    Combining the pharmacophore features of coumarins and 1,4-substituted 1,2,3-triazoles to design new acetylcholinesterase inhibitors : fast and easy generation of 4-methylcoumarins/1,2,3-triazoles conjugates via Click Chemistry

    Coumarins are a large class of compounds that display a range of interesting biological properties, being considered privileged structures because of the ability of their 2H-chromen-2-one nuclei to bind to multiple pharmacological targets. We hypothesized that the linkage of a second pharmacophore nucleus to the 2H-chromen-2-one core, the 1,2,3-triazole moiety, would entail more selective and pharmacologically active coumarins. Therefore, we describe the synthesis of fourteen 4-methylcoumarins/1,4-substituted 1,2,3-triazole conjugates, which were predicted by in silico methods to inhibit acetylcholinesterase (AChE) and proved to be moderate in vitro inhibitors of this enzyme. Molecular docking simulations suggest that the most active of these compounds has a putative binding mode similar to donepezil, both occupying the peripheral anionic site of AChE, which is associated with the secondary noncholinergic functions of the enzyme. This highlights the potential of this series for further optimization in the search of new coumarins for the treatment of Alzheimer’s disease

Centro de Ciencias Fisicas e MatematicasDescreve-se a síntese de cristais líquidos ferroelétricos e precursores quirais derivados do reagente (S)-(-)-lactato de etila, bem como as propriedades mesomórficas de séries homólogas sintetizadas. Foram também sintetizadas duas séries homólogas de cristais líquidos derivados do reagente quiral (S)-(-)-2-metil-1-butanol. As rotas de síntese descritas e executadas combinam simplicidade, produtos de fácil purificação, materiais de partida acessíveis, baixos custos e apresentam rendimentos ópticos e químicos adequados. A importância destas novas metodologias sintéticas desenvolvidas foi ampliada no sentido de obtenção de novos materiais com propriedades mesogênicas e ferroelétricas.     An old dog with new tricks : schiff bases for liquid crystals materials based on isoxazolines and isoxazoles

    Schiff bases are in action again showing an exuberant mesophase range despite their chemical and thermal weakness. Two series of Schiff bases (SB), based on isoxazolines 9 a–f and isoxazoles 10 a–f, are described. 9 a–f were synthesized by a [3+2] cycloaddition 1,3-dipolar of arylnitrile oxide from p-nitrobenzaldehyde (1) and alkenes. And, subsequently oxidized to 10 a–f by MnO2. The isoxazolines and isoxazoles thus obtained were reduced to aniline derivatives and condensed with appropriated arylaldehydes. The SBs series isoxazolines 9 a–f presented a narrow mesophase range, while SBs series isoxazoles 10 a–f showed a large mesophase range. Nematic mesophase was observed for SBs with short and non-polar groups, while SmA and SmC mesophase for SBs with long and more polar terminal groups. The stability of the SBs is also dependent on the clearing temperature. For 10 a–f with high clearing temperature, decomposition induced by heat was observed during the first cycle of heating and in solutions of CDCl3. Series 9 a–f was more resistant to the thermal decomposition
