    A 66-year-old man presented with subacute cough and worsening dyspnea. Labs were notable for moderate peripheral eosinophilia, and computed tomography (CT) scan demonstrated extensive crazy-paving throughout bilateral upper lung fields. Bronchoalveolar lavage (BAL) revealed macrophages with lipid-filled vacuoles and negative periodic acid-Schiff (PAS) stain. Further history obtained from the patient and family was notable for daily application of commercially available vapor rub to nares and intentional deep inhalation of nebulized fluids containing scented oils. The patient was diagnosed with exogenous lipoid pneumonia through an unusual route of lipid administration

    poster abstractArt, Race, Space is a collaborative research project that takes as its starting point E Pluribus Unum, a public art installation proposed for the Indianapolis Culture Trail by renowned artist Fred Wilson that was cancelled in 2011 due to controversy surrounding Wilson’s appropriation of a freed slave figure from the Soldiers and Sailors Monument. Art, Race, Space” goes beyond examining the visual legacies of racial bondage to explore how the public responses to sculptures, memorials, and archaeology reveal our society’s faultlines of race and inequality. Building on the ideas about race, class, visual culture, and democratic debate that emerge from the Indianapolis project, the faculty have designed a multifaceted program to advance scholarship and promote civic dialogue about these significant issues. The faculty members organized an interdisciplinary symposium in January, 2013. Supported by an IAHI grant, the symposium explored the complicated relationships between art, race, and civic space with presentations by Wilson, community representatives who supported and opposed the sculpture, and scholars from a variety of disciplines who examined historical and cultural contexts of the controversy that had revealed Indianapolis’ longstanding racial and class tensions. The dialogue was expanded with the presentation of historical and contemporary examples from other parts of the United States. In order to encourage public dialogue, the symposium provided opportunities for audience members and presenters to engage in conversations, and it deployed social media (Twitter and Facebook) to encourage broader participation. The project's goal is to further scholarship and encourage public conversation on race and materiality. To this end the faculty have created a website, a Facebook page, Twitter account, and are working on an open-access curriculum to support dialogue in schools and informal learning settings about the complex issues of art, race, and representation. The faculty are also collaborating on academic publications, including selected proceedings and an article on the symposium's "hybrid discourse" that combined university and community resources, expertise, and communication practices and brought together diverse voices in constructive conversation about the challenging issues surrounding E Pluribus Unum

    IDENTIFICATION OF POTENTIAL TARGETS OF THE GRR1P SCF UBIQUITIN LIGASE IN FUNGI

    The opportunistic human pathogen Candida albicans causes both superficial and life-threatening systemic infections and is a leading cause of fungal disease in immunocompromised individuals.  C. albicans can grow in different cell shapes, or morphologies, including yeast-like cells and a variety of filamentous forms, such as true hyphae and pseudohyphae.   Yeast, hyphae and pseudohyphae have been observed at the sites of Candida infection and there is strong evidence that morphogenesis, the transition between yeast and filamentous growth forms, is essential for virulence. Several studies have implicated ubiquitin-dependent proteolysis in the regulation of morphogenesis, yet the mechanism by which this pathway does so is largely unknown.  Previously, we have shown that deletion of the GRR1 gene results in the constitutive formation of filamentous growth forms.  The Grr1 protein is a component of an SCF ubiquitin ligase system that selectively targets proteins for degradation.  Thus, the loss of Grr1-mediated proteolysis presumably leads to the aberrant accumulation, and inappropriate activity, of a protein or proteins that induce filamentous growth.  The spectrum of proteins targeted for degradation by Grr1 is not known.  The goal of this project is to identify Grr1 targets in Saccharomyces cerevisiae, an experimentally tractable model system for pathogenic fungi.  We are using a novel proteomics-based approach to isolate and characterize proteins that are ubiquitinated in a Grr1-dependent fashion. The successful identification of Grr1p targets will be important for developing a working model of the pathways involved in the yeast to filamentous growth transition in pathogenic fungi

    Visualization of actin filaments and monomers in somatic cell nuclei

The function and form (monomer, filament, or noncanonical oligomer) of nuclear actin are hotly debated, and its localization and dynamics are largely unknown. To determine the distribution of nuclear actin in live somatic cells and evaluate its potential functions, we constructed and validated fluorescent nuclear actin probes. Monomeric actin probes concentrate in nuclear speckles, suggesting an interaction of monomers with RNA-processing factors. Filamentous actin probes recognize discrete structures with submicron lengths that are excluded from chromatin-rich regions. In time-lapse movies, these actin filament structures exhibit one of two types of mobility: 1) diffusive, with an average diffusion coefficient of 0.06–0.08 μm2/s, or (2) subdiffusive, with a mobility coefficient of 0.015 μm2/s. Individual filament trajectories exhibit features of particles moving within a viscoelastic mesh. The small size of nuclear actin filaments is inconsistent with a role in micron-scale intranuclear transport, and their localization suggests that they do not participate directly in chromatin-based processes. Our results instead suggest that actin filaments form part of a large, viscoelastic structure in the nucleoplasm and may act as scaffolds that help organize nuclear contents.This bulk of this work was supported by a grant from the National Institutes of Health to R.D.M.     Tooth Whitening Effects on Bracket Bond Strength In Vivo

    Objective: To test the hypothesis that there is no difference between the bracket survival rate of brackets bonded to bleached and unbleached teeth. Materials and Methods: Thirty-eight patients who required comprehensive orthodontic treatment were included in the study. A split mouth technique was used with one arch exposed to in-office whitening gel containing 38% hydrogen peroxide for 30 minutes, while the unbleached arch served as the control. Patients were divided into two groups: Brackets bonded within 24 hours after bleaching and brackets bonded 2–3 weeks after bleaching. The bracket survival rate was computed using the log-rank test (Kaplan-Meier Analysis). Results: A significantly higher rate of bracket failure was found with bleached teeth (16.6%) compared with unbleached teeth (1.8%) after 180 days. Brackets bonded within 24 hours of bleaching resulted in significantly higher clinical failure (14.5%) compared with those bonded after 3 weeks (2.1%). Adhesive Remnant Index scores of failed brackets revealed that the majority of failure in bleached teeth occurred in the enamel/resin interface. Conclusions: The hypothesis was rejected. Brackets bonded within 24 hours after bleaching have a significantly higher risk for bond failure. Orthodontic bonding should be delayed for 2–3 weeks if patients have a history of in-office bleaching with 38% hydrogen peroxide
