    Effect of inducers against tobamovirus infection in tomato and bell Pepper

    Tomato and bell pepper seeds were treated with salicylic acid (50 mM), neem oil (5%) and Pseudomonas fluorescens (slurry). The seedlings were sprayed with salicylic acid (50 mM) and neem oil (5%). The concentration of Tomato mosaic tobamovirus (ToMV) and Tobacco mosaic tobamovirus (TMV) was assessed based on the number of local lesions on Nicotiana glutinosa. The results showed that the seed/seedling treatment with inducers reduced the number of local lesions when compared to untreated ones. Salicylic acid was an effective inducer

    Aqueous leaf extracts of Schefflera Venulosa and S. Wallichiana (araliaceae) protects the liver against carbon tetrachloride (ccl4) - induced hepatic damage in albino rats

    The hepatoprotective activity of the aqueous extracts of S. venulosa (AESV) and S. wallichiana (AESW) leaves on carbon tetrachloride (CCl4)-induced liver damage in albino rats was investigated. Animals were pretreated with the AESV (250 and 500 mg/kg body weight) and AESW (250 and 500 mg/kg body weight) for 15 days and then challenged with CCl4 (1 ml/kg body weight) in olive oil (1:1 v/v) on the 15th day. The degree of protection was measured by using biochemical parameters such as serum glutamate oxalate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), alkaline phosphatase (ALP), total protein and total bilirubin. Further the effects of both the extracts on catalase (CAT), peroxidase (Px) and superoxide dismutase (SOD) were estimated in both CCl4 treated and extract treated groups. Oral administration of AESV and AESW (500 mg/kg body weight) significantly protected from CCl4-induced elevation in SGOT, SGPT, ALP, total bilirubin, total protein and decrease in the activities of hepatic antioxidant enzymes namely SOD, CAT and Px. The extracts (AESV and AESW at 500 mg/kg body weight) also protected against histopathological damage induced by CCl4 such as distorted hepatocytes and necrosis. The present study suggests that AESV has potent antioxidant and hepatoprotective activity when compared to AESW

    Diversity studies on the endophytic fungi of Vitex negundo L

    Abstract A total of 143 endophytic fungal isolates were recovered from 1350 bark, twig and leaf segments of Vitex negundo L. which has important traditional medicinal value in the Indian system of medicine. The isolates belonged to ascomycetes (24%), coelomycetes (21%), hyphomycetes (51%) and mycelia sterilia (4%) groups. Lasiodiplodia sp. was the dominant endophyte in all the plant parts. The endophytic taxa were identified on the basis of morphology and DNA sequence data. Twig samples harboured higher number of endophytic taxa than the bark and leaf tissues of V. negundo. Phylogenetic relationships of the endophytic fungi were estimated from the sequences of the ITS region. Our studies indicate that diverse endophytic genera are grouped into six clades with respective fungal endophytes grouped in separate clades

    Squeezing of a coupled state of two spinors

    The notion of spin squeezing involves reduction in the uncertainty of a component of the spin vector below a certain limit. This aspect has been studied earlier for pure and mixed states of definite spin. In this paper, this study has been extended to coupled spin states which do not possess sharp spin value. A general squeezing criterion has been obtained by requiring that a direct product state for two spinors is not squeezed.     B\"acklund transformations for the second Painlev\'e hierarchy: a modified truncation approach

    The second Painlev\'e hierarchy is defined as the hierarchy of ordinary differential equations obtained by similarity reduction from the modified Korteweg-de Vries hierarchy. Its first member is the well-known second Painlev\'e equation, P2. In this paper we use this hierarchy in order to illustrate our application of the truncation procedure in Painlev\'e analysis to ordinary differential equations. We extend these techniques in order to derive auto-B\"acklund transformations for the second Painlev\'e hierarchy. We also derive a number of other B\"acklund transformations, including a B\"acklund transformation onto a hierarchy of P34 equations, and a little known B\"acklund transformation for P2 itself. We then use our results on B\"acklund transformations to obtain, for each member of the P2 hierarchy, a sequence of special integrals.Comment: 12 pages in LaTeX 2.09 (uses ioplppt.sty), to appear in Inverse Problem

    Expression of Regulatory Platelet MicroRNAs in Patients with Sickle Cell Disease

    Background: Increased platelet activation in sickle cell disease (SCD) contributes to a state of hypercoagulability and confers a risk of thromboembolic complications. The role for post-transcriptional regulation of the platelet transcriptome by microRNAs (miRNAs) in SCD has not been previously explored. This is the first study to determine whether platelets from SCD exhibit an altered miRNA expression profile. Methods and Findings: We analyzed the expression of miRNAs isolated from platelets from a primary cohort (SCD = 19, controls = 10) and a validation cohort (SCD = 7, controls = 7) by hybridizing to the Agilent miRNA microarrays. A dramatic difference in miRNA expression profiles between patients and controls was noted in both cohorts separately. A total of 40 differentially expressed platelet miRNAs were identified as common in both cohorts (p-value 0.05, fold change>2) with 24 miRNAs downregulated. Interestingly, 14 of the 24 downregulated miRNAs were members of three families - miR-329, miR-376 and miR-154 - which localized to the epigenetically regulated, maternally imprinted chromosome 14q32 region. We validated the downregulated miRNAs, miR-376a and miR-409-3p, and an upregulated miR-1225-3p using qRT-PCR. Over-expression of the miR-1225-3p in the Meg01 cells was followed by mRNA expression profiling to identify mRNA targets. This resulted in significant transcriptional repression of 1605 transcripts. A combinatorial approach using Meg01 mRNA expression profiles following miR-1225-3p overexpression, a computational prediction analysis of miRNA target sequences and a previously published set of differentially expressed platelet transcripts from SCD patients, identified three novel platelet mRNA targets: PBXIP1, PLAGL2 and PHF20L1. Conclusions: We have identified significant differences in functionally active platelet miRNAs in patients with SCD as compared to controls. These data provide an important inventory of differentially expressed miRNAs in SCD patients and an experimental framework for future studies of miRNAs as regulators of biological pathways in platelets. © 2013 Jain et al

    Nitrite augments tolerance to ischemia/reperfusion injury via the modulation of mitochondrial electron transfer

    Nitrite (NO2−) is an intrinsic signaling molecule that is reduced to NO during ischemia and limits apoptosis and cytotoxicity at reperfusion in the mammalian heart, liver, and brain. Although the mechanism of nitrite-mediated cytoprotection is unknown, NO is a mediator of the ischemic preconditioning cell-survival program. Analogous to the temporally distinct acute and delayed ischemic preconditioning cytoprotective phenotypes, we report that both acute and delayed (24 h before ischemia) exposure to physiological concentrations of nitrite, given both systemically or orally, potently limits cardiac and hepatic reperfusion injury. This cytoprotection is associated with increases in mitochondrial oxidative phosphorylation. Remarkably, isolated mitochondria subjected to 30 min of anoxia followed by reoxygenation were directly protected by nitrite administered both in vitro during anoxia or in vivo 24 h before mitochondrial isolation. Mechanistically, nitrite dose-dependently modifies and inhibits complex I by posttranslational S-nitrosation; this dampens electron transfer and effectively reduces reperfusion reactive oxygen species generation and ameliorates oxidative inactivation of complexes II–IV and aconitase, thus preventing mitochondrial permeability transition pore opening and cytochrome c release. These data suggest that nitrite dynamically modulates mitochondrial resilience to reperfusion injury and may represent an effector of the cell-survival program of ischemic preconditioning and the Mediterranean diet

    Characterization of xenotime from Datas (Brazil) as a potential reference material for in situ U-Pb geochronology

These crystals belong to a 300 g xenotime assortment, collected from alluvial deposits in SE Brazil. Electron microprobe and Laser Ablation‐Inductively Coupled Plasma‐Mass Spectrometry (LA‐ICP‐MS) analyses show that the selected crystals are internally homogeneous for most rare earth element, (REE, except some light REE) but are relatively heterogeneous for U and Th. The xenotime REE patterns are consistent with an origin from hydrothermal quartz veins in the Datas area that cut greenschist‐facies metasediments and that locally contain other accessory phases such as rutile and monazite. High‐precision U‐Pb Isotope Dilution‐Thermal Ionization Mass Spectrometry (ID‐TIMS) analyses showed slight age heterogeneity for the XN01 crystal not observed in the XN02 sample. The two crystals have slightly different average 206Pb/238U ages of 513.4 ± 0.5 Ma (2 s) and 515.4 ± 0.2 Ma (2 s), respectively. In situ U‐Pb isotope data acquired via LA‐(Q,SF,MC)‐ICP‐MS are within the uncertainty of the ID‐TIMS data, showing homogeneity at the 1% precision of the laser ablation (and probably ion microprobe) technique. U‐Pb LA‐(MC, SF)‐ICP‐MS analyses, using XN01 as a primary RM, reproduced the ages of other established RMs within less than 1% deviation. Other Datas crystals (XN03‐05) also display a reproducibility in Pb/U dates better than 1% on LA‐ICP‐MS, making them good candidates for further testing by ID‐TIMS.https://agupubs.onlinelibrary.wiley.com/doi/10.1029/2017GC007412Publisher's versio
