Resource efficient processing and communication in sensor/actuator environments
The future of computer systems will not be dominated by personal computer like hardware platforms but by embedded and cyber-physical systems assisting humans in a hidden but omnipresent manner. These pervasive computing
devices can, for example, be utilized in the home automation sector to create sensor/
actuator networks supporting the inhabitants of a house in everyday life. The efficient usage of resources is an important topic at design time and operation time of mobile embedded and cyber-physical systems. Therefore, this thesis presents methods which allow an efficient use of energy and processing resources in sensor/actuator networks. These networks comprise different nodes cooperating for a âsmartâ joint control function. Sensor/actuator nodes are typical cyber-physical systems comprising sensors/actuators and processing and communication components. Processing components of todayâs sensor nodes can comprise many-core chips. This thesis introduces new methods for optimizing the code and the application mapping of the aforementioned systems and presents novel results with regard to design space explorations for energy-efficient and embedded many-core systems. The considered many-core systems are graphics processing units. The application code for these graphics processing units is optimized for a particular platform variant with the objectives of minimal energy consumption and/or of minimal runtime. These two objectives are targeted with the utilization of multi-objective optimization techniques. The mapping optimizations are realized by means of multi-objective design space explorations. Furthermore, this thesis introduces new techniques and functions for a resource-efficient middleware design employing service-oriented architectures.
Therefore, a service-oriented architecture based middleware framework is presented which comprises a lightweight service orchestration. In addition to that,
a flexible resource management mechanism will be introduced. This resource management
adapts resource utilization and services to an environmental context and provides methods to reduce the energy consumption of sensor nodes
Diagnostic Benefit of High b-Value Computed Diffusion-Weighted Imaging in Patients with Hepatic Metastasis
Diffusion-weighted imaging (DWI) has rapidly become an essential tool for the detection of malignant liver lesions. The aim of this study was to investigate the usefulness of high b-value computed DWI (c-DWI) in comparison to standard DWI in patients with hepatic metastases. In total, 92 patients with histopathologic confirmed primary tumors with hepatic metastasis were retrospectively analyzed by two readers. DWI was obtained with b-values of 50, 400 and 800 or 1000 s/mm2 on a 1.5 T magnetic resonance imaging (MRI) scanner. C-DWI was calculated with a monoexponential model with high b-values of 1000, 2000, 3000, 4000 and 5000 s/mm2. All c-DWI images with high b-values were compared to the acquired DWI sequence at a b-value of 800 or 1000 s/mm2 in terms of volume, lesion detectability and image quality. In the group of a b-value of 800 from a b-value of 2000 s/mm2, hepatic lesion sizes were significantly smaller than on acquired DWI (metastases lesion sizes b = 800 vs. b 2000 s/mm2: mean 25 cm3 (range 10â60 cm3) vs. mean 17.5 cm3 (range 5â35 cm3), p < 0.01). In the second group at a high b-value of 1500 s/mm2, liver metastases were larger than on c-DWI at higher b-values (b = 1500 vs. b 2000 s/mm2, mean 10 cm3 (range 4â24 cm3) vs. mean 9 cm3 (range 5â19 cm3), p < 0.01). In both groups, there was a clear reduction in lesion detectability at b = 2000 s/mm2, with hepatic metastases being less visible compared to c-DWI images at b = 1500 s/mm2 in at least 80% of all patients. Image quality dropped significantly starting from c-DWI at b = 3000 s/mm2. In both groups, almost all high b-values images at b = 4000 s/mm2 and 5000 s/mm2 were not diagnostic due to poor image quality. High c-DWI b-values up to b = 1500 s/mm2 offer comparable detectability for hepatic metastases compared to standard DWI. Higher b-value images over 2000 s/mm2 lead to a noticeable reduction in imaging quality, which could hamper diagnosis
Magnetic Resonance Imaging of Peritoneal Carcinomatosis: Evaluation of High b-Value Computed Diffusion-Weighted Imaging
Over the last few years, diffusion-weighted imaging (DWI) has become increasingly
relevant in the diagnostic assessment of peritoneal carcinomatosis. The aim of this study was
to investigate the benefits of high-b DWI (c-DWI) compared to standard DWI in patients with
peritoneal carcinomatosis. A cohort of 40 patients with peritoneal carcinomatosis were included in
this retrospective study. DWI was performed with b-values of 50, 400, and 800 or 1000 s/mm2 on a
1.5-T magnetic resonance imaging (MRI) scanner. C-DWI was calculated using a mono-exponential
model with high b-values of 1000, 2000, 3000, 4000, and 5000 s/mm2. All c-DWI images with high
b-values were compared in terms of volume, detectability of peritoneal lesions, and image quality
with the DWI sequence acquired with a b-value of 800 or 1000 s/mm2 by two readers. In the group
with a b-value of 800 s/mm2, there was no statistically significant difference in terms of lesion volume.
In the second group with a b-value of 1000 s/mm2, peritoneal carcinomatosis lesions were statistically
significantly larger than in the c-DWI with a- high b-value of 2000 s/mm2 (median 7 cm3, range
1â26 cm3vs. median 6 cm3, range 1â83 cm3, p < 0.05). In both groups, there was a marked decrease
in the detectability of peritoneal lesions starting at b = 2000 s/mm2. In addition, image quality
decreased noticeably from c-DWI at b = 3000 s/mm2. In both groups, all images with high b-values
at b = 4000 s/mm2 and 5000 s/mm2 were not diagnostically valuable due to poor image quality.
The c-DWI technique offers good diagnostic performance without additional scanning time. High
c-DWI b-values up to b = 1000 s/mm2 provide comparable detectability of peritoneal carcinomatosis
compared to standard DWI. Higher b-values over 1500 s/mm2 result in lower image quality, which
might lead to misdiagnosis
The TreaT-Assay: A Novel Urine-Derived Donor Kidney Cell-Based Assay for Prediction of Kidney Transplantation Outcome
Donor-reactive immunity plays a major role in rejection after kidney transplantation, but analysis of donor-reactive T-cells is not applied routinely. However, it has been shown that this could help to identify patients at risk of acute rejection. A major obstacle is the limited quantity or quality of the required allogenic stimulator cells, including a limited availability of donor-splenocytes or an insufficient HLA-matching with HLA-bank cells. To overcome these limitations, we developed a novel assay, termed the TreaT (Transplant reactive T-cells)-assay. We cultivated renal tubular epithelial cells from the urine of kidney transplant patients and used them as stimulators for donor-reactive T-cells, which we analyzed by flow cytometry. We could demonstrate that using the TreaT-assay the quantification and characterization of alloreactive T-cells is superior to other stimulators. In a pilot study, the number of pre-transplant alloreactive T-cells negatively correlated with the post-transplant eGFR. Frequencies of pre-transplant CD161+ alloreactive CD4+ T-cells and granzyme B producing alloreactive CD8+ T-cells were substantially higher in patients with early acute rejection compared to patients without complications. In conclusion, we established a novel assay for the assessment of donor-reactive memory T-cells based on kidney cells with the potential to predict early acute rejection and post-transplant eGFR
Immune Response in Moderate to Critical Breakthrough COVID-19 Infection After mRNA Vaccination
SARS-CoV-2 variants of concern (VOCs) can trigger severe endemic waves and vaccine breakthrough infections (VBI). We analyzed the cellular and humoral immune response in 8 patients infected with the alpha variant, resulting in moderate to fatal COVID-19 disease manifestation, after double mRNA-based anti-SARS-CoV-2 vaccination. In contrast to the uninfected vaccinated control cohort, the diseased individuals had no detectable high-avidity spike (S)-reactive CD4+ and CD8+ T cells against the alpha variant and wild type (WT) at disease onset, whereas a robust CD4+ T-cell response against the N- and M-proteins was generated. Furthermore, a delayed alpha S-reactive high-avidity CD4+ T-cell response was mounted during disease progression. Compared to the vaccinated control donors, these patients also had lower neutralizing antibody titers against the alpha variant at disease onset. The delayed development of alpha S-specific cellular and humoral immunity upon VBI indicates reduced immunogenicity against the S-protein of the alpha VOC, while there was a higher and earlier N- and M-reactive T-cell response. Our findings do not undermine the current vaccination strategies but underline a potential need for the inclusion of VBI patients in alternative vaccination strategies and additional antigenic targets in next-generation SARS-CoV-2 vaccines
Providing Information by Resource- Constrained Data Analysis
The Collaborative Research Center SFB 876 (Providing Information by Resource-Constrained Data Analysis) brings together the research fields of data analysis (Data Mining, Knowledge Discovery in Data Bases, Machine Learning, Statistics) and embedded systems and enhances their methods such that information from distributed, dynamic masses of data becomes available anytime and anywhere. High incidence and viral load of HHV-6A in a multi-centre kidney transplant cohort
Human herpesvirus 6 (HHV-6) is a common opportunistic pathogen in kidney transplant recipients. Two distinct species of HHV-6, HHV-6A and HHV-6B, have been identified, of which the latter seems to be dominant. However, it is unclear whether they increase the likelihood of other viral reactivations. We characterized a multi-centre cohort of 93 patients along nine study visits for viral load. We tested for the following viruses: HHV-6A and HHV-6B, the herpesviruses cytomegalovirus (CMV) and Epstein-Barr virus (EBV) and the polyomavirus BK (BKV). We detected HHV-6A viral load in 48 (51.6%) patients, while the incidence of HHV-6B was much lower, being detected in 6 (6.5%) patients. The incidence of HHV-6A was higher than of BKV, CMV and EBV. HHV-6A also demonstrated higher viral loads than the rest of viruses. There was a non-significant trend of association between HHV-6A and HHV-6B as co-infection, whereas no increased incidence of other viruses among patients with HHV-6A reactivation was observed. There was no negative effect of high HHV-6A (>10,000â
copies/ml) load on markers of renal graft and hepatic function or blood count twelve months post-transplant. In contrast to previously published data, our results show a clear dominance of HHV-6A in peripheral blood when compared to HHV-6B, with higher incidence and viral load levels. Despite the high HHV-6A loads observed, we did not identify any negative effects on posttransplant outcome
