    Surface crack growth in fiber composites

    The results of an experimental study of damage extension and failure in glass and graphite/epoxy laminates containing partially through-thickness surface cracks are presented. The laminates studied are divided between those containing four plies, 90/0/0/90, 15/-15/-15/15, and 45/-45/-45/45, and those containing 12-16 plies of the general configurations 0/90, + or - 45, and 0/+ or - 60. Most of the results are for surface cracks of various lengths and several depths. Stable damage extension in laminates containing surface cracks is predominantly delamination between plies, and tends to be much more extensive prior to failure than is the case with through-thickness cracks, resulting in approximately notch-insensitive behavior in most cases. A greater tendency for notch-sensitive behavior is found for 0/90 graphite/epoxy laminates for which stable damage extension is more limited. The rate of damage extension with increasing applied stress depends upon the composite system and ply configuration as well as the crack length and depth. An approximate semiempirical method is presented for estimating the growth rate of large damage-regions

    A general procedure for software cost estimation in any environment is outlined. The basic concepts of work and effort estimation are explained, some popular resource estimation models are reviewed, and the accuracy of source estimates is discussed. A software cost prediction procedure based on the experiences of the Software Engineering Laboratory in the flight dynamics area and incorporating management expertise, cost models, and historical data is described. The sources of information and relevant parameters available during each phase of the software life cycle are identified. The methodology suggested incorporates these elements into a customized management tool for software cost prediction. Detailed guidelines for estimation in the flight dynamics environment developed using this methodology are presented

    A cell-permeable biscyclooctyne as a novel probe for the identification of protein sulfenic acids

    Reactive oxygen species act as important second messengers in cell signaling and homeostasis through the oxidation of protein thiols. However, the dynamic nature of protein oxidation and the lack of sensitivity of existing molecular probes have hindered our understanding of such reactions; therefore, new tools are required to address these challenges. We designed a bifunctional variant of the strained bicyclo[6.1.0]nonyne (BCN-E-BCN) that enables the tagging of intracellular protein sulfenic acids for biorthogonal copper-free click chemistry. In validation studies, BCN-E-BCN binds the sulfenylated form of the actin-severing protein cofilin, while mutation of the cognate cysteine residues abrogates its binding. BCN-E-BCN is cell permeable and reacts rapidly with cysteine sulfenic acids in cultured cells. Using different azide-tagged conjugates, we demonstrate that BCN-E-BCN can be used in various applications for the detection of sulfenylated proteins. Remarkably, cycloaddition of an azide-tagged fluorophore to BCN-E-BCN labelled proteins produced in vivo can be visualized by fluorescence microscopy to reveal their subcellular localization. These findings demonstrate a novel and multifaceted approach to the detection and trapping of sulfenic acids

    Rapid quenching effects in PVC films

    Using a specially constructed microbalance for hydrostatic weighing, density changes in PVC thin films (with no additives, 30-100 micrometers thick), due to rapid quenching (approximately 300 C/sec) through the glass transition temperature, have been observed. The more severe the quench, the greater is the free volume content. Isobaric volume recovery of PVC has also been studied by volume dilatometry. Both show aging of relaxing molecular rearrangements takes place as a linear function of logarithmic aging time at room temperature. Distribution of retardation times and Primak's distributed activation energy spectra have been applied to the volume recovery data. The concomitant changes in mechanical properties of PVC after quenching have been monitored by tensile creep and stress-strain to failure. All reflect the presence of excess free volume content, due to rapid quenching
