Syntheses and Biological Activity of the HDAC Class I Inhibitor Largazole
Histone deacetylase inhibitors are an emerging class of antiproliferative agents that have the potential to combat cancer on an epigenetic level. The recently reported marine natural product largazole has been recently isolated in trace amounts from cyanobacteria and displays a very
potent and selective antiproliferative activity towards a number of cell lines. Several lines of evidence suggested it might act as a histone deacetylase inhibitor. These compelling properties have generated considerable interest in the synthetic community which resulted in several total syntheses
of largazole. The present review offers a brief overview of the synthetic routes and some early structureâactivity relationships
Identification and Heterologous Expression of the Albucidin Gene Cluster from the Marine Strain Streptomyces Albus Subsp. Chlorinus NRRL B-24108
Herbicides with new modes of action and safer toxicological and environmental profiles are
needed to manage the evolution of weeds that are resistant to commercial herbicides. The unparalleled
structural diversity of natural products makes these compounds a promising source for new herbicides.
In 2009, a novel nucleoside phytotoxin, albucidin, with broad activity against grass and broadleaf
weeds was isolated from a strain of Streptomyces albus subsp. chlorinus NRRL B-24108. Here, we report
the identification and heterologous expression of the previously uncharacterized albucidin gene
cluster. Through a series of gene inactivation experiments, a minimal set of albucidin biosynthetic
genes was determined. Based on gene annotation and sequence homology, a model for albucidin
biosynthesis was suggested. The presented results enable the construction of producer strains for
a sustainable supply of albucidin for biological activity studies
The Synthesis of Certain Phomentrioloxin A Analogues and Their Evaluation as Herbicidal Agents
A series of 28 analogues of the phytotoxic geranylcyclohexentriol (â)-phomentrioloxin A (1) has been synthesized through cross-couplings of various enantiomerically pure haloconduritols or certain deoxygenated derivatives with either terminal alkynes or borylated alkenes. Some of these analogues display modest herbicidal activities, and physiological profiling studies suggest that analogue 4 inhibits photosystem II in isolated thylakoids in vitro
Enantioselective rhodium-catalyzed C-C bond activations for the construction of quaternary stereogenic centers
Rhodium(I)-Catalyzed 1,4-Silicon Shift of Unactivated Silanes from Aryl to Alkyl: Enantioselective Synthesis of Indanol Derivatives
Enantioselective Rh-promoted activation and 1,4-positional swap of unactivated tetraorganosilanes, e.g., 1-ethyl-3-methyl-3-(2-(trimethylsilyl)phenyl)cyclobutanol (I) to give (1S,3S)-1-ethyl-3-methyl-3((trimethylsilyl)methyl)indanol (II). E.g., reaction of silylphenyl tert-cyclobutanol I with 2.5 mol% [Rh(cod)OH]2/6.0 mol% (R)-Difluorphos ligand at 100ÂŹâ in mesitylene gave 82% yield of trans-indanol II (97% ee)
Enantioselective metal-catalyzed activation of strained rings
A review. Activation of otherwise inert bonds has significant potential in the design of efficient and synthetically useful transformations. While general catalytic carbon-carbon single bond activations are still in their infancy, this emerging area examines recent developments in the activation of strained rings, focusing on enantioselective reactions
Beta-carbon elimination from cyclobutanols: a clean access to alkylrhodium intermediates bearing a quaternary stereogenic center
A review. The catalytic activation of C-C bonds bears significant ecol. and economical advantages. In this account, the authors describe their results for an enantioselective C-C activation of sym. substituted tert-cyclobutanols. Subsequent downstream reactions of the obtained alkylrhodium intermediate give rise to a wide range of synthetically versatile products bearing all-carbon quaternary stereogenic centers with excellent enantioselectivities
