    Spatial and Temporal Variation in the Distribution of Fishes of the LaBranche Wetlands Area of the Lake Pontchartrain Estuary, Louisiana

    The LaBranche Wetlands are located along the southwestern shore of Lake Pontchartrain. During early summer of 1989 and 1990, an electroflshing survey of 6 stations In Bayou LaBranche and Bayou Trepagnier, a tributary, yielded 10,644 specimens representing 38 species in 19 families. All fishes were typical of brackish to freshwater, low gradient systems of the Gulf Coastal Plain. Members of the Lepisosteidae, Clupeidae, Fundulidae, Poecillidae and Centrarchidae were numerical dominants. Results from rotenone samples at five of the same stations by Louisiana Department of Environmental Quality in 1986, and net samples at one common site by Thompson and Verret (1980) bring the total number of species for the LaBranche Wetlands to 52. Differences in the physicochemical characters of the two bayou systems were reflected in the dissimilar faunas between the two bayous. Results from detrended correspondence analysis (DCA) and community similarity indices indicate a high degree of variance at sites within the same bayou system over time

    Nonnative structure in a peptide model of the unfolded state of SOD1: Implications for ALS-linked aggregation

    Dozens of mutations throughout the sequence of the gene encoding superoxide dismutase 1 (SOD1) have been linked to toxic protein aggregation in the neurodegenerative disease amyotrophic lateral sclerosis (ALS). A parsimonious explanation for numerous genotypes resulting in a common phenotype would be mutation-induced perturbation of the folding free-energy surface that increases the populations of high-energy states prone to aggregation. The absence of intermediates in the folding of monomeric SOD1 suggests that the unfolded ensemble is a potential source of aggregation. To test this hypothesis, here we dissected SOD1 into a set of peptides end-labeled with FRET probes to model the local behavior of the corresponding sequences in the unfolded ensemble. Using time-resolved FRET, we observed that the peptide corresponding to the loop VII-beta8 sequence at the SOD1 C-terminus was uniquely sensitive to denaturant. Utilizing a two-dimensional form of maximum entropy modeling, we demonstrate that the sensitivity to denaturant is the surprising result of a two-state-like transition from a compact to an expanded state. Variations of the peptide sequence revealed that the compact state involves a nonnative interaction between the disordered N-terminus and the hydrophobic C-terminus of the peptide. This nonnative intramolecular structure could serve as a precursor for intermolecular association and result in aggregation associated with ALS. We propose that this precursor would provide a common molecular target for therapeutic intervention in the dozens of ALS-linked SOD1 mutations

    Passive, broadband and low-frequency suppression of laser amplitude noise to the shot-noise limit using hollow-core fibre

    We use hollow-core fibre to preserve the spectrum and temporal profile of picosecond laser pulses in CBD to suppress 2.6 dB of amplitude noise at MHz noise frequencies, to within 0.01 dB of the shot-noise limit. We provide an enhanced version of the CBD scheme that concatenates circuits to suppress over multiple frequencies and over broad frequency ranges --- we perform a first demonstration that reduces total excess amplitude noise, between 2 - 6 MHz, by 85%. These demonstrations enable passive, broad-band, all-guided fibre laser technology operating at the shot-noise limit.Comment: 8 pages, 8 figure

    Education Differences in Life Expectancy With Cognitive Impairment

    Background. Low education has an impact on life expectancy and level of cognition, but little is known on its effect on life expectancy with cognitive impairment. Methods. The Medical Research Council Cognitive Function and Ageing Study (MRC CFAS) collected population-based longitudinal data on people aged 65 years and older including measures of education and cognitive impairment, using the Mini-Mental State Examination (MMSE), for five geographically diverse areas around England and Wales interviewed between 1991 and 2003. Transitions between health states were calculated using Markov chain methods. Life expectancy in different states of cognitive function as measured by MMSE were further explored for different education groups. The effect of fixed and educationally based cut points for cognitive impairment are investigated. Results. Life expectancy spent with cognitive impairment is fairly constant with increasing age at around 1.4 years in men and 2.5 years in women, though this reflects a large increase in the proportion of life spent with cognitive impairment. The differences seen between education groups for the proportion of total life with cognitive impairment (men 13% and women 22% of life lived for low education vs men 7% and women 12% in high education group) disappear when education-adjusted cut points are used (10% in men and 17% in women at age 65 for all education groups). Conclusions. The results show that there is a substantial amount of life expectancy with cognitive impairment in both men and women. The impairment burden is just as great for those with high education as the lowest educated group. © 2009 The Author(s)

    Folding of the RNA recognition motif (RRM) domains of the amyotrophic lateral sclerosis (ALS)-linked protein TDP-43 reveals an intermediate state

    Pathological alteration of TDP-43 (TAR DNA-binding protein-43), a protein involved in various RNA-mediated processes, is a hallmark feature of the neurodegenerative diseases amyotrophic lateral sclerosis and frontotemporal lobar degeneration. Fragments of TDP-43, composed of the second RNA recognition motif (RRM2) and the disordered C terminus, have been observed in cytoplasmic inclusions in sporadic amyotrophic lateral sclerosis cases, suggesting that conformational changes involving RRM2 together with the disordered C terminus play a role in aggregation and toxicity. The biophysical data collected by CD and fluorescence spectroscopies reveal a three-state equilibrium unfolding model for RRM2, with a partially folded intermediate state that is not observed in RRM1. Strikingly, a portion of RRM2 beginning at position 208, which mimics a cleavage site observed in patient tissues, increases the population of this intermediate state. Mutually stabilizing interactions between the domains in the tethered RRM1 and RRM2 construct reduce the population of the intermediate state and enhance DNA/RNA binding. Despite the high sequence homology of the two domains, a network of large hydrophobic residues in RRM2 provides a possible explanation for the increased stability of RRM2 compared with RRM1. The cluster analysis suggests that the intermediate state may play a functional role by enhancing access to the nuclear export signal contained within its sequence. The intermediate state may also serve as a molecular hazard linking productive folding and function with pathological misfolding and aggregation that may contribute to disease

    Quantum walks of correlated photon pairs in two-dimensional waveguide arrays

    We demonstrate quantum walks of correlated photons in a 2D network of directly laser written waveguides coupled in a 'swiss cross' arrangement. The correlated detection events show high-visibility quantum interference and unique composite behaviour: strong correlation and independence of the quantum walkers, between and within the planes of the cross. Violations of a classically defined inequality, for photons injected in the same plane and in orthogonal planes, reveal non-classical behaviour in a non-planar structure.Comment: 5 pages, 5 figure
