    The DnaA AAA+ Domain His136 Residue Directs DnaB Replicative Helicase to the Unwound Region of the Replication Origin, oriC

    Chromosomal replication initiation requires dynamic mechanisms in higher-order nucleoprotein complexes that are constructed at the origin of replication. In Escherichia coli, DnaA molecules construct functional oligomers at the origin oriC, enabling localized unwinding of oriC and stable binding of DnaB helicases via multiple domain I molecules of oriC-bound DnaA. DnaA-bound DnaB helicases are then loaded onto the unwound region of oriC for construction of a pair of replisomes for bidirectional replication. However, mechanisms of DnaB loading to the unwound oriC remain largely elusive. In this study, we determined that His136 of DnaA domain III has an important role in loading of DnaB helicases onto the unwound oriC. DnaA H136A mutant protein was impaired in replication initiation in vivo, and in DnaB loading to the unwound oriC in vitro, whereas the protein fully sustained activities for oriC unwinding and DnaA domain I-dependent stable binding between DnaA and DnaB. Functional and structural analyses supported the idea that transient weak interactions between DnaB helicase and DnaA His136 within specific protomers of DnaA oligomers direct DnaB to a region in close proximity to single stranded DNA at unwound oriC bound to DnaA domain III of the DnaA oligomer. The aromatic moiety of His136 is basically conserved at corresponding residues of eubacterial DnaA orthologs, implying that the guidance function of DnaB is common to all eubacterial species

    ベッド上での水平移動時における看護師の腕の差し入れの深さからみた作業効率性の検討

     ベッド上での水平移動時の看護師の腕の差し入れの深さからみた作業効率性について明らかにすることを目的とした。研究デザインは準実験研究である。被験者は、女性看護師10 名とし、実験は1 名の被験者が3 名の模擬患者に、腕の差し入れの深い群と浅い群の2 種類についてベッドの右側へ引き寄せる水平移動を設定して実施した。測定項目は重心軌跡として、足底の荷重中心の軌跡を重心線の軌跡、仙骨部のマーカーを重心の上下の動きを反映するデータとして測定した。腕の差し入れの深い群のほうが浅い群に比べ、荷重中心の総軌跡長が有意( p < 0.05)に短く、所要時間は有意( p < 0.01)に短く、動作回数は有意( p < 0.05)に少なかった。また、腕の差し入れの深い群をやりやすいと選択した被験者が有意( p < 0.01)に多かった。これらの結果を得たことから腕の差し入れの深い群のほうが作業効率があがっていたことが明らかとなった

    同志社女子大学における薬学部と看護学部の協働でのInterprofessional Education の取り組み

    In the spring semester of 2021, the Faculty of Pharmaceutical Sciences and the Faculty of Nursing of Doshisha Women’s College of Liberal Arts jointly conducted wheelchair experience training for the 1st-year students as a multidisciplinary education. We conducted a questionnaire survey on pharmacy and Nursing students before and after training in order to assess the effect of interprofessional education and analyzed the report after the training using text mining method. Questionnaire items are the importance of each item related to interprofessional collaboration, KiSS-18 (Kikuchi’s scale of social skills: 18 items), RIPLS (readiness for interprofessional learning scale). The students of both faculties evaluated themselves quite high for even before the training and retained after the training. However, they may misunderstand or understand not quite well about their professional roles and mutual cooperation in the real clinical settings.     地域に根付く米国ホスピスのフィールドワーク:ボランティアとの協働からみた本邦の看護教育についての考察

    Portland, Oregon, has attracted attention in Japan as the “most desirable city to live in the United States” and “a model of urban development.” The purpose of this study was to examine nursing education in Japan through care that supports people who live and die in the community by visiting the Hopewell House Hospice in Portland. The hospice is run by approximately 70 volunteers and facility staff who provide facility care for 11 beds and about 150 home care patients. From fieldwork conducted at the facility and from an interview with the volunteer staff manager who was a former midwife and nursing manager, we found that the users and the staff themselves have “a special place called a bed,” a “record of their personality and health,” and “prayer and hope”. In addition, the nurses relied on their ingenuity to manage their environment and provide continuous care in order to reach the end stage while keeping the patient connected to the community, which facilitated feelings of joy. Nursing practice in the Hopewell House Hospice was influenced by the climate of the region and the culture of the local residents, so there was an overlap between life and death, which we believe should be incorporated into nursing education in Japan.論

    Uma abordagem promissora para a obtenção de coquetéis enzimáticos eficientes para a etapa de hidrólise na produção de etanol de 2ª geração é o enriquecimento em termos de atividades que lhes faltam, mas podem ser obtidas através de sistemas heterólogos. O trabalho teve como objetivo avaliar o sistema toxina -antitoxina (TA) para estabilização plasmidial em E. coli na produção de expansina e endoglucanase recombinantes. Os resultados indicaram que o sistema de expressão com estabilização TA é tão eficaz quanto o dependente de antibiótico para estabilização plasmidial. Além disso, mostrou-se mais eficiente pelo fato de não permitir a sobrevivência de células sem o plasmídeo. Estudos indicaram a quantidade mínima de 0,05mM de IPTG para expressão das proteínas nesta linhagem, cerca de 20 vezes menos que a concentração usualmente aplicada no momento da indução. Sendo assim, o sistema de estabilização plasmidial TA mostrou-se uma ótima ferramenta para o desenvolvimento de uma plataforma alternativa para a produção de proteínas recombinantes.A promising approach to obtain efficient enzyme cocktails for the hydrolysis step in the production of 2nd generation ethanol is the enrichment in terms of activities that are lacking, but can be obtained by heterologous systems. The study aimed to evaluate the toxina-antitoxin (TA) system for plasmid stabilization in E. coli in the production of recombinant expansin and endoglucanase. The results indicated that the expression system with TA stabilization is as effective as the antibiotic dependent for plasmid stabilization. Moreover, it proved to be more efficient by not allo wing the survival of cells without the plasmid. Studies have indicated the minimum amount of 0.05 mM IPTG for expression of proteins in this strain, about 20 times less than the concentration usually applied for induction. Thus, the plasmid stabilization TA system proved to be a great tool for the development of an alternative platform for producing recombinant proteins
