    Increased levels of RNA oxidation enhance the reversion frequency in aging pro-apoptotic yeast mutants

    Despite recent advances in understanding the complexity of RNA processes, regulation of the metabolism of oxidized cellular RNAs and the mechanisms through which oxidized ribonucleotides affect mRNA translation, and consequently cell viability, are not well characterized. We show here that the level of oxidized RNAs is markedly increased in a yeast decapping Kllsm4Δ1 mutant, which accumulates mRNAs, ages much faster that the wild type strain and undergoes regulated-cell-death. We also found that in Kllsm4Δ1 cells the mutation rate increases during chronological life span indicating that the capacity to han- dle oxidized RNAs in yeast declines with aging. Lowering intracellular ROS levels by antioxidants recovers the wild- type phenotype of mutant cells, including reduced amount of oxidized RNAs and lower mutation rate. Since mRNA oxidation was reported to occur in different neurodegen- erative diseases, decapping-deficient cells may represent a useful tool for deciphering molecular mechanisms of cell response to such conditions, providing new insights into RNA modification-based pathogenesis

    Test of Einstein Equivalence Principle for 0-spin and half-integer-spin atoms: Search for spin-gravity coupling effects

    We report on a conceptually new test of the equivalence principle performed by measuring the acceleration in Earth's gravity field of two isotopes of strontium atoms, namely, the bosonic 88^{88}Sr isotope which has no spin vs the fermionic 87^{87}Sr isotope which has a half-integer spin. The effect of gravity upon the two atomic species has been probed by means of a precision differential measurement of the Bloch frequency for the two atomic matter waves in a vertical optical lattice. We obtain the values η=(0.2±1.6)×107\eta = (0.2\pm 1.6)\times10^{-7} for the E\"otv\"os parameter and k=(0.5±1.1)×107k=(0.5\pm1.1)\times10^{-7} for the coupling between nuclear spin and gravity. This is the first reported experimental test of the equivalence principle for bosonic and fermionic particles and opens a new way to the search for the predicted spin-gravity coupling effects.Comment: 5 pages, 4 figures.     ATLAS is a general purpose experiment approved for the LHC collider at CERN. An important component of the detector is the central hadronic calorimeter; for its construction more than 600,000 Wave Length Shifting (WLS) fibres (corresponding to a total length of 1,120 Km) have been used. We have built and put into operation a dedicated instrument for the measurement of light yield and attenuation length over groups of 20 fibres at a time. The overall accuracy achieved in the measurement of light yield (attenuation length) is 1.5% (3%).     Intrasexual vibrational behavior of Philaenus spumarius in semi-field conditions

    Insects that communicate by vibrational signals live in a complex interactive network of communication. Most studies on insect intrasexual behavior, based on plant-borne vibrational signals, have targeted few individuals. Despite their importance, behaviors that occur within groups were often overlooked. The study of multiple individuals, when insects occur in high density could simulate the environment in which they live and provide more reliable information on their behavior. In semi-field conditions, we investigated the intrasexual behavior of the meadow spittlebug, Philaenus spumarius. Vibrational signals exchanged among individuals of the same sex were recorded throughout their adult stage, from late spring to early autumn, and during the day, from the morning to the evening using a laser vibrometer. Males were less active than females throughout the season and their interactions were less frequent compared to females. Intrasexual interactions were characterized by signal overlapping in both unisex groups, in addition to signal alternating only in the case of males. In conclusion, the study of signaling behavior in intrasexual groups contributed to a better understanding of P. spumarius social behavior.     Vibrational disruption of feeding behaviors of a vector of plant pathogen

    Interference with the behaviors associated to host plant recognition, and inter-and intra-specific communication of insect vectors of plant pathogens, could represent a sustainable strategy for reducing or disrupting pathogen transmission Here, we show that the transmission over a suitable host plant (sunflower) of a vibrational stimulus significantly affects the probing and feeding behavior of the spittlebug Philaenus spumarius (Hemiptera: Aphrophoridae), the main European vector of the fastidious bacterium Xylella fastidiosa. Specifically, ca. 30% of the individuals did not even attempt to probe the sunflower plants to which the stimulus was transmitted, while the remaining showed a sex-independent reduction in inges-tion of the xylem sap, i.e., P. spumarius\u2019 main food source, of ca. 67% compared to the control. Even so, the stimulus did not affect the feeding behavior when transmitted to olive plants. The possible reflection of a signal-based vector behavior disturbance on the epidemiology of X. fastidiosa, together with future research needs are discussed

    Water level and volume estimations of the Albano and Nemi lakes (central Italy)

    In April 2006 an airborne laser scanning (LIDAR) survey of the Albano and Nemi craters was carried out to obtain a high resolution digital terrain model (DTM) of the area. We have integrated the LIDAR survey of the craters and the recent bathymetry of the Albano lake to achieve a complete DTM, useful for morphological studies. In addition, with a GPS RTK survey (July 2007) we estimated the Albano and Nemi mean lake levels respectively at 288.16 m and 319.02 m (asl). Based on the integrated DTM and the newly estimated water level values, we evaluated about 21.7·106 m3 the water volume loss of the Albano lake from 1993 to 2007, with an average rate of about 1.6·106 m3/yr

    Towards Understanding Photodegradation Pathways in Lignins:The Role of Intramolecular Hydrogen Bonding in Excited States

    The photoinduced dynamics of the lignin building blocks syringol, guaiacol, and phenol were studied using time-resolved ion yield spectroscopy and velocity map ion imaging. Following irradiation of syringol and guaiacol with a broad-band femtosecond ultraviolet laser pulse, a coherent superposition of out-of-plane OH torsion and/or OMe torsion/flapping motions is created in the first excited 1ππ* (S1) state, resulting in a vibrational wavepacket, which is probed by virtue of a dramatic nonplanar → planar geometry change upon photoionization from S1 to the ground state of the cation (D0). Any similar quantum beat pattern is absent in phenol. In syringol, the nonplanar geometry in S1 is pronounced enough to reduce the degree of intramolecular H bonding (between OH and OMe groups), enabling H atom elimination from the OH group. For guaiacol, H bonding is preserved after excitation, despite the nonplanar geometry in S1, and prevents O–H bond fission. This behavior affects the propensities for forming undesired phenoxyl radical sites in these three lignin chromophores and provides important insight into their relative “photostabilities” within the larger biopolymer
