    Semi-free forearm flap for pharyngeal-esophageal reconstruction after radiation therapy

    AbstractReconstruction of the pharynx and upper esophagus uses various procedures, including pedicled or free flap. Pharyngoplasty with free forearm flap provides excellent functional results. In radiation-related pharyngeal stenosis, recipient vascularization is often poor, especially in the venous system. The authors describe pharyngeal reconstruction with semi-free forearm flap, pedicled on the cephalic vein, to minimize the risk of venous thrombosis, which is the main factor of free forearm flap necrosis. Taking the case of a laryngectomy with complete pharyngeal stenosis after radiation therapy and iterative neck surgery, the technique of pharyngeal-esophageal reconstruction by semi-free forearm flap is described in a context of impaired vascularization

    International audienceMost of the Coq proteins involved in coenzyme Q (ubiquinone or Q) biosynthesis are interdependent within a multiprotein complex in the yeast Saccharomyces cerevisiae. Lack of only one Coq polypeptide, as in Δcoq strains, results in the degradation of several Coq proteins. Consequently, Δcoq strains accumulate the same early intermediate of the Q(6) biosynthetic pathway; this intermediate is therefore not informative about the deficient biosynthetic step in a particular Δcoq strain. In this work, we report that the overexpression of the protein Coq8 in Δcoq strains restores steady state levels of the unstable Coq proteins. Coq8 has been proposed to be a kinase, and we provide evidence that the kinase activity is essential for the stabilizing effect of Coq8 in the Δcoq strains. This stabilization results in the accumulation of several novel Q(6) biosynthetic intermediates. These Q intermediates identify chemical steps impaired in cells lacking Coq4 and Coq9 polypeptides, for which no function has been established to date. Several of the new intermediates contain a C4-amine and provide information on the deamination reaction that takes place when para-aminobenzoic acid is used as a ring precursor of Q(6). Finally, we used synthetic analogues of 4-hydroxybenzoic acid to bypass deficient biosynthetic steps, and we show here that 2,4-dihydroxybenzoic acid is able to restore Q(6) biosynthesis and respiratory growth in a Δcoq7 strain overexpressing Coq8. The overexpression of Coq8 and the use of 4-hydroxybenzoic acid analogues represent innovative tools to elucidate the Q biosynthetic pathway

    Characterization of the Phytochelatin Synthase of Schistosoma mansoni

    Treatment for schistosomiasis, which is responsible for more than 280,000 deaths annually, depends exclusively on the use of praziquantel. Millions of people are treated annually with praziquantel and drug resistant parasites are likely to evolve. In order to identify novel drug targets the Schistosoma mansoni sequence databases were queried for proteins involved in glutathione metabolism. One potential target identified was phytochelatin synthase (PCS). Phytochelatins are oligopeptides synthesized enzymatically from glutathione by PCS that sequester toxic heavy metals in many organisms. However, humans do not have a PCS gene and do not synthesize phytochelatins. In this study we have characterized the PCS of S. mansoni (SmPCS). The conserved catalytic triad of cysteine-histidine-aspartate found in PCS proteins and cysteine proteases is also found in SmPCS, as are several cysteine residues thought to be involved in heavy metal binding and enzyme activation. The SmPCS open reading frame is considerably extended at both the N- and C-termini compared to PCS from other organisms. Multiple PCS transcripts are produced from the single encoded gene by alternative splicing, resulting in both mitochondrial and cytoplasmic protein variants. Expression of SmPCS in yeast increased cadmium tolerance from less than 50 ”M to more than 1,000 ”M. We confirmed the function of SmPCS by identifying PCs in yeast cell extracts using HPLC-mass spectrometry. SmPCS was found to be expressed in all mammalian stages of worm development investigated. Increases in SmPCS expression were seen in ex vivo worms cultured in the presence of iron, copper, cadmium, or zinc. Collectively, these results indicate that SmPCS plays an important role in schistosome response to heavy metals and that PCS is a potential drug target for schistosomiasis treatment.     A Tradeoff Drives the Evolution of Reduced Metal Resistance in Natural Populations of Yeast

    Various types of genetic modification and selective forces have been implicated in the process of adaptation to novel or adverse environments. However, the underlying molecular mechanisms are not well understood in most natural populations. Here we report that a set of yeast strains collected from Evolution Canyon (EC), Israel, exhibit an extremely high tolerance to the heavy metal cadmium. We found that cadmium resistance is primarily caused by an enhanced function of a metal efflux pump, PCA1. Molecular analyses demonstrate that this enhancement can be largely attributed to mutations in the promoter sequence, while mutations in the coding region have a minor effect. Reconstruction experiments show that three single nucleotide substitutions in the PCA1 promoter quantitatively increase its activity and thus enhance the cells' cadmium resistance. Comparison among different yeast species shows that the critical nucleotides found in EC strains are conserved and functionally important for cadmium resistance in other species, suggesting that they represent an ancestral type. However, these nucleotides had diverged in most Saccharomyces cerevisiae populations, which gave cells growth advantages under conditions where cadmium is low or absent. Our results provide a rare example of a selective sweep in yeast populations driven by a tradeoff in metal resistance

    L’analyse des emplois prĂ©positionnels et des emplois conjonctifs d’ “exceptĂ©â€, de “sauf” et d’ “hormis” permet d’envisager les trois prĂ©positions/conjonctions comme le pivot d’un binĂŽme, comme la plaque tournante d’une structure bipolaire. PlacĂ©es au milieu du binĂŽme, ces prĂ©positions sont forcĂ©es par leur sĂ©mantisme originaire dĂ»ment mĂ©taphorisĂ© de jouer le rĂŽle de marqueurs d’inconsĂ©quence systĂ©matique entre l’élĂ©ment se trouvant Ă  leur gauche et celui qui se trouve Ă  leur droite. L’opposition qui surgit entre les deux Ă©lĂ©ments n’est donc pas une incompatibilitĂ© naturelle, intrinsĂšque, mais extrinsĂšque, induite. Dans la plupart des cas (emplois limitatifs), cette opposition prend la forme d’un rapport entre une « classe » et le « membre (soustrait) de la classe », ou bien entre un « tout » et une « partie » ; dans d’autres (emplois exceptifs), cette opposition se manifeste au contraire comme une attaque de front portĂ©e par un « tout » Ă  un autre « tout ». De plus, l’inconsĂ©quence induite mise en place par la prĂ©position/conjonction paraĂźt, en principe, tout Ă  fait insurmontable. Dans l’assertion « les Ă©cureuils vivent partout, sauf en Australie » (que l’on peut expliciter par « Les Ă©cureuils vivent partout, sauf [qu’ils ne vivent pas] en Australie »), la prĂ©position semble en effet capable d’impliquer le prĂ©dicat principal avec signe inverti, et de bĂątir sur une telle implication une sorte de sous Ă©noncĂ© qui, Ă  la rigueur, est totalement inconsĂ©quent avec celui qui le prĂ©cĂšde (si « les Ă©cureuils ne vivent pas en Australie », le fait qu’ils « vivent partout » est faux). NĂ©anmoins, l’analyse montre qu’alors que certaines de ces oppositions peuvent enfin ĂȘtre dĂ©passĂ©es, d’autres ne le peuvent pas. C’est, respectivement, le cas des relations limitatives et des relations exceptives. La relation limitative, impliquant le rapport « tout » - « partie », permet de rĂ©soudre le conflit dans les termes d’une somme algĂ©brique entre deux sous Ă©noncĂ©s pourvus de diffĂ©rent poids informatif et de signe contraire. Les valeurs numĂ©riques des termes de la somme Ă©tant dĂ©sĂ©quilibrĂ©es, le rĂ©sultat est toujours autre que zĂ©ro. La relation exceptive, au contraire, qui n’implique pas le rapport « tout » - « partie », n’est pas capable de rĂ©soudre le conflit entre deux sous Ă©noncĂ©s pourvus du mĂȘme poids informatif et en mĂȘme temps de signe contraire : les valeurs numĂ©riques des termes de la somme Ă©tant symĂ©triques et Ă©gales, le rĂ©sultat sera toujours Ă©quivalent Ă  zĂ©ro

    Effect of potential cycling on structure and activity of Pt nanoparticles dispersed on different carbon supports

    International audiencePlatinum particles synthesized via the Bonnemann methods were dispersed on two different Vulcan XC72 carbon Supports. One was used after thermal treatment at 400 degrees C under nitrogen atmosphere, the other after oxidation of its Surface by acid leaching using diluted HNO3 in water (1/3). Characterization of the carbon support indicated that HNO3 treatment leads to the decrease of the BET surface and to the increase of the surface acidity of the carbon support. After dispersion of the platinum catalyst, TEM results indicated that the mean particle size was a little higher on the non-oxidized support (Pt/XC72) than that on the functionalized one (Pt/XC72(HNO3)), being 2.5 and 2.0 nm, respectively. However, potential cycles from 0.05 to 1.25 V vs. RHE led to a higher increase of the particle size when catalyst is dispersed on the functionalized support, reaching after 400 potential cycles 5.5 nm against 4.0 nm with the non-functionalized one. The effect of the upper limit (1.0 and 1.25 V vs. RHE) of the potential cycles on the active surface area and on the activity towards the oxygen reduction reaction (orr) was determined for both catalysts. Results indicated that the particle growth was not the main degradation process over the whole duration of the electrochemical experiments, but that dissolution/redeposition (Otswald ripening) was also involved. The predominant role of each degradation process depends on the number of cycles, on the upper potential limit and on the carbon surface state, and could be temporally separated. However, the lower activity towards orr was recorded for the (Pt/XC72(HNO3)) cycled Lip to 1.0 V vs. RHE
