    Variational Temporal IRT: Fast, Accurate, and Explainable Inference of Dynamic Learner Proficiency

    Dynamic Item Response Models extend the standard Item Response Theory (IRT) to capture temporal dynamics in learner ability. While these models have the potential to allow instructional systems to actively monitor the evolution of learner proficiency in real time, existing dynamic item response models rely on expensive inference algorithms that scale poorly to massive datasets. In this work, we propose Variational Temporal IRT (VTIRT) for fast and accurate inference of dynamic learner proficiency. VTIRT offers orders of magnitude speedup in inference runtime while still providing accurate inference. Moreover, the proposed algorithm is intrinsically interpretable by virtue of its modular design. When applied to 9 real student datasets, VTIRT consistently yields improvements in predicting future learner performance over other learner proficiency models.Comment: 9 pages, 16th International Conference on Educational Data Mining (EDM'23

    Algorithms for a class of single-machine weighted tardiness and earliness problems

    We address the problem of determining schedules for static, single-machine scheduling problems where the objective is to minimize the sum of weighted tardiness and weighted earliness. We develop optimal and heuristic procedures for the special case of weights that are proportional to the processing times of the respective jobs. The optimal procedure uses dominance properties to reduce the number of sequences that must be considered, and some of the heuristic use these properties as a basis for constructing good initial sequences. A pairwise interchange procedure is used to improve the heuristic solutions.     Development and Evaluation of An Educational Video For High School Students On Vaping And Other Nicotine Products

    Background: Electronic nicotine delivery systems (ENDS), especially vaping, have increased in use and resulted in severe lung injuries (EVALI). In 2020, there were nearly 3.6 million middle school and high school (HS) students who vape. More research is needed to identify effective teaching strategies for adolescents on ENDS. Our previous research found that high school students’ experiences with vaping were related to social and behavioral influences, a knowledge deficit, and media exposure. They suggested a video educational intervention. Purpose: To develop and evaluate the implementation of a vaping survey and educational video in a high school classroom setting and to further explore the knowledge, attitudes, and behaviors of high school students regarding nicotine products. Methods: For the educational video, we videotaped interviews with six college-aged vapers. They were asked seven open-ended questions that elicited dialogue regarding their experiences with ENDS products. A process evaluation was then conducted. Students from a rural high school in a health education completed our validated TABS survey, viewed the developed video, and then participated in a semi-structured focus group to evaluate the video and survey. An interview was conducted with their teacher. COVID-19 restrictions dictated the process take place through a virtual platform. Results: Feedback obtained from the students indicated that there was a knowledge deficit about ENDS ingredients; the survey was easy to complete and understand, and the video was an appropriate length with dramatic relief achieved. The teacher, with no prior education on these products, suggested the video be used as an introduction to ENDS education for students. We then developed a follow up demonstration module. Conclusion/Implications: This vaping video educational module aims to reduce the knowledge deficit and usage of ENDS in adolescents.     Vaping: Exploring perceptions and behaviors of students and ENDS products

    Background: Electronic nicotine delivery systems (ENDS) have increased in use and media exposure with a 78% rise in usage between 2017 to 2018. While a gateway to addictive products, users are also unaware of the ingredients and their effects on the body. Interventions are limited. Further research is needed to investigate perceptions of ENDS and identify effective cessation strategies for young adults. Purpose: To explore the knowledge and attitudes of HS students regarding ENDS products and compare data collected over the year to determine effects of recent media exposure. Methods: Six focus groups (n = 45) were held in a rural high school. Students were asked four open ended questions which elicited dialogue regarding attitudes and behaviors about ENDS and suggestions for educational interventions. Results: Emerging themes were social and behavioral influences (peer pressure; nicotine addiction) ; knowledge deficit; media promotion or prevention. Suggested educational methods were explanation of the ingredients in ENDS, dramatic relief and visual depiction of the consequences. There was more focus in the media on prevention than last year. Conclusion/Implications: A knowledge deficit regarding the dangers of ENDS, including the damaging effects of nicotine, still exists.     Promotion of physical activity in rural, remote and northern settings: a Canadian call to action

    Introduction: The lack of policy, practice and research action on physical activity and features of the physical (built and natural) environments in rural, remote and northern settings is a significant threat to population health equity in Canada. This paper presents a synthesis of current evidence on the promotion of physical activity in non-urban settings, outcomes from a national priority-setting meeting, and a preliminary call to action to support the implementation and success of population-level initiatives targeting physical activity in non-urban settings. Methods: We conducted a “synopses of syntheses” scoping review to explore current evidence on physical activity promotion in rural, remote, northern and natural settings. Next, we facilitated a collaborative priority-setting conference with 28 Canadian experts from policy, research and practice arenas to develop a set of priorities on physical activity in rural, remote and northern communities. These priorities informed the development of a preliminary Canadian call to action. Results: We identified a limited number of reviews that focused on physical activity and the built environment in rural, remote and northern communities. At the priority-setting conference, participants representing rural, remote and northern settings identified top priorities for policy, practice and research action to begin to address the gaps and issues noted in the literature. These priorities include self-identifying priorities at the community level; compiling experiences; establishing consistency in research definitions and methods; and developing mentorship opportunities. Conclusion: Coordinated action across policy, practice and research domains will be essential to the success of the recommendations presented in this call to action

    A multiple case history and systematic review of adoption, diffusion, implementation and impact of provincial daily physical activity policies in Canadian schools

    BACKGROUND : Few children meet physical activity (PA) recommendations, and are therefore at increased risk for overweight/obesity and adverse health outcomes. To increase children\u27s opportunities for PA, several Canadian provinces have adopted school-based daily PA (DPA) policies. It is not clear why some jurisdictions have adopted DPA policies, and others have not, nor whether these policies have been implemented and have achieved their intended outcomes. The purpose of this study was to understand the processes underlying adoption and diffusion of Canadian DPA policies, and to review evidence regarding their implementation and impact. METHODS: We adopted a multiple case history methodology in which we traced the chronological trajectory of DPA policies among Canadian provinces by compiling timelines detailing key historical events that preceded policy adoption. Publicly available documents posted on the internet were reviewed to characterize adopter innovativeness, describe the content of their DPA policies, and explore the context surrounding policy adoption. Diffusion of Innovations theory provided a conceptual framework for the analyses. A systematic literature search identified studies that had investigated adoption, diffusion, implementation or impact of Canadian DPA policies. RESULTS: Five of Canada\u27s 13 provinces and territories (38.5%) have DPA policies. Although the underlying objectives of the policies are similar, there are clear differences among them and in their various policy trajectories. Adoption and diffusion of DPA policies were structured by the characteristics and capacities of adopters, the nature of their policies, and contextual factors. Limited data suggests implementation of DPA policies was moderate but inconsistent and that Canadian DPA policies have had little to no impact on school-aged children\u27s PA levels or BMI. CONCLUSIONS: This study detailed the history and current status of Canadian DPA policies, highlighting the conditional nature of policy adoption and diffusion, and describing policy and adopter characteristics and political contexts that shaped policy trajectories. An understanding of the conditions associated with successful policy adoption and diffusion can help identify receptive contexts in which to pioneer novel legislative initiatives to increase PA among children. By reviewing evidence regarding policy implementation and impact, this study can also inform amendments to existing, and development of future PA policies

    Integrative Genomics Analysis Unravels Tissue-Specific Pathways, Networks, and Key Regulators of Blood Pressure Regulation

    Blood pressure (BP) is a highly heritable trait and a major cardiovascular disease risk factor. Genome wide association studies (GWAS) have implicated a number of susceptibility loci for systolic (SBP) and diastolic (DBP) blood pressure. However, a large portion of the heritability cannot be explained by the top GWAS loci and a comprehensive understanding of the underlying molecular mechanisms is still lacking. Here, we utilized an integrative genomics approach that leveraged multiple genetic and genomic datasets including (a) GWAS for SBP and DBP from the International Consortium for Blood Pressure (ICBP), (b) expression quantitative trait loci (eQTLs) from genetics of gene expression studies of human tissues related to BP, (c) knowledge-driven biological pathways, and (d) data-driven tissue-specific regulatory gene networks. Integration of these multidimensional datasets revealed tens of pathways and gene subnetworks in vascular tissues, liver, adipose, blood, and brain functionally associated with DBP and SBP. Diverse processes such as platelet production, insulin secretion/signaling, protein catabolism, cell adhesion and junction, immune and inflammation, and cardiac/smooth muscle contraction, were shared between DBP and SBP. Furthermore, “Wnt signaling” and “mammalian target of rapamycin (mTOR) signaling” pathways were found to be unique to SBP, while “cytokine network”, and “tryptophan catabolism” to DBP. Incorporation of gene regulatory networks in our analysis informed on key regulator genes that orchestrate tissue-specific subnetworks of genes whose variants together explain ~20% of BP heritability. Our results shed light on the complex mechanisms underlying BP regulation and highlight potential novel targets and pathways for hypertension and cardiovascular diseases

    Bacterial Delivery of Nuclear Proteins into Pluripotent and Differentiated Cells

    Numerous Gram negative pathogens possess a type III secretion system (T3SS) which allows them to inject virulent proteins directly into the eukaryotic cell cytoplasm. Injection of these proteins is dependent on a variable secretion signal sequence. In this study, we utilized the N-terminal secretion signal sequence of Pseudomonas aeruginosa exotoxin ExoS to translocate Cre recombinase containing a nuclear localization sequence (Cre-NLS). Transient exposure of human sarcoma cell line, containing Cre-dependent lacZ reporter, resulted in efficient recombination in the host chromosome, indicating that the bacterially delivered protein was not only efficiently localized to the nucleus but also retained its biological function. Using this system, we also illustrate the ability of P. aeruginosa to infect mouse embryonic stem cells (mESC) and the susceptibility of these cells to bacterially delivered Cre-NLS. A single two-hour infection caused as high as 30% of the mESC reporter cells to undergo loxP mediated chromosomal DNA recombination. A simple antibiotic treatment completely eliminated the bacterial cells following the delivery, while the use of an engineered mutant strain greatly reduced cytotoxicity. Utility of the system was demonstrated by delivery of the Cre-NLS to induced pluripotent stem cells to excise the floxed oncogenic nuclear reprogramming cassette. These results validate the use of T3SS for the delivery of transcription factors for the purpose of cellular reprogramming
