    Matrix effect in bio-analysis of illicit drugs with LC-MS/MS: Influence of ionization type, sample preparation, and biofluid

    AbstractThe purpose of the present work was to evaluate the synergistic effect of ionization type, sample preparation technique, and bio-fluid on the presence of matrix effect in quantitative liquid chromatography (LC)-MS/MS analysis of illicit drugs by post-column infusion experiments with morphine (10-μg/mL solution). Three bio-fluids (urine, oral fluid, and plasma) were pretreated with four sample preparation procedures [direct injection, dilution, protein precipitation, solid-phase extraction (SPE)] and analyzed by both LC-electrospray ionization (ESI)-MS/MS and LC-atmospheric pressure chemical ionization (APCI)-MS/MS. Our results indicated that both ionization types showed matrix effect, but ESI was more susceptible than APCI. Sample preparation could reduce (clean up) or magnify (pre-concentrate) matrix effect. Residual matrix components were specific to each bio-fluid and interfered at different time points in the chromatogram. We evaluated matrix effect in an early stage of method development and combined optimal ionization type and sample preparation technique for each bio-fluid. Simple dilution of urine was sufficient to allow for the analysis of the analytes of interest by LC-APCI-MS/MS. Acetonitrile protein precipitation provided both sample clean up and concentration for oral fluid analysis, while SPE was necessary for extensive clean up of plasma prior to LC-APCI-MS/MS

    Modern human activity fueled by economic development is profoundly altering our relationship with microorganisms. This altered interaction with microbes is believed to be the major driving force behind the increased rate of emerging infectious diseases from animals. The spate of recent infectious disease outbreaks, including Ebola virus disease and Middle East respiratory syndrome, emphasize the need for development of new innovative tools to manage these emerging diseases. Disseminating vaccines are one such novel approach to potentially interrupt animal to human (zoonotic) transmission of these pathogens

    ATPMN: accurate positions and flux densities at 5 and 8 GHz for 8,385 sources from the PMN survey

    We present a source catalogue of 9,040 radio sources resulting from high-resolution observations of 8,385 PMN sources with the Australia Telescope Compact Array. The catalogue lists flux density and structural measurements at 4.8 and 8.6 GHz, derived from observations of all PMN sources in the declination range -87 deg < delta < -38.5 deg (exclusive of galactic latitudes |b| 70 mJy (50 mJy south of delta = -73 deg). We assess the quality of the data, which was gathered in 1992-1994, describe the population of catalogued sources, and compare it to samples from complementary catalogues. In particular we find 127 radio sources with probable association with gamma-ray sources observed by the orbiting Fermi Large Area Telescope.Comment: 20 pages, 21 figure

    Thresholds for tracing ships' ballast water: An Australian case study

    To limit the spread of non-indigenous marine species, ships can be legally required to conduct ballast water exchange (BWE) prior to discharging ballast water. It has been proposed to verify BWE by measuring concentrations of coastal tracers in ballast tanks, which should track their removal. Using 3 Australian ports as case studies (Port Botany, Port Curtis and Port Phillip Bay), each representing a different BWE verification difficulty level, the spatial and temporal variability of chromophoric dissolved organic matter (CDOM) and 3 trace elements (manganese [Mn], barium [Ba] and phosphorus [P]), were measured to assess their utility as tracers of coastal (unexchanged) ballast water. CDOM fluorescence at λex/λem = 320/414 nm (C2*) and 370/494 nm (C3*) and Mn concentrations were significantly higher in ports than in the adjacent Tasman Sea, except near port entrances and at a few sites in Port Botany. Ba concentrations demonstrated the least power to discriminate coastal sources, but P easily discriminated water from mesotrophic Port Phillip Bay. In general, tracers showed greater variation between and within ports, rather than between seasons. Conservative BWE thresholds were calculated to be 1.6 quinine sulphate equivalents for C2*, 0.9 quinine sulphate equivalents for C3*, 1.4 μg l-1 for Mn and 6.9 μg l -1 for Ba. Overall, these thresholds would allow water sourced from eastern Australian ports to be identified as coastal at 92%, 69% and 74% of sites examined using C3*, Mn and Ba, respectively, requiring 71 ± 26%, 54 ± 40% and 59 ± 38% replacement with mid-ocean water to be within ocean baseline concentration ranges. © Inter-Research 2010 · www.int-res.com

    The MASSIVE Survey II: Stellar Population Trends Out to Large Radius in Massive Early Type Galaxies

    We examine stellar population gradients in ~100 massive early type galaxies spanning 180 < sigma* < 370 km/s and M_K of -22.5 to -26.5 mag, observed as part of the MASSIVE survey (Ma et al. 2014). Using integral-field spectroscopy from the Mitchell Spectrograph on the 2.7m telescope at McDonald Observatory, we create stacked spectra as a function of radius for galaxies binned by their stellar velocity dispersion, stellar mass, and group richness. With excellent sampling at the highest stellar mass, we examine radial trends in stellar population properties extending to beyond twice the effective radius (~2.5 R_e). Specifically, we examine trends in age, metallicity, and abundance ratios of Mg, C, N, and Ca, and discuss the implications for star formation histories and elemental yields. At a fixed physical radius of 3-6 kpc (the likely size of the galaxy cores formed at high redshift) stellar age and [alpha/Fe] increase with increasing sigma* and depend only weakly on stellar mass, as we might expect if denser galaxies form their central cores earlier and faster. If we instead focus on 1-1.5 R_e, the trends in abundance and abundance ratio are washed out, as might be expected if the stars at large radius were accreted by smaller galaxies. Finally, we show that when controlling for \sigmastar, there are only very subtle differences in stellar population properties or gradients as a function of group richness; even at large radius internal properties matter more than environment in determining star formation history.Comment: 17 pages, 9 figures, accepted by ApJ; resubmitted with updated reference
