    thesisA significant amount of effort has been put into the research and development of peptide-based drugs for the treatment of human disease. The use of anticonvulsant neuropeptides for the treatment of pain and epilepsy has recently shown promise. Neuropeptides such as galanin (Gal), neuropeptide Y (NPY), neurotensin (NT), and neuropeptide W (NPW) modulate their biological activities through their respective receptor targets in the brain. Introduced intracerebroventricularly, these compounds have been shown to reduce pain and seizure activity. Upon systemic administration, however, these compounds do not exhibit the same potency. The metabolic instability and inability to penetrate the blood-brain barrier (BBB) has precluded their widespread use as drugs for neurological disorders. This work will describe the application of the lipidizationcationization strategy to several endogenous neuropeptides towards increasing their metabolic stability and improving systemic bioavailability. Based on existing structure activity relationship (SAR) information, analogs of Gal, NPY NT, and NPW were designed and chemically synthesized. The combination of increased cationic character (oligo-Lys motifs) and increased lipophilicity (lipoamino acid) resulted in analogs with increased octanol-water partitioning coefficients (logD), significantly improved metabolic stabilities, while retaining high affinities for their native receptors. Furthermore, these analogs were shown to modulate seizure activity in animal models of epilepsy upon systemic administration. Our results suggest that the cationizationlipidization strategy may be broadly applicable across a wide variety of neuropeptides, opening up a large repertoire of new drug candidates for the treatment of neurological disorders

    Psychology has much to contribute to engagement in community organizing and direct action. Such organizing and action is psychological: it is motivational, cognitive, attitudinal, and emotional. Psychological knowledge is a cost-free resource. It can be used to counter unjust power structures and their tendency to maintain policies and systems of social harm and inequality. A theoretical model of organizing and direct action that integrates knowledge from community psychology and related disciplines with historic and modern action campaigns is needed. Theory helps us learn from the past to guide us toward more effective future outcomes

    Nuclear receptor agonist-driven modification of inflammation and amyloid pathology enhances and sustains cognitive improvements in a mouse model of Alzheimer's disease

    BACKGROUND: Alzheimer's disease (AD) is a highly prevalent neurodegenerative disorder characterized by pathological hallmarks of beta-amyloid plaque deposits, tau pathology, inflammation, and cognitive decline. Treatment remains a clinical obstacle due to lack of effective therapeutics. Agonists targeting nuclear receptors, such as bexarotene, reversed cognitive deficits regardless of treatment duration and age in murine models of AD. While bexarotene demonstrated marked efficacy in decreasing plaque levels following short-term treatment, prolonged treatment did not modulate plaque burden. This suggested that plaques might reform in mice treated chronically with bexarotene and that cessation of bexarotene treatment before plaques reform might alter amyloid pathology, inflammation, and cognition in AD mice. METHODS: We utilized one-year-old APP/PS1 mice that were divided into two groups. We treated one group of mice for 2 weeks with bexarotene. The other group of mice was treated for 2 weeks with bexarotene followed by withdrawal of drug treatment for an additional 2 weeks. Cognition was evaluated using the novel-object recognition test either at the end of bexarotene treatment or the end of the withdrawal period. We then analyzed amyloid pathology and microgliosis at the conclusion of the study in both groups. RESULTS: Bexarotene treatment enhanced cognition in APP/PS1 mice similar to previous findings. Strikingly, we observed sustained cognitive improvements in mice in which bexarotene treatment was discontinued for 2 weeks. We observed a sustained reduction in microgliosis and plaque burden following drug withdrawal exclusively in the hippocampus. CONCLUSIONS: Our findings demonstrate that bexarotene selectively modifies aspects of neuroinflammation in a region-specific manner to reverse hippocampal-dependent cognitive deficits in AD mice and may provide insight to inform future studies with nuclear receptor agonists
