    Moving Away from Understanding: Personal Therapy in Contemporary Doctoral Education

    Personal therapy has never been a training requirement for clinical psychology in the American Psychological Association (APA). This became more evident when the field of Clinical Psychology at the Boulder Conference over 30 years ago in which personal therapy was omitted as a requirement of doctoral education because there was a lack of sufficient empirical evidence to support such a mandate in the United States. While there has been research in the field of psychodynamic theory on personal therapy, the stance taken during the Boulder Conference (and to some extent the Chicago and Vail Conferences thereafter) continues to be the standard in regards to the training of psychologists. This has resulted in new psychologists having limited understanding of themselves and, at times, difficult or impaired relationships with their clients. This dissertation will look at the original psychoanalytic writings on training analysis and will use these points to explain the more recent literature on the utility of personal therapy, particularly in countries that still require it. In addition to these main points, the arguments against personal therapy will also be included to help showcase both sides of the issue. Using three major studies on personal therapy, this dissertation will use the themes found in the literature review to see if there is still a justification for not requiring personal therapy, as well as providing personal opinions on why the field has moved away from self-understanding and towards a predominantly outcome-focused approach to psychotherapy

    Human and Machine Learning in Non-Markovian Decision Making

    Humans can learn under a wide variety of feedback conditions. Reinforcement learning (RL), where a series of rewarded decisions must be made, is a particularly important type of learning. Computational and behavioral studies of RL have focused mainly on Markovian decision processes, where the next state depends on only the current state and action. Little is known about non-Markovian decision making, where the next state depends on more than the current state and action. Learning is non-Markovian, for example, when there is no unique mapping between actions and feedback. We have produced a model based on spiking neurons that can handle these non-Markovian conditions by performing policy gradient descent. Here, we examine the model’s performance and compare it with human learning and a Bayes optimal reference, which provides an upper-bound on performance. We find that in all cases, our population of spiking neurons model well-describes human performance

    Linking perceptual learning with identical stimuli to imagery perceptual learning

    Perceptual learning is usually thought to be exclusively driven by the stimuli presented during training (and the underlying synaptic learning rules). In some way, we are slaves of our visual experiences. However, learning can occur even when no stimuli are presented at all. For example, Gabor contrast detection improves when only a blank screen is presented and observers are asked to imagine Gabor patches. Likewise, performance improves when observers are asked to imagine the non-existing central line of a bisection stimulus to be offset either to the right or left. Hence, performance can improve without stimulus presentation. As shown in the auditory domain, performance can also improve when the very same stimulus is presented in all learning trials and observers were asked to discriminate differences which do not exist (observers were not told about the set up). Classic models of perceptual learning cannot handle these situations since they need proper stimulus presentation, i.e., variance in the stimuli, such as a left vs. right offset in the bisection stimulus. Here, we show that perceptual learning with identical stimuli occurs in the visual domain, too. Second, we linked the two paradigms by telling observers that only the very same bisection stimulus was presented in all trials and asked them to imagine the central line to be offset either to the left or right. As in imagery learning, performance improved

    Trametinib-Induced Epidermal Thinning Accelerates a Mouse Model of Junctional Epidermolysis Bullosa

    Junctional epidermolysis bullosa (JEB) patients experience skin and epithelial fragility due to a pathological deficiency in genes associated with epidermal adhesion. Disease severity ranges from post-natal lethality to localized skin involvement with persistent blistering followed by granulation tissue formation and atrophic scarring. We evaluated the potential of utilizing Trametinib, an MEK inhibitor previously shown to target fibrosis, with and without the documented EB-anti-fibrotic Losartan for reducing disease severity in a mouse model of JEB; Lamc2jeb mice. We found that Trametinib treatment accelerated disease onset and decreased epidermal thickness, which was in large part ameliorated by Losartan treatment. Interestingly, a range of disease severity was observed in Trametinib-treated animals that tracked with epidermal thickness; those animals grouped with higher disease severity had thinner epidermis. To examine if the difference in severity was related to inflammation, we conducted immunohistochemistry for the immune cell markers CD3, CD4, CD8, and CD45 as well as the fibrotic marker αSMA in mouse ears. We used a positive pixel algorithm to analyze the resulting images and demonstrated that Trametinib caused a non-significant reduction in CD4 expression that inversely tracked with increased fibrotic severity. With the addition of Losartan to Trametinib, CD4 expression was similar to control. Together, these data suggest that Trametinib causes a reduction in both epidermal proliferation and immune cell infiltration/proliferation, with concurrent acceleration of skin fragility, while Losartan counteracts Trametinib’s adverse effects in a mouse model of JEB

    The Right to Counsel in Illinois: Evaluation of Adult Criminal Trial-Level Indigent Defense Services

    This study shows there are two overarching reasons why the State of Illinois is defaulting on its constitutional right to counsel obligations. First, the state requires counties and courts to provide and predominantly fund indigent defense systems in a way that bakes in governmental interference with the right to counsel. Second, as one of only seven states with no state-level mechanism to oversee any aspect of trial-level right to counsel services, Illinois lacks information about every aspect of the varied indigent defense systems implemented by the county governments and courts in their efforts to fulfill the Sixth Amendment right to counsel responsibilities that the state has delegated to them. There is a path forward, and it is important for Illinois to get this right

    APRIL, a New Ligand of the Tumor Necrosis Factor Family, Stimulates Tumor Cell Growth

    Members of the tumor necrosis factor (TNF) family induce pleiotropic biological responses, including cell growth, differentiation, and even death. Here we describe a novel member of the TNF family designated APRIL (for a proliferation-inducing ligand). Although transcripts of APRIL are of low abundance in normal tissues, high levels of mRNA are detected in transformed cell lines, and in human cancers of colon, thyroid, and lymphoid tissues in vivo. The addition of recombinant APRIL to various tumor cells stimulates their proliferation. Moreover, APRIL-transfected NIH-3T3 cells show an increased rate of tumor growth in nude mice compared with the parental cell line. These findings suggest that APRIL may be implicated in the regulation of tumor cell growth
