    Biomarkers of aging in HIV: inflammation and the microbiome

    Purpose: HIV-infected subjects present increased levels of inflammatory cytokines and T cell activation in the peripheral blood despite suppressive combination antiretroviral therapy which renders them susceptible to premature aging. The purpose of the present work was to review existing evidence on the ways in which the anatomical and microbiological abnormalities of the gastrointestinal tract can represent a major cause of organ disease in HIV infection. Methods: We conducted a systematic review of the Pubmed database for articles published from 2014 to 2018. We included studies on inflammatory/activation biomarkers associated with cardiovascular and bone disease, neurocognitive impairment and serious non-AIDS events in HIV-infected subjects. We also included researches which linked peripheral inflammation/activation to the anatomical, immune and microbiological alterations of the gastrointestinal tract. Results: Recent literature data confirm the association between non-infectious comorbidities and inflammation in HIV infection which may be driven by gastrointestinal tract abnormalities, specifically microbial translocation and dysbiosis. Furthermore, there is mounting evidence on the possible role of metabolic functions of the microbiota in the pathogenesis of premature aging in the HIV-infected population. Conclusions: Biomarkers need to be validated for their use in the management of HIV infection. Compounds which counteract microbial translocation, inflammation and dysbiosis have been investigated as alternative therapeutic strategies in viro-suppressed HIV-infected individuals, but appear to have limited efficacy, probably due to the multifactorial pathogenesis of non-infectious comorbidities in this setting

    On a Low-Frequency and Contrast Stabilized Full-Wave Volume Integral Equation Solver for Lossy Media

    In this article, we present a new regularized electric flux volume integral equation (D-VIE) for modeling high-contrast conductive dielectric objects in a broad frequency range. This new formulation is particularly suitable for modeling biological tissues at low frequencies, as it is required by brain epileptogenic area imaging, but also at higher ones, as it is required by several applications, including, but not limited to, deep brain stimulation (DBS). When modeling inhomogeneous objects with high complex permittivities at low frequencies, the traditional D-VIE is ill-conditioned and suffers from numerical instabilities that result in slower convergence and less accurate solutions. In this work, we address these shortcomings by leveraging a new set of volume quasi-Helmholtz projectors. Their scaling by the material permittivity matrix allows for the rebalancing of the equation when applied to inhomogeneous scatterers and, thereby, makes the proposed method accurate and stable even for high complex permittivity objects until arbitrarily low frequencies. Numerical results, canonical and realistic, corroborate the theory and confirm the stability and the accuracy of this new method both in the quasi-static regime and at higher frequencies

    Synchrotron radiation μ X-ray diffraction in transmission geometry for investigating the penetration depth of conservation treatments on cultural heritage stone materials

    The assessment of the penetration depth of conservation treatments applied to cultural heritage stone materials is a burning issue in conservation science. Several analytical approaches have been proposed but, at present, many of them are not fully exhaustive to define in a direct way the composition and location of the conservation products formed after inorganic mineral treatments. Here, we explored, for the first time, the analytical capability of synchrotron radiation m X-ray diffraction in transmission geometry (SR-mTXRD) for the study of the crystal chemistry and penetration depth of the consolidating phases formed after the application of diammonium hydrogen phosphate (DAP) treatments on a porous carbonatic stone (Noto limestone). The SR-mTXRD approach provided unambiguous information on the nature of the newly formed calcium phosphates (hydroxyapatite, HAP, and octacalcium phosphate, OCP) with depth, supplying important indications of the diffusion mechanism and the reactivity of the substrate. Qualitative and semi-quantitative data were obtained at the microscale with a non-destructive protocol and an outstanding signal-to-noise ratio. The SR-mTXRD approach opens a new analytical scenario for the investigation of a wide range of cultural heritage materials, including natural and artificial stone materials, painted stratigraphies, metals, glasses and their decay products. Furthermore, it can potentially be used to characterize the penetration depth of a phase \u201cA\u201d (or more crystalline phases) in a matrix \u201cB\u201d also beyond the cultural heritage field, demonstrating the potential wide impact of the study

    On a Fast Solution Strategy for a Surface-Wire Integral Formulation of the Anisotropic Forward Problem in Electroencephalography

    This work focuses on a quasi-linear-in-complexity strategy for a hybrid surface-wire integral equation solver for the electroencephalography forward problem. The scheme exploits a block diagonally dominant structure of the wire self block— that models the neuronal fibers self interactions—and of the surface self block—modeling interface potentials. This structure leads to two Neumann iteration schemes further accelerated with adaptive integral methods. The resulting algorithm is linear up to logarithmic factors. Numerical results confirm the performance of the method in biomedically relevant scenarios

    Grazing incidence synchrotron X-ray diffraction of marbles consolidated with diammonium hydrogen phosphate treatments : non-destructive probing of buried minerals

    Diammonium hydrogen phosphate (DAP)-based consolidating treatments react with carbonatic stones and form calcium phosphates phases, whose composition depends on the availability of free calcium ions. In this work, an innovative non-destructive approach based on grazing incidence X-ray diffraction (GIXRD) with synchrotron radiation (SR) is used to investigate DAP-treated Carrara marble specimens and to study the influence of the substrate composition on the crystallization of calcium phosphate phases. The outcomes indicate that the presence of compositional micro-heterogeneity of Carrara marble favours the formation of specific phases. Dicalcium phosphate dihydrate, a calcium phosphate with a low Ca/P molar ratio, is formed on carbonatic phases with a low Ca amount, such as dolomite grains and Mg-containing veins. Furthermore, this study highlights the potentialities of SR-GIXRD as a powerful non-destructive tool for the diagnostic of Cultural Heritage objects since it allows investigating the conservation history of stone materials and their interaction with the environment

    Conformal invariance studies of the Baxter-Wu model and a related site-colouring problem

    The partition function of the Baxter-Wu model is exactly related to the generating function of a site-colouring problem on a hexagonal lattice. We extend the original Bethe ansatz solution of these models in order to obtain the eigenspectra of their transfer matrices in finite geometries and general toroidal boundary conditions. The operator content of these models are studied by solving numerically the Bethe-ansatz equations and by exploring conformal invariance.     Synthesis of calcium oxalate trihydrate: new data by vibrational spectroscopy and synchrotron X-ray diffraction

    Calcium oxalate is found in nature in three different crystalline states determined by the number of H2O in the unit formula (whewellite CaC2O4\ub7H2O, COM; weddellite CaC2O4\ub7(2 + x)H2O, COD and caoxite CaC2O4\ub73H2O, COT). The properties of these materials are relevant in the field of biomedicine, cultural heritage and mineralogy. In two previous papers, we have used X-ray diffraction and vibrational spectroscopy (infrared and Raman) to derive information on crystal and molecular structures of COM and COD. In this paper, we complete the synthesis and analysis on the third form, COT, and present a comparative study of the data collected from the three crystalline states. The experiments clearly highlight the role played by the H2O molecules linked within the structure by different kinds of hydrogen bonds. The vibrational assignment of the infrared and Raman bands are critically proposed. The fact relevant for the work in biomedicine, cultural heritage and crystallography is that a simple examination of the spectra allows quickly to determine the chemical nature of the material in an unknown sample even in a minute quantity or in awkward experimental conditions

    Body composition, muscle strength, and physical function of patients with Bethlem myopathy and Ullrich congenital muscular dystrophy.

    Objective. To determine the contributions of body mass, adiposity, and muscularity to physical function and muscle strength in adult patients with Bethlem myopathy (BM) and Ullrich congenital muscular dystrophy (UCMD). Materials and Methods. Evaluation involved one UCMD and 7 BM patients. Body composition was determined by body mass index (BMI) and dual-energy-X-ray-absorptiometry (DXA), muscle strength by dynamometry, physical function by the distance walked in 6 minutes (6MWD), forced vital capacity (FVC) by a spirometer. Results. Six participants were of normal weight and 2 overweight based on BMI; all were sarcopenic based on appendicular fat free mass index (AFFMI); and 7 were sarcopenic obese based on AFFMI and % fat mass. Average muscle strength was reduced below 50% of normal. The 6MWD was in BM patients 30% less than normal. FVC was reduced in 4 of the BM patients. Muscle strength had a good correlation with the physical function variables. Correlation between muscle strength and BMI was poor; it was very high with AFFMI. AFFMI was the best single explicator of muscle strength and physical function. Conclusion. Muscle mass determined by DXA explains most of the variability of the measures of muscle strength and physical function in patients with BM and UCMD

    Doxorubicin and congo red effectiveness on prion infectivity in golden Syrian hamster

    The effect of doxorubicin and Congo Red on prion protein (PrP) infectivity in experimental scrapie was studied to better understand the effect of these compounds in prion diseases and to establish whether a dose-response correlation exists for Congo Red. This was performed in order to test the effectiveness of compounds that may easily be used in human prion diseases. Brain homogenate containing membrane bound PrPSc monomers was used as inoculum and was previously incubated with doxorubicin 10(-3) M and with increasing concentrations of Congo Red ranging from 10(-7) to 10(-2) M. This study shows for the first time that doxorubicin, and confirms that Congo Red, may interact with pathological PrP monomers modifying their infectious properties. Pre-incubation of infected brain homogenate with Congo Red resulted in prolonged incubation time and survival, independently of Congo Red concentration (p<0.05). Doxorubicin and Congo Red effects do not depend upon interaction with PrP amyloid material
