    inborn errors of immunity associated with atopy (IEIs-A) are a group of inherited monogenic disorders that occur with immune dysregulation and frequent skin involvement. several pathways are involved in the pathogenesis of these conditions, including immune system defects, alterations of skin barrier and metabolism perturbations. current technological improvements and the higher accessibility to genetic testing, recently allowed the identification of novel molecular pathways involved in IEIs-A, also informing on potential tailored therapeutic strategies. compared to other systemic therapy for skin diseases, biologics have the less toxic and the best tolerated profile in the setting of immune dysregulation.     Human germline heterozygous gain-of-function STAT6 variants cause severe allergic disease

    sharma et al. define a new primary atopic disorder caused by heterozygous gain-of-function variants in STAT6. this results in severe, early-onset allergies, and is seen in 16 patients from 10 families. Anti-IL-4R & alpha; antibody and JAK inhibitor treatment were highly effective.STAT6 (signal transducer and activator of transcription 6) is a transcription factor that plays a central role in the pathophysiology of allergic inflammation. we have identified 16 patients from 10 families spanning three continents with a profound phenotype of early-life onset allergic immune dysregulation, widespread treatment-resistant atopic dermatitis, hypereosinophilia with esosinophilic gastrointestinal disease, asthma, elevated serum IgE, IgE-mediated food allergies, and anaphylaxis. the cases were either sporadic (seven kindreds) or followed an autosomal dominant inheritance pattern (three kindreds). all patients carried monoallelic rare variants in STAT6 and functional studies established their gain-of-function (GOF) phenotype with sustained STAT6 phosphorylation, increased STAT6 target gene expression, and T(H)2 skewing. Precision treatment with the anti-IL-4R & alpha; antibody, dupilumab, was highly effective improving both clinical manifestations and immunological biomarkers. this study identifies heterozygous GOF variants in STAT6 as a novel autosomal dominant allergic disorder. We anticipate that our discovery of multiple kindreds with germline STAT6 GOF variants will facilitate the recognition of more affected individuals and the full definition of this new primary atopic disorder

    Nel 1987, sul Kōya-san, la montagna sacra al buddhismo Shingon, vi fu il primo incontro tra il professor Shodo Abukawa e Luigi Giussani. Da questo incontro nacque una grande amicizia, la quale non fu solo un esempio vissuto di dialogo interreligioso, ma anche un percorso di comunione, di compagnia Introduzione alla realtà totale lungo la strada che conduce al Mistero. Di questo percorso Giussani ha scritto in diverse occasioni, lasciando precisi elementi per la costruzione di una “teologia dell’incontro” con coloro che non credono in Cristo, ma sono mossi dalle grandi domande di senso che da sempre agitano l’animo umano. Scopo del presente contributo è quello di o"rire uno studio ragionato e documentato della “teologia dell’incontro” di Giussani e di utilizzarne i contenuti per offrire un’interpretazione dei concetti di “opera missionaria” e di “dialogo sincero” utilizzati, rispettivamente, dai cann. 781 e 787, § 1, del Codice di Diritto Canonico latino. In particolare, si dimostrerà come per il nostro autore il dialogo di cui al canone 787 è sincero quando si struttura in un avere cura, in un’attenzione alla persona dell’altro, la quale si pone come immagine dell’esperienza di comunione in Cristo e con Cristo che il fedele è chiamato a vivere: attenzione che da un lato impone il riconoscimento della tradizione religiosa come qualcosa di vivo e operante, dall’altro richiede la conservazione e il rispetto della religiosità di ogni popolo nella sua tradizione, fatta salva la libertà di coscienza

    This study explores the conceptual evolution of secularism into a sustainable legal and political paradigm within European constitutionalism, guided by the influence of Robert Audi’s theory and the jurisprudence of the European Court of Human Rights (ECtHR). The transformation from confessional states to secular nations in Europe, marked notably by the institutional recognition of religious freedom as detailed in Article 9 of the European Convention on Human Rights, signifies a shift towards a “sustainable secularism.” This new model advocates for a dynamic balance between religious freedom, state neutrality, and social cohesion, adapting to societal changes while promoting equal respect and opportunities for all citizens, irrespective of their religious affiliations. The study discusses the implications of sustainable secularism for the legal frameworks of European states and in the industrial relations, emphasizing the need for laws and norms to maintain neutrality and avoid favoritism towards any religion or worldview, adopting instead a governance model that integrates and respects diverse religious perspectives within public life. Sustainable secularism, therefore, is positioned as a crucial, adaptable framework that ensures the ongoing relevance and effectiveness of secularism in contemporary society. It aims to uphold foundational values of freedom of conscience and equality, fostering a stable, pluralistic, and inclusive environment where diverse beliefs can coexist without competition. This framework not only prevents sectarianism but also promotes a public policy that respects individual rights and maintains formal equality, adapting to the unique requirements of different societal situations based on the principles of fundamental rights and human dignity
