    Screening of Fusarium graminearum isolates for enzymes extracellular and deoxynivalenol production

    Fusarium graminearum, the main etiological agent of Fusarium Head Bligh, has high intraspecific genetic diversity, which is related to the variability in the aggressiveness among isolates against wheat. The aggressiveness involves different mechanisms as the production and liberation of extracellular enzymes and mycotoxins. In the present report, several F. graminearum isolates obtained from wheat spikes from Pampas region, Argentina, were screened for polygalacturonase (pectinase), proteolytic and lipase extracellular enzymatic activities production, as well as by the capacity to produce deoxynivalenol. The enzymatic production in terms of magnitude was varied among isolates, which could be related to a differential capacity to infect wheat. Both polygalacturonase as proteolytic activities had a maximum activity in the first days of incubation. Instead, the lipase activity reached its maximum activity advanced incubation time. Deoxynivalenol production was delayed over time with respect to the first enzymatic activities, which would infer its relation with the progress of the disease in the host, more than with the early stages of infection.     Characterization of <i>Fusarium graminearum</i> isolates recovered from wheat samples from Argentina by Fourier transform infrared spectroscopy: phenotypic diversity and detection of specific markers of aggressiveness

    Fusarium graminearum is the primary causal agent of Fusarium head blight of wheat in Argentina. This disease affects crop yields and grain quality also reducing the wheat end-use, and causing mycotoxin contamination. The aim of this work was to analyze the phenotypic characteristics associated with phenotypic diversity and aggressiveness of 34 F. graminearum sensu stricto isolates recovered from Argentinean fields in the 2008 growing season using the Fourier Transform Infrared (FTIR) dried film technology. We applied this technique also to search for spectral specific markers associatedwith aggressiveness. The combination of FTIR technologywith hierarchical cluster analysis allowed us to determine that this population constitutes a highly diverse and heterogeneous group of fungi with significant phenotypic variance. Still, when the spectral features of a set of these isolates were compared against their aggressiveness, as measured by disease severity, thousand grains weight, and relative yield reduction, we found that the more aggressive isolates were richer in lipid content. Therefore, we could define several spectroscopic markers (> C — H stretching modes in the 3000–2800 window, > C=O and — C — O vibrational modes of esters at 1765–1707 cm−1 and 1474–900 cm−1, respectively),, mostly assigned to lipid content that could be associated with F. graminearum aggressiveness. All together, by the application of FTIR techniques and simple multivariate analyses, it was possible to gain significant insights into the phenotypic characterization of F. graminearum local isolates, and to establish the existence of a direct relationship between lipid content and fungal aggressiveness. Considering that lipids have a major role as mediators in the interaction between plants and fungi our results could represent an attractive outcome in the study of Fusarium pathogenesis.Centro de Investigación y Desarrollo en Fermentaciones Industriale

    Occurrence of Different Species of <i>Fusarium</i> from Wheat in Relation to Disease Levels Predicted by a Weather-Based Model in Argentina Pampas Region

    Fusarium head blight (FHB) is an important disease throughout many of the world wheat-growing areas that have humid to semi-humid climate. The infection happens mainly during the anthesis of the wheat, when there have been favorable conditions of moisture and temperature. The direct relation of the infection to environmental factors makes possible the formulation of mathematical models that predict the disease. The causal agent of the FHB of the spike of wheat is attributed principally to Fusarium graminearum. High economic losses due yield decrease have been recorded in Argentina. In the present work, 67 isolates of Fusarium spp. were obtained from samples of wheat grains from Pampas region from 15 locations distributed in Buenos Aires, Entre Rios, Santa Fe and Corboba provinces during 2006 and 2007 wheat-growing seasons. The identification of species from monosporic isolates was carried out by morphological characterization and use of species-specific PCR-based assays. Both identification criteria were necessary and complementary for the species determination, since in some cases the molecular identification was not specific. Scanty presence of F. graminearum was observed in 2006 wheat-growing season coinciding with the lack of favorable meteorological conditions for producing FHB infection events. High presence of F. graminearum isolates was observed in 2007 wheat-growing season, in accordance with moderate incidence of the disease according to spatial distribution of FHB incidence values.     Fusarium graminearum isolates from three different agroecological regions in Argentina were examined according to the production of different extracellular enzyme activities of potential biotechnological interest: pectinases (PGase: polygalacturonase and PMGase: polymethylgalacturonase), cellulase (CMCase: carboxymethylcellulase) and hemicellulase (xylanase). The isolates were grown in minimum salt medium supplemented with 0.25% glucose, 0.125% citric pectin and 0.125% oat bran as carbon sources and/or enzyme inducers. PGase activity was detected early (after two days of incubation) in all the cultures; it was found to be the highest for all the isolates. PMGase was high only for those isolates of the II region. CMCase and endoxylanase activities were particularly found at late stages (after four and seven days of incubation, respectively) and the maximum values were lower than pectinase activities
