy is 92.85%. Colposcopy helps to reduce false negative cases seen in cytology.Conclusions: In our study overall incidence of CIN was 19, among them CIN1 in 9%, CIN2 in 4%, CIN3 in 4% and malignancy in 2% cases. Higher sensitivity in colposcopy and directed biopsy 94%, higher specificity 95% in cytology as compared to colposcopy. This emphasizes the use of all 3 methods Papincolaou cytology, colposcopy and histology is complementary to each other and helps to reduce false negative cases

    Secure and Efficient Utilization of Encrypted Cloud Data using Multi-Keyword Ranked Search

    Cloud Computing is a technology that provides services to users such as software as a service, platform as a service and storage as a service. These services are provided based on Pay-per-Use basis so these services are cost effective and flexible. Due to this advantage of cloud computing, the individuals as well as the enterprises are getting motivated to shift their local sensitive and huge data management system to cloud storage. But the sensitive data has to be encrypted before outsourcing in order to provide security to the data. After the data has outsourced it has to be utilized efficiently without losing the originality as it was stored. In this paper we provide a mechanism called ”Multi-keyword Ranked Search over Encrypted cloud data” that gives better and efficient searched result over the encrypted data taking multiple keywords as query, which obsoletes the tradition searching scheme based on plain text search. And we use a “Coordinate Matching” technique to find as many matches as possible and use “inner product similarity” to retrieve relevance search results. So if user wants to retrieve the data stored on cloud, he can specify the multiple keywords and rank for relevance retrieval of results. Finally results the user with top ranked files.     Plants are among the richest sources of bioactive compounds throughout the world for thousands of years and continue to provide new remedies to mankind. Roots of Hemidesmus indicus R. Br. is an important plant drug which is used to cure leprosy, leucoderma, itching, skin disease, asthma, bronchitis, leucorrhoea, dysentery, piles, syphilis, paralysis, urinary disorders and diabetes mellitus. As per Acharya Charaka has mentioned suitable season for collection of Moola i.e. Greeshma and Shishira Rutu. Here an attempt to study to check the potency of root collected as per classical reference. The present study focused on the pharmacognostical, phytochemical investigation as well as HPLC study were performed by taking different solvent extracts of Hemidesmus indicus root. This study highlights the detailed HPLC study on Hemidesmus indicus root by taking different solvent extracts with their increasing polarity which is a referential information for identification parameters and improves our confidence level of acceptability of herbal drugs

    Lamin A Rod Domain Mutants Target Heterochromatin Protein 1ι and β for Proteasomal Degradation by Activation of F-Box Protein, FBXW10

    Lamins are major structural proteins of the nucleus and contribute to the organization of various nuclear functions. Mutations in the human lamin A gene cause a number of highly degenerative diseases, collectively termed as laminopathies. Cells expressing lamin mutations exhibit abnormal nuclear morphology and altered heterochromatin organization; however, the mechanisms responsible for these defects are not well understood.The lamin A rod domain mutants G232E, Q294P and R386K are either diffusely distributed or form large aggregates in the nucleoplasm, resulting in aberrant nuclear morphology in various cell types. We examined the effects of these lamin mutants on the distribution of heterochromatin protein 1 (HP1) isoforms. HeLa cells expressing these mutants showed a heterogeneous pattern of HP1alpha and beta depletion but without altering HP1gamma levels. Changes in HP1alpha and beta were not observed in cells expressing wild-type lamin A or mutant R482L, which assembled normally at the nuclear rim. Treatment with proteasomal inhibitors led to restoration of levels of HP1 isoforms and also resulted in stable association of lamin mutants with the nuclear periphery, rim localization of the inner nuclear membrane lamin-binding protein emerin and partial improvement of nuclear morphology. A comparison of the stability of HP1 isoforms indicated that HP1alpha and beta displayed increased turnover and higher basal levels of ubiquitination than HP1gamma. Transcript analysis of components of the ubiquitination pathway showed that a specific F-box protein, FBXW10 was induced several-fold in cells expressing lamin mutants. Importantly, ectopic expression of FBXW10 in HeLa cells led to depletion of HP1alpha and beta without alteration of HP1gamma levels.Mislocalized lamins can induce ubiquitin-mediated proteasomal degradation of certain HP1 isoforms by activation of FBXW10, a member of the F-box family of proteins that is involved in E3 ubiquitin ligase activity
