    The employment of waste paper to microbial production of cellulases

    Studium možnosti využití odpadního papíru na mikrobiální produkci celulas bylo prováděno za pomoci kultivace Aspergillus niger na zdroji uhlíku, kterým byl odpadní kancelářský papír a karton, zvlhčený bezuhlíkatým médiem, nebo destilovanou vodou. Kultivace probíhala způsobem SSF v Erlenmeyerových baňkách a kolonách. Byla sledována koncentrace extracelulárních proteinů, jejich celulasová akivita a u vybraných vzorků i aktivita proteasová. Bylo zjištěno, že nejvýhodnějším způsobem kultivace z hlediska produkce celulas je kultivace v koloně, promývané bezuhlíkatým médiem v třídenních intervalech.Study of the employment of waste paper to microbial production of cellulase was carried out using Aspergillus niger cultivation on carbon sources that have been waste office paper and cardboard, humidified by no – carbon medium or distiled water. Cultivation took place in the SSF way in Erlenmeyer flasks and columns. Concentration of extracellular proteins, cellulase and protease activity for selected samples were monitored. It was found that the most advantageous method of cultivation in terms of cellulase activity production is the cultivation in the column washing by no – carbon medium in three day intervals.

    Structure Formation in Tailor-Made Buriti Oil Emulsion During Simulated Digestion

    Functional food emulsions enriched in health-promoting nutrients can help to maintain and improve health and lifestyle. The oil extracted from the Amazonian buriti fruit is renowned for its high levels of carotenoid, vitamin E, and unsaturated fatty acids, which have been linked to improvements in cardiometabolic health. Here, buriti oil in water emulsions are developed and their colloidal transformations are investigated in an advanced digestion model with oral, gastric, and intestinal parts with in situ synchrotron small-angle X-ray scattering, cryogenic electron microscopy, and dynamic light scattering under simulated “healthy” and “compromised” digestive conditions. The interior oil phase of whey-stabilized buriti oil-in-water emulsion transforms into highly ordered lyotropic liquid crystalline structures during simulated intestinal digestion at compromised bile conditions. Simulated gastric digestion influences intestinal digestion by slowing it down, resulting in less ordered structures. The digestion-triggered structure formation is pH- and bile salt-dependent and can be modulated by adding vitamin E to the oil. This tailoring of structures during digestion offers a new pathway to steer digestion kinetics and nutrient bioavailability.ISSN:1616-3028ISSN:1616-301

    Rapid single-step upconversion-linked immunosorbent assay for diclofenac

    The non-steroidal anti-inflammatory drug and analgesic diclofenac is a common micropollutant in water. A direct competitive upconversion-linked immunosorbent assay (ULISA) for diclofenac has been developed that is based on a nanoparticulate tracer consisting of a NaYF4:Yb, Er core (diameter of similar to 90 nm, excitation and emission wavelenghts 980 and 535 nm, respectively) enclosed by a carboxylated silica shell and finally coated with diclofenac-conjugated bovine gamma-globulin. The proteinaceous coating prevents non-specific adsorption of the tracer to the microtiter plate and provides a structurally flexible linker for surface-exposed diclofenac to warrant efficient competition with free (analyte) diclofenac in real water samples. The tracer was purified by gel electrophoresis and lyophilized. Both processes have no adverse effects on the immunoassay. All assay components can be stored in a dry state without a cooling chain, and can be reactivated on demand. Hence, this ULISA is well suited for environmental monitoring in low-resource settings. The ULISA has a similar limit of detection (20 pg mL(-1); equivalent to 70 pM) as the conventional ELISA, but the time for analysis is reduced to 70 min because no enzymatic amplification steps are involved

    PDTSC 1.0 is a multi-purpose corpus of spoken language. 768,888 tokens, 73,374 sentences and 7,324 minutes of spontaneous dialog speech have been recorded, transcribed and edited in several interlinked layers: audio recordings, automatic and manual transcription and manually reconstructed text. PDTSC 1.0 is a delayed release of data annotated in 2012. It is an update of Prague Dependency Treebank of Spoken Language (PDTSL) 0.5 (published in 2009). In 2017, Prague Dependency Treebank of Spoken Czech (PDTSC) 2.0 was published as an update of PDTSC 1.0
