Quantitative analysis of the dystrophin gene by real-time PCR
Duchenne and Becker muscular dystrophy (DMD/BMD) are severe X-linked neuromuscular disorders caused by mutations in the dystrophin gene. Our aim was to optimize a quantitative real-time PCR method based on SYBR® Green I chemistry for routine diagnostics of DMD/BMD deletion carriers. Twenty female relatives of DMD/BMD patients with previously detected partial gene deletions were studied. The relative quantity of the target exons was calculated by a comparative threshold cycle method (ΔΔCt). The carrier status of all subjects was successfully determined. The gene dosage ratio for non-carriers was 1.07±0.20, and for carriers 0.56±0.11. This assay proved to be simple, rapid, reliable and cost-effective

Owocem tych zainteresowań, współpracy i badań własnych są prezentowane
tu artykuły, których tematykę można ująć w pięć bloków zagadnień.
Pierwszy z nich dotyczy czynników rozwoju gospodarczego zarówno
w aspekcie mikroekonomicznym, takim jak przedsiębiorczość, kapitał intelektualny,
wzrost wydajności pracy, jak i makroekonomicznym, takim jak systemy
prawne, podatkowe, dotacje, uwarunkowania społeczne, infrastrukturalne
i technologiczne.
Drugi blok, prezentuje zagadnienia, które nabierają obecnie coraz większego
znaczenia, mimo iż pozornie tylko pośrednio wpływają na rozwój gospodarczy.
Filozofia, etyka, umiejętność rozwiązywania konfliktów staje się
dziś kluczem do skutecznego zarządzania, ale tylko takiego, które zostanie
zaakceptowane w wymiarze osobowym, społecznym, kulturowym, międzynarodowym.
Trzeci blok dotyka zagadnień z dziedziny finansów. Rozwój tego rynku
daje szerokie możliwości wykorzystania produktów i usług finansowych nie
tylko dużym przedsiębiorstwom i instytucjom, ale także gospodarstwom domowym.
Zaawansowane metody ekonometryczne dają z kolei szanse na wielowymiarową
i specjalistyczną analizę zjawisk finansowych, ułatwiając ich
zrozumienie i podejmowanie decyzji w tym obszarze. Czwarty blok obejmuje opracowania koncentrujące się na systemach
ułatwiających funkcjonowanie i rozwój przedsiębiorstw. Ich elementem jest
przepływ informacji wewnątrz firmy, a także relacje partnerskie z kooperantami
i klientami.
Piąta grupa prac prezentuje doświadczenia w dziedzinie zarządzania edukacją
w krajach Unii Europejskiej. Proces Boloński wymusza działania dostosowawcze podmiotów realizujących zadania w dziedzinie nauki i edukacji
do wspólnych standardów. Ich wprowadzanie wymaga zdolności menadżerskich
nie tylko na poziomie systemu, ale także uczelni, a nawet poszczególnego
wykładowcy i nauczyciela.
Te i inne zagadnienia były też przedmiotem dyskusji na VI Międzynarodowej
konferencji naukowej, która odbyła się w dniach 28-29 maja 2006 w
Krakowie. Konferencja ta organizowana corocznie od sześciu lat przez Krakowską
Szkołę Wyższą im. A.F. Modrzewskiego miała wspólny dla wszystkich
wydziałów tytuł: „Państwo, Gospodarka, Społeczeństwo”. Wydział Zarządzania
i Marketingu w trakcie tej konferencji obradował w sekcji o tytule,
który stał się inspiracją do stworzenia prezentowanej książki.
Niniejsza publikacja, oprócz trzech już serii wydziałowych Zeszytów Naukowych,
jest jedną z dwu pozycji prezentujących dorobek naukowy roku
akademickiego 2005/06. Ze względu na ograniczony rozmiar tej publikacji,
zagadnienia związane z zarządzaniem w aspekcie jakości i ekologii będą
przedmiotem osobnego opracowania
Place and Significance of Mountain Tourism in Modern Tourist Movements
At the end of the 80’s of last century began the trend of reducing the demand for mass tourism and there is a new form of demand for tourism products. Modern tourism demand is directed towards the so-called “6E” (experience, excitement, escape, education, entertaiment, ecology). Development of retail market in Serbia
The aim of this paper is to analyze retail development in Serbia under the conditions of globalization and internationalization in business. The starting point is in current trends in trade in both EU and transitional countries. Macro environmental factors affecting trade development have been investigated as well as indicators related to the number of trade companies, real turnover, employment rate, etc. On the basis of the indicators outlined, the present state of trade in Serbia has been observed and prospect trends have been pointed out. Some assumptions about future successful development along with the increasing competitiveness of Serbian trade have also been made
Effect of hydrogen evolution reaction on morphology and crystal structure of electrolytically produced copper powder particles
Morphologies of copper powder particles obtained by potentiostatic regime of electrolysis under different hydrogen evolution conditions were correlated with their crystal structure. In the dependence of quantity of evolved hydrogen, the either dendritic or cauliflower-like particles were formed. In the both types of the particles, Cu crystallites were random oriented. This random orientation is ascribed to the existence of the spherical morphology and to similar morphology at the micro level, because the both types of particles were constructed from small agglomerates of approximately spherical Cu grains at the micro level. The particle size distribution (PSD) of the obtained particles is also examined and it was shown the strong effect of the shape and size of particles on it, with a tendency of the decrease of particle size with increasing the overpotential of electrolysis.Morfologije praškastih čestica bakra dobijene potenciostatskim režimom elektrolize pod različitim uslovima izdvajanja vodonika su korelisane sa njihovom kristalnom strukturom. U zavisnosti od količine izdvojenog vodonika, formirane su ili dendritične ili karfiolaste čestice. U oba tipa čestica, kristaliti bakra su bili slučajno orijentisani. Ova slučajna orijentacija je pripisana postojanju sferične morfologije i sličnoj morfologiji na mikro nivou, zato što oba tipa čestica su bila izgrađena od malih aglomerata približno sferičnih zrna bakra na mikro novou. Raspodela veličine čestica (PSD) dobijenih čestica je takođe ispitana i pokazan je strog uticaj oblika i veličine čestica na raspodelu, sa tendencijom smanjenja veličine čestica sa povećavajućom prenapetošću elektrolize
Comprehensive qualitative evaluation of the first ever full-time online course for medical students in one middle-income country
ABSTRACTThe measures to control the COVID-19 pandemic in the academic 2020/2021 included full-time online courses for medical students in Serbia for the first time since the faculty was founded in 1920. The purpose of this study was to examine the attitudes and opinions about the full-time online course among medical students and their teachers. A qualitative study was carried out in the autumn of 2021. In-depth interviews were conducted with 38 participants (27 students in the 4th study year and 11 teachers who teach in the 3rd study year) about their experiences in the past academic year. To select study participants, purposive sampling was employed. The interviews were audio-recorded and transcribed verbatim. Qualitative content analysis was applied. Five topics emerged: 1) Theoretical online learning, 2) Practical online learning, 3) Motivation, 4) Technical aspects of online learning and 5) Evaluation of knowledge. Overall, students’ attitudes toward theoretical classes were mostly negative, but the attitudes of teachers were mostly positive. However, the attitudes of both students and teachers toward practical online classes were mostly negative. Students were, in general, less motivated to follow online classes compared to conventional courses, even though the teachers felt that they had to put more effort into making a suitable online course. Most participants considered that the online classes had a negative impact on their knowledge and practical skills, but their academic achievement was not inferior compared to previous years. The full-time online course should include conventional on-site teaching. The relevance of acquiring practical skills is strongly emphasized
Liming of anthropogenically acidified soil promotes phosphorus acquisition in the rhizosphere of wheat
We studied the effect of liming and P fertilization of extremely acid soil (accidently acidified by sulfidic mining waste) on P availability and the subsequent adaptive responses of wheat roots. The wheat plants were grown in rhizoboxes allowing precise sampling of rhizosphere and bulk soil for sequential extraction of P fractions and determination of exchangeable Al. Root exudates were collected by pieces of paper for electrophoresis and subjected to HPLC analysis. Expression of organic anions and P-i transporter genes was analyzed by a real-time quantitative PCR. The concomitant application of lime with P fertilization increased the concentrations of plant-available P fractions in both rhizosphere and bulk compartments. The applied soil amendments strongly affected plant growth, biomass partitioning and shoot P accumulation. Liming enhanced root exudation of citrate in P unfertilized plants, while the high malate efflux was maintained until both P deficiency and Al toxicity were eliminated by the amendments. We showed the importance of liming for recovering of P acquisition potential of wheat roots, which can be strongly impaired in acid soils. Our results clearly demonstrated that P-deficient roots not subjected to Al stress in the limed soil can maintain high efflux of malate and even increase efflux of citrate along with the enhanced expression of related anion transporters (TaMATE1 and TaALMT1)
